EFFECT OF SUBSTRATE CONCENTRATION ON ENZYME ACTIVITY

Data Gathered:

How would you explain for the values 16 and 32 at 40% and 80% substrate
concentration as well as the zero values at 20% to 80%substrate concentration?
What is the meaning of these values? (6pts)

16 and 32 corresponds to the demanded total number of concentrations per


Substrate Concentrations 40% and 80% respectively. The zero values signify the
number of unoccupied active sites per unit volume per unit time. These values
show us the relationship between the varying concentrations of substrates and
enzymes. On this activity, the concentration of enzymes is constant, while the
substrate concentrations were modified for each tube. The total number of active
sites for all test tubes is 8. When substrate concentration is increased, it means that
the number of substrate molecules are also increased. This increasing of
concentration caused to the decreasing sites of enzymatic reactions in the test
tubes.

Interpretation: (9pts)

We have to keep in mind that the number of active sites is constant to all tubes. The
concentration of the substrates is being increased per set up.

On this graph we see a significant increase in enzymatic activity between 2.5% to


20.0%, this is due to the set ups having lesser or equal number of substrates than or
to the number of active sites. At 20% we see that it is the graph’s peak. The plots
that follows next to this point didn’t have an increase. The graph reached its peak
and began to plateau. Similar to one of our past activities, there was a shift on what
factor is considered the limiting factor. At the first three concentrations, the
substrate concentration was the limiting factor for there are greater number of
unoccupied active sites on these set ups. At the fourth set up, both active sites and
substrate concentration is equal to each other so we can say that at this point it is at
its optimum performance where all substrates are being processed by the active
sites. And because of the fact that the number of active sites is controlled, this
factor then becomes the limiting factor, limiting or restraining the progress of the
results as shown in the graph. This effect caused the graph to plateau. Therefore we
can say that substrate concentration is directly proportional to enzymatic activities
and vice versa.

Conclusion: (3pts)
1)At the end of this activity, I was able to find out that substrate
concentration is directly proportional to enzymatic activity.

2) At the end of this activity, I was able to know that the trend of graph on
the rate of reaction is a slanted straight line signifying a positive progress
between the two variables.

3) At the end of this activity, I have interpreted the given graph which
showed us that the graph had a positive start at the beginning up to the
middle and started to plateau onwards.

1. On the basis of the collision theory of chemical reaction, what are the
conditions needed to cause a chemical reaction to take place? (4pts)
Collision theory states that in order for a chemical reaction to occur,
collisions between two or more molecules must occur. However,
simply colliding the reactants does not result in the transformation of
the reactants into products. Molecules must collide with the correct
spatial orientation and have enough energy.

2. How was the effect of substrate concentration on the enzyme activity


investigated? What are the factors considered? (6ts)
The effect of substrate concentration on the enzyme activity was
investigated through the variations of its concentrations on different
set up. A controlled number of active sites was considered, in order to
have that unbiased data regarding the effect of the substrate on the
enzyme activity. The graph then helped in visualizing this effect and
made the interpretation easier.

3. If we are to keep the substrate concentration as well the pH and temperature


constant, what kind of graph should be expected? (4pts)
If we are to keep the substrate concentration as well the pH and
temperature constant, the kind of graph that should be expected is a
straight horizontal line. Because if all factors are kept constant, and
there is no decrease nor increase in a variable, therefore

4. How will substrate or enzyme concentration affect the rate of reaction?
(3pts)
At first, increasing the substrate concentration boosts the rate of an
enzyme-catalyzed process. As the enzyme molecules become
saturated with substrate, this increase in reaction rate slows down and
eventually turns off. The rate of an enzyme-catalyzed process
increases as the concentration of the enzyme increases.
