We conducted our experiment on a sandwich that was left aside for a couple of days.

Then, we put 10 g
of it in the stomacher bag to be digested with 90 ml broth (diluted by 1/10). After that we took 1 ml to a
tube containing 9 ml broth (dilution by 1/100), then from this tube 1 ml to another (diluted by 1/1000)
and finally 1 ml from the second to a third tube (diluted by 1/1000)

We took 0.1 ml from each tube and spread on a PCA and MSA mediums.

In the PCA plates that grow everything the first one has no colonies but the dilution is 1/100 so we have
less than 100 cfu/g.

In the second two plates the colonies are too numerous to count.

Concerning the MSA plates which grow staphylococcus aureus, in the 3 plates we have less than 20
colonies so we pick the least dilute (1/100)

We have 14 colonies

Cfu/ml = (14 x 100)/0.1 = 14000 cfu/ml
