DISCUSSION
Based on Glucose fermentation test, the pH indicator phenol red is used to detect the change in pH.
Glucose fermentation will produce acidic waste and inverted tube called Durham tube is inserted in
the media to accumulate carbon dioxide produced as a waste product of fermentation. From this
test, the E. coli and S. sonnei test tube changed from red phenol to yellow which shows a positive
reaction where the bacteria are capable of fermenting the provided sugar and result in a drop of the
pH value. In test tube inoculated and P.aeruginosa the liquid is orange in color which shows
negative reaction where there's no acid produce and the glucose fermentation did not occur. Also,
there is gas production along with acid production in E. coli test tube because there is bubble
present at the top of the Durham tube.

Referring to figure 2 in starch hydrolysis. Part A, which is Bacillus subtilis indicates a


positive result that there's no longer any starch presents as we can see there is clear zone extending
out from the growth. This is caused by the bacterium which produces Exoenzyme that hydrolyzes
the starch. While in part B, which is E. coli the result shows negative result that the starch is not
hydrolyze and the starch is still present.

Next, from figure 3 of Gelatin hydrolysis. Test tube A which is Staphylococcus aureus
remain liquid shows a positive result which these bacteria contain the enzyme gelatinase. However,
in test tube B which is Staphylococcus epidermidis become solidify indicates negative result and
does not contain the enzyme gelatinase.

From figure 4 in Indole test. Test tube A which Proteus vulgaris, there is a formation of
maroon layer on top of the liquid which indicates positive result, and the organism produces an
enzyme called tryptophanase. Besides, it also shows the ability of Proteus vulgaris to degrade the
amino acid tryptophan and produce indole. However, in Test tube B, Porteus mirabilis there is
formation of yellow layer on top of the liquid which considered to be negative indole result.
