1. Summary

이 논문의 주요 내용은 UPS(ubiquitin-proteasome system)의 DUB(deubiquitinating

enzymes)중 하나인 USP14 이 어떤 영향을 주는지 그리고 USP14 inhibitor 인 IU1 을
처리해주어 protein degradation 이 어떻게 변화하는지 확인하는 것이다.

Proteasome 에 의해 hydrolysing activity 를 가지는 Ub-AMC 를 이용한 assay 와 Western

blotting 을 이용해 USP14 의 deubiquitinating activity 가 proteasome 에 의해 activate 되고
chain trimming 을 통해 proteasome 에 의한 protein degradation 을 inhibit 함을 확인하였다.

USP14 이 protein degradation 에 어떻게 관여하는지 확인하기 위해 wild-type USP14,

USP14(CA) (non catalytic effect), USP14(SF) (UBL domain 과 catalytic domain 사이 mRNA
서열을 delete), USP14(△UBL) (proteasome binding site 를 delete)을 이용하여 western
blotting 을 진행하였다. Neurodegenerative disease 에 critical 한 protein 인 Tau 와 TDP-43
에 대하여 USP14 이 protein degradation 을 inhibit 하고 USP14(CA)의 경우 degradation 을
accelerate 함을 알 수 있었다.

여러 분자 중에서 screening 을 통해 IU1 이라는 분자를 선택하였다. IU1 은 USP14 의

active site 에 결합하는 inhibitor 로 USP14 에는 영향을 주지만 다른 DUB 에는 영향을 주지
않다는 것을 확인하였다. Proteasome 과 함께 처리해주었을 때 USP14 에 영향을 주는 것을
보아 USP14 의 activated form 에 specific 하게 결합하는 것을 알 수 있다. USP14 을 inhibit
하기 때문에 protein degradation 을 가속시킨다.

In-vitro 뿐만 아니라 in-vivo 에서도 IU1 을 처리하여 주었을 때 Tau degradation 을

enhance 함을 확인할 수 있었다. 또한 추가적으로 proteotoxic mechanism 에 연관된 protein
들도 IU1 에 의해서 degradation 이 일어남을 확인하였다.

저자들은 IU1 의 특징을 이용하면 misfolding 등으로 인하여 toxic 한 protein 들을

proteasome 의 activity 를 높임으로써 제거하여 neurodegenerative disease 와 같은 disease
에 치료에 사용될 가능성을 이야기하고 있다.

2. Results 부분의 verb

When such ‘VS-proteasomes’ were reconstituted with recombinant USP14(Supplementary

Fig. 2), UB-AMC hydrolysing activity was increased 800-fold over that of isolated USP14.

The amino-terminal ubiquitin-like(UBL) domain of USP14(Fig. 1d) is its principal

proteasome-binding site, and accordingly deletion of the UBL domain abrogated the
USP14 effect (Fig, 1c).

IU1 induced dose-dependent reduction in tau levels, with a strong effect seen at 50 μM .

3. Introduction & Discussion 부분의 verb

The extension and disassembly of chains at the proteasome may alter substrate
degradation rates by changing substate affinity for the proteasome.

May alter: 바꿀 수 있다.

Thus, removal of the ubiquitin chain by RPN11 can promote substrate translocation into
the CP to be hydrolyesd.

Promote: 촉진시키다.

Whether chain trimming by UCH37 can suppress proteasome activity as powerfully as

USP14 will require further study.

Suppress: 막다. 억제하다.
