9.1. DETECTION OF UREA IN URINE

Part I. Sodium Hypobromite Test

Materials
urine sample test tube
sodium hypobromite (NaBrO) dropper
solution measuring cylinder

Procedure
1. Take ~2 mL urine sample and pour it into a test tube.
2. Using a dropper, take sodium hypobromite solution and add few drops of the solution to the
urine sample.
3. Brisk effervescence of nitrogen appears in the test tube which indicates presence of urea in
the urine.

Part II. Urease Test

Materials
urine sample urease phenol red indicator
powder measuring cylinder
2% sodium carbonate (Na2CO3) dropper spatula
1% acetic acid (CH3COOH)

Procedure
1. Take a test tube and pour ~2 mL urine sample into the test tube.
2. Using a dropper, take some phenol red indicator and add 4-5 drops of phenol red indicator to
the test tube.
3. Use a fresh dropper to take few drops of 2% Na2CO3. Add Na2CO3 solution drop by drop
until a pink color develops in the test tube.
4. Take few drops of 1% acetic acid using a fresh dropper. Add 1% acetic acid to the test tube
drop by drop until the pink color disappears.
5. Using a spatula, take some urease powder and add urease powder to the test tube and shake it
well.
6. The pink or red color appearing in the solution indicates the presence of urea in urine.

Sodium Hypobromite While watching the Detection of Urea in


Urine video (by amritacreate), when the
speaker added a few drops of sodium
hypobromite solution to the test tube (which
had urine already), there was a brisk
effervescence in the tube which indicated the
presence of urea in the urine.
Urease While I was watching the Detection of Urea in
Urine video (by amritacreate), when the
speaker demonstrated by adding phenol red
indicator the urine sample turned reddish,
next by adding 2% of Sodium Carbonate the
sample turned pink, and finally, by adding
1% of Acetic acid the pink color
dissappeared. The speaker then added urease
powder to the sample and shook it. You will
know that urease is present in the urine if the
color pink or red appeared in the solution.

9.2. DETECTION OF ALBUMIN IN URINE

Part I. Sulfosalicylic Acid Test

Materials
urine sample test tube holder
30% sulfosalicylic acid (C7H6O6S) measuring cylinder
hot water bath test dropper
tube

Procedure
1. Take a test tube and pour the urine sample in the test tube.
2. Using a dropper, take some sulfosalicylic acid. Add few drops of sulfosalicylic acid in the
test tube containing urine. A whitish color appears in the solution.
3. Using a test tube holder, put the test tube in a hot water bath.
4. A whitish or cloudy turbid solution indicates the presence of albumin in the urine sample.

Part II. Heller’s Test

Materials
urine sample test tube holder
concentrated nitric acid (HNO3) measuring cylinder
test tube dropper

Procedure
1. Put a small amount of concentrated nitric acid in a test tube.
2. Using a dropper, take some urine sample.
3. Now incline the test tube and add sample of urine by means of a dropper along the inner side
of the test tube so that it forms a layer over the nitric acid.

4. The presence of white ring at the junction of two layers indicates the presence of albumin in
the urine.

Test Observations
Sulfosalicylic Acid While I was watching the Detection of Albumin
in Urine video (by amritacreate), I have noticed
that by adding a few drops of sulfosalicylic acid
in the test tube containing urine, a whitish color
appears in the solution and by heating it, a
whitish or cloudy turbid solution appears
indicating the presence of albumin in the urine
sample.
Heller’s During the video, I have noticed that this was a
different test among all the other experiments
performed by the speaker. I have also noticed
that by adding a small sample of urine in the test
tube containing concentrated nitric acid, it
formed a layer above the concentrated nitric
acid. The presence of a white ring at the junction
of two layers indicates the presence of albumin
in the urine.

Guide Questions

1. What does urea in urine indicate about a person’s health?

When you urinate, your kidneys expel urea, which is a waste product. The urine urea
nitrogen test assesses the quantity of protein breakdown by determining how much urea is
present in the urine. The test can tell you how effectively your kidneys are working and
whether your protein intake is too high or too low.

2. What does it mean when there is albumin detected in the urine?

The protein albumin is found in the blood. Albumin from the blood is not allowed to
flow into the urine by a healthy kidney.
