1. SAFETY INSTRUCTIONS

1 .1 GENERAL
The contents of this manual including all drawings, tables and graphic
representations are intellectual property of E. Merck.

1.2 WARNING SIGN

This warning sign reminds the user of important items with respect to handling
and safety measures. The warning sign is located on the back panel of the
analyser and warns the user for the following:

2. INSTALLATION

2.1 SHIPPING
The Merck Microlab 200 is shipped in one box, which contains:

The box should be carefully inspected for damage. If there is any major
damage, this should be reported to your supplier. Missing parts should be
reported immediately to E. Merck or their representative.

2.2 UNPACKING
Where the Merck Microlab 200 is to be unpacked and installed by trained
service personnel from E. Merck, the user should not attempt to unpack
the Merck Microlab 200 and accessories. Where the user is carrying out
the installation, the following steps should be followed:
Carefully open the boxes and remove the inserts which hold the
analyser in place.
Place the Merck Microlab 200 on a flat level surface.
Ensure that all accessories have been removed, before
discarding the packing box.

2.4 INSTRUMENT LOCATION
The Merck Microlab 200 is intended for indoor use. It should be located
on a flat surface, which is not exposed to vibration (i.e. centrifuges, etc.)
and direct sunlight.

Installation category 11
The Merck Microlab 200 is, without any switching, suitable for the whole
specified voltage range of 110...240 V nominal.

The fuse holder is located in the mains switch block. at the rear panel of
the analyser.

To replace the fuses, pull out the fuse holder lid. The fuses will come out
together with the lid. Pull the fuses out of the lid and replace them with
the ones of the correct rating. Push back the lid.

For the 110...240 V range, 2 fuses of 2.5 A slow blow (T) 250 V should be
mounted.

Spare or replacement fuses are supplied with the Merck Microlab 200.

2.7 CONNECTIONS
The mains connector should have a solid ground connection, to assure
safety and trouble free operation of the Merck Microlab 200.

The optional DPIJ 411 Il-printer should be connected via the connector
marked "PRINTER" at the rear side. Other printers or data collection
systems have to be connected to the connector marked "RS232".
The DPIJ 411 Il-printer is available in two options: one with its own AC
adapter and one without. In the first case, simply connect the power
cable to the mains. In the other case, connect the power cable with the
small connector to the 5 V DC power outlet which is located on the rear
panel of the Microlab 200 (see figure 3-2 on p. 12).

With the parallel Centronics connection no settings are required in the


analyser. When a printer or data collection system is connected via the
serial RS232 interface, settings should be made in the System Parameters
(see chapter 4).

3. SYSTEM OVERVIEW

3.1 INTRODUCTION
The Merck Microlab 200 is an analyser capable of meeting all the manual
or discrete demands of the clinical laboratory. The Merck Microlab 200
has been designed with flexibility in mind, to cater for the urgent
demands of the medical as well as laboratory staff. The advanced
software is user orientated and friendly to meet the individual
requirements of the Clinical
Laboratory.

3.2 OPTICAL BLOCK


The measurement takes place in a micro flowcell. The measurement
volume is only 32 Ill. The reagent sample mixture is aspirated into the
flowcell by a bellows pump. The volume is continuously programmable up
to 1500 gl- The flowcell is designed for low sample-to-sample carry over,
allowing the use of small volumes. Typically the mixture volumes used
will be in the range 350...500 "l. A flush mode is provided to wash the
flowcell thoroughly between different assays.

The flowcell is electronically thermostatted by means of Peltier elements.


The construction and materials used, makes the temperature response
very rapidly.

The wavelength of the light used for the measurement is selected by


means of high quality narrow band interference filters, mounted in a
filterwheel. The filterwheel is controlled by the program and
measurement can be either monochromatic or bichromatic.

As standard 6 filters are mounted, with the following wavelengths: 340,


405, 505, 546, 578 and 620 nm.

The lightsource used is a long-life quartz Iodine lamp.

3.3 ANALYSER CONTROL


The Merck Microlab 200 can be programmed for 40 different test
methods. After the parameters of a test have been programmed, the
information on how to perform the test is displayed on the screen. In fact

it is the Merck Microlab 200 that guides the user step by step through the
necessary test procedures.

Battery backup saves the programmed parameters for at least 30 days


after the instrument is switched off.

Depending on the parameters programmed for each test, the analyser


selects the temperature, delay time, measurement mode and time,
calculates the result and prints a report. Two types of reports are
available:
Standard reports which provides patient information, names
and results of each requested test, plus flagging of high/low
results and non-linear kinetic results.
Extended reports include, in addition to that above, starting and
end-point absorbances, initial kinetic absorbance and
kinetic reaction rates in AAbs/min, % of non-linearity and
calculation factors.

3.4 QUALITY CONTROL
The system is programmed to allow two quality control surveys for each
test. The system calculates the mean, standard deviation and coefficient
of variation on a maximum of 30 controls measured. This information is
displayed on the first page of the statistics program and the results can be
printed. If the PAGE key is pressed, then a Levey-Jennings plot of the
results is displayed on the screen. The PRINT key can be pressed to obtain
a print-out. An automatic print out will be made every 30 results.

3.5 SCREEN DISPLAY


The display used is a high contrast Liquid Crystal Display (LCD). It can
16 linag of PO characters each and can disolav araohics as well. A
contrast control allows the display to be adapted to the viewing angle of
the operator. The potentiometer is located on the rear panel.

3.6 PRINTER
The Merck Microlab 200 can be equipped with an external 40 column
printer. Once the analyser is switched ON, check that the paper is loaded.
For paper loading, refer to chapter 8.2 The printer uses special thermal
paper.

3.7 KEYBOARD
The keyboard consists of 6 softkeys, 4 hardware function keys, and a
QWERTY-alphanumerical keyboard. The softkeys are directly located
under the LCD screen. The function of these keys is projected on the
screen with the first 3 or 4 letters of the functions.

3.7.1 SOFT KEYS


Selects (re)calibration for the assay in progress.

Starts the system to calculate a non linear curve after the measurement
CALC of the calibrators.

CHNG Gives the possibility to change the data of a calibration curve.

Selects the control measurement mode. The measured results will be


CONI added automatically to the statistical memory used for the Quality
Control calculations, programmed in the Microlab 200.

This function is used to delete data. This can be a complete programmed
DELE test, patient or control data.

When starting a measurement this function will change the replication to


DUPL a duplicate test.

END This function is used to start the End of Day Procedure (see chapter 6.2).

If it is necessary to look into the details of a measurement, this function


INFO will switch to the info screen on which all available information
concerning the sample is given, including a plot of kinetic test.

This function has the same effect as TEST, but is used in other modes (e.g.
MEAS Statistics etc.) and is used to return to the Test Selection Menu.

Allows the entry of a patient name in the measurement mode. This name
NAME will be printed with the result. The name can contain up to 15 characters.

This function is used to give a negative answer to a question from the


NO system or to deselect a calibrator in the Program Mode.

Allows the entry of a patient identification number during a


NUMB measurement. This number will be printed with the result. The number
can be alpha numeric and can be up to 8 characters long.

This key is used to change display pages, when the user is


PAGE entering/reviewing data consisting of more than one screen page.

After operating this function the printer will print the information present
PRIN on the screen. If this information is contained in more pages, all the pages
are printed.

Must be pressed to continue with the test after calculation of a


RDY calibration curve.

REAG This will force the Merck Microlab 200 in a (re)measurement of the Reagent Blank.

RPT During measurement this function is used to repeat a measurement. Mean


calculations will be done with the new result. This function is also used to

return to the test programming after the non-linear curve has been
plotted.

SING When starting a measurement this function will change the replication to a single
test.

SKIP During measurement this function is used to move fotward in the analyses
sequence, without measuring the requested reagent or sample. SKIP is
also used to override a program sequence.

TEST This function is used to return to the Test Selection Menu.

TRIP When starting a measurement this function will change the replication to a triplicate
test.

UTIL Operating this function will return the system to the Utilities Menu.

YES This function is used to give a positive answer to a question from the
system.

3.7.2 HARDWARE FUNCTION KEYS


Starts (re)measurement on what is present in the flowcell without
ABS sipping, or what is present in the cuvette. The sequence number will be
increased by 1 and patient data will be erased. If ABS is selected after
operating the PREV function, sequence number and patient data will be
maintained.
FLUSH Pressing this control will switch on the sipper pump in a continuous
mode, pumping 1500 VI. Pressing the key again will stop the pumping
action at the end of a complete cycle.

PAPER Advances the printer paper several lines.

PRIME Operating this key will activate the sipper system for 1 cycle at the
programmed volume, without starting a measurement.

3.7.3 NUMERICAL AND ALPHA KEYS


CLEAR This key clears an entry during data entry.

ENTER This key is used to confirm an entry via the keyboard.

for decreasing kinetic reactions, in order to obtain a positive result. The
minus key can also be used to include a "dash" in names etc.

3.8 SIPPER SYSTEM


When the bar below the Sipper Tube is pressed, the Sipper Pump is
activated. Shortly after aspiration of the sample, the measurement takes
place.

The red LED above the Sipper Tube indicates the status of the pump
action: i.e. continuously burning means aspirating, flashing means
dispens

4. SYSTEM FUNCTION

This section describes the operation to be performed on the Merck


Microlab 200 before the instrument is used for routine operation.

4.1 SWITCH ON
Switch on the mains power to the analyser. As soon as the Merck Microlab
200 is switched on it performs a self-check after which the following
screen is displayed (this screen is the main screen of the analyser):

When the Microlab 200 is switched on for the first time, figure 4-2 is
displayed. The system parameters then must be checked. This is done by
the routine SET SYSTEM PARAMETERS.

4.2 SET SYSTEM PARAMETERS


After pressing the UTIL key, the following screen is displayed:

after the measurement is finished, thus providing a better separation

I-FILTER SETTING
The Merck Microlab 200 is supplied with a standard set of 6 filters: 340,
405, 505, 546, 578 and 620 nm. The two positions marked are empty and
can be mounted with optional extra filters. By selecting I-FILTER SETTING
the user may change a value if a new filter is fitted.
Skip through the wavelength values by pressing ENTER until the desired
position is reached. Add the wavelength by keying in the right value
followed by ENTER.

lingual instrument. By pressing 2, a choice can be made between English,
German and Spanish.

and parity can be altered to meet the
communication protocol of the host computer. This is achieved by
pressing the appropriate number. The baud rate however is fixed at 2400.
Information about the computer protocol is shown in Appendix 1.
When all parameters have been set, press MEAS to return to the Test
Selection Menu.

5. PROGRAM MODE

5.1 OVERVIEW
The Merck Microlab 200 has been designed to provide flexibility in test
measurement, as well as a high degree of accuracy. There are 40 spaces
available for users to program their own test parameters. Tests already
programmed may be overwritten if necessary.

It is important, when the Merck Microlab 200 is new and the memory is
empty, to program the instrument in the right sequence:

5.2 PROGRAMMING CONTROLS


Press UTIL in the Test Selection Menu to obtain the Utilities Menu.

In total there are 15 free positions available, divided over 2 pages.


Select a free position to program a new control. The Merck Microlab 200
will ask to enter the name and the batch number of the control. Values
and limits are programmed during the programming of test parameters of
a test.
To re-program a control position, it first has to be deleted. Select DELE to
obtain the Delete Control Screen:

5.3 PROGRAMMING/EDITING A TEST


Follow the sequence as described in Programming Controls but select
3TEST PARAMETERS and the Test Survey Screen will be displayed.

The 40 tests are grouped into 4 blocks of tests. Each page contains 10
tests.

By pressing DELE a test can be deleted. The mode line on top of the
screen will change into "DELETE TEST". PROG will return the operator to
the Test Selection Menu in the
Program Mode.

The Test Survey Menu consists of a total of 40 positions, of which only


blocks of 10 tests can be displayed at the time. The other pages can be
displayed by pressing the PAGE key.

This selection is mainly used for enzyme activity tests. The reaction is
monitored during the whole programmed measuring time, at intervals of
0.5 second. The measurement is checked for linearity. If the non-linearity
is greater than the set limit for NON-LIN (see Chapter 5.3.2) and
AAbs/min is > 15 mAbs/min, there will be a warning message both on the
screen and on the printout.
ENDPOINT
This selection is used for measurement of normal endpoint reactions. The
endpoint of the reaction has normally been reached outside the analyzer
before the measurement run has started. Actual measurement is
performed in 2 seconds.
TWOPOINT
This selection is used for kinetic substrate measurements or kinetic
measurements where linearity check is not needed.

If a factor is to be entered press 2 and enter the factor value. The factor
may consist of a 5 digit number with a decimal point. If the reaction is a
decreasing reaction, the factor should be entered starting with a minus
sign

The position of the decimal point in the factor will be used to set
the decimal point in the final result.
3-S-BLANK
Press 3 to toggle sample blank between YES and NO. If YES is selected the
Merck Microlab 200 expects 2 different sample-/reagent mixtures to

To reach the programmed temperature in the flowcell of the reaction
mixture, the following minimum delay times are necessary:

The maximum programmable delay time is 999 seconds.


2-MEAS TIME
For kinetic tests the measuring time can be programmed in intervals of 2
seconds with a maximum of 998 seconds. Concerning the measuring
times please refer to the application sheet or to the package insert.

The Merck Microlab 200 always reports a reaction in AA/min, irrespective


of the measuring time program.

5.3.3 TWO-POINT
The process of programming for a two-point test is identical to the
options as listed under kinetic test.

The Merck Microlab 200 in this case does not calculate a rate reaction of
AAbs/min, but uses the difference in absorbance between the two
measuring points (dAbs).

5.4 NON LINEAR TESTS
When more than one calibrator has been programmed for a test, the
Merck Microlab 200 will automatically calculate a calibration curve. Values
can be entered manually or they can be obtained by means of
measurements. After calculation, the calibration curve is displayed:

The analyser is capable of producing a calibration curve from a series of up


to 10 standards. The mathematical approach is that of a cubic spline fit. A
3rd degree regression curve is calculated through each grouping of 3
successive calibration points and the complete combination of the derived
curve is fitted together to produce 1 curve.

The cubic spline fit does not always produce the best results with the
calibration curves which show an inherent logarithmic relationship. In
these cases, the software has been designed to systematically test the
absorbance/concentration data against both logarithmic and an inverse
logarithmic function in order to obtain the best fit.

In total, 25 different approaches are tried, including various polynomial


equations, to obtain the best universal solution and the most "linear"
relationship between absorbance and concentration.

Once the "best fit curve" has been established, various smoothness factors
are applied to obtain the required accuracy of fit.

The required accuracy is user-selectable in percentage terms and may be


changed at any time without the need of having to re-run the standards.

Changing the calibration curve can be done by pressing CHNG, after which
a new accuracy percentage can be entered. The higher this figure, the
straighter the curve will be.

Pressing RPT will allow the possibility of reprogramming the test.

must be cleaned first with detergent and rinsed afterwards with water.
Place a bottle with a 5% detergent solution (e.g. Extran MA 01, Merck art.
number 1.07555.) under the sipper tube and press GO. The flowcell is
cleaned for about 2 minutes. After this the Merck Microlab 200 asks to
place a bottle with water under the sipper tube. The flowcell is rinsed with

6.4 BICHROMATIC MEASUREMENTS
When in the test parameters of the test the second wavelength is selected
for an end-point test, the Merck Microlab 200 automatically measures the
reaction mixture at 2 different wavelengths. Each actual measurement
takes 2 seconds and some time is needed to change the filter. The results
of the 2 measurements are subtracted. This value is then used in the
further calculations.

7. STATISTICS

7.1 INTRODUCTION
The statistics program is located under position 3 of the Utilities Menu.
This program gives the possibility of statistical evaluation of controls
measured on the Merck Microlab 200.

For all 40 test programs in the unit, two controls can be defined, which can
be selected from 15 different control sera.

The system will store the last 30 results on a rotating basis, so that as
each new value is added, the oldest value is deleted. After each 30
results, an automatic print-out of the Levey-Jennings plot is made.
The statistical data that are available include:

The Levey-Jennings Plot displays the high and low ranges. The most recent

8. REPLACEMENTS

8.1 REPLACING /ADJUSTING LAMP


Proceed to the UTILITIES menu, and select 4-SERVICE, 2-ADJUST LAMP.
The following screen is displayed:

Remove the sipper tube and remove the aspirate lever by simply pulling it
down from the black block.

Remove the three mounting screws at the top of the rear panel (marked
with arrows), and gently take off the top cover by moving it towards the
front of the instrument and then lifting it. The cover can be placed on its
side, at the right hand side of the unit.

Lamp Block

Remove the lamp block completely by loosening screws 1 and 2. The lamp
block can be removed by lifting it out carefully.
Pull the lamp out of its lamp holder.

Insert a new lamp in the lamp holder.

Reinsert the lamp block in its place and lock screw 1.

Adjust screw block in its middle position with screw 3 (see Figure 8-2).
Then press GO.

Adjust screws 1 and 3 in sequence in order to obtain a minimum


absorbance reading. The absorbance is displayed in the upper right corner
of the display. Press END and fasten screw 2.

Carefully put the top cover back in position, lock the three mounting
screws at the rear panel and reinsert the sipper tube and aspirate lever.

8.2 REPLACING PAPER ROLL


Open the paper holder on top of the printer by pulling it backwards and
then lifting it up. Load the printer paper into the printer so that the paper
feed is from under the roll (see Figure 8-3).

Press PAPER on the keyboard to load the paper.

8.3 REPLACING FUSES


The fuses are located at the rear panel of the analyser. They are mounted
in the fuse holder between the power switch and the mains cable
entrance. Pull out the fuse holder lid and the fuses can be replaced
easily.

The fuse holder will only take the European style 5x20mm glass fuses.

Should the fuse blow immediately again after replacement, never mount a
more powerful one, but contact your local service organization.

9. SERVICE AND MAINTENANCE

9.1 DAILY
The Merck Microlab 200 may be cleaned with a damp cloth and some
detergent. Make sure to remove the mains connector before cleaning and
take care that no water gets into the instrument.

The only daily maintenance of the Merck Microlab 200 is:


Check printer paper (if connected) and assure that there is
uay 1 um.
Empty waste container.
Follow the instructions on the screen at the end of the test
procedure and the end of the day procedure.

9.2 OCCASIONALLY
Sometimes due to insufficient washing or long periods of inactivity, the
silicon tubing in the pinch valve might get blocked, causing:
non aspirating of sample, or
liquid leakage from the pressure relief system of the pump, through
the outlet at the bottom of the instrument.
If this happens, proceed as follows:
Disconnect the instrument from the power outlet.
Remove the top cover (see Change Lamp)
Stretch the silicon tubing passing through the pinch valve and roll
between the fingers, trying to "unstick" the tubing in the pinch
area.
Check for correct operation before mounting the top cover. If the
Merck Microlab 200 functions properly, replace the cover. The
internal tubing system has to be replaced only once a year.
However, tubings might get polluted due to lack of maintenance,
and may cause damage to the instrument. When this is observed,
no warranty claims will be accepted.

9.3.1 ABSORBANCE ON SCREEN
This mode can be used to read absorbances from different samples without
the use of a full program.
Select the Utilities Menu followed by 4-SERVICE. The following screen is
displayed:

Analyser Control

I-ANALYSER CONTROL
With the selections 1-5 all functions can be set and checked for proper
operation. On the second half of the screen, readings of the settings are
made. For example: when 2s-TEMPERATURE is set to 25, after a while the
reading TEMPERATURE will change from O to 1 indicating that the
selected temperature has been reached.
2-SIGNAL CONVERSION
This function is extensively described in the Service Manual and is for
service purposes only.

APPENDIX 2 - FAULT FINDING

In this appendix errors and their causes are described. Possible problems
are divided in 3 sections: Flowcell
Results
General
In case a given remedy does not solve the problem, please contact your
local service organisation.

2.1 FLOWCELL
2.1.1 NO SAMPLE ASPIRATION
System does not aspirate sample in any mode (also not in flush), but
the prime LED indicates normal operation and the pump seems to
operate normally.

CauseRemedy

Obstruction in the fluid system. Check all tubing and flowcell for
any blockage.
