CONTENT Genetic Engineering and Applications of Recombinant DNA

I. Enumeration

Give the steps in Genetic Recombination Technology.

a. Isolation of Genetic Material

b. Restriction Enzyme Digestion
c. PCR Amplification
d. Ligation of DNA Molecules
e. Insertion of Recombinant DNA into Host
f. Isolation of Recombinant Cells

II. Definition of Terms

In your own words, define or describe the functions of each term used in recombinant DNA
technology:

a. Recombinant DNA

 It is the joining of DNA molecules of two different species into a host organism to
produce and form a new genetic combination.

b. DNA Ligase

 It is an enzyme that acts as a glue that joins the DNA strands that have matching ends
to form a single DNA molecule.

c. Restriction enzyme

 Restriction enzyme mostly found in bacteria, acts as a molecular scissors in cutting the
target or desired sequence in a DNA called the restriction sites.

d. PCR
 PCR or Polymerase Chain Reaction is the third step in Genetic Recombination, and it is
the step where we will be making lots of copies of the desired fragment of a DNA.
