2. Explain How a colorimetric technique determines the concentration of a sample.
Discussion:

Analytical methods based on the absorption of radiant energy are called absorptiometry methods,
which utilize the visible region of the spectrum. They are also known as colorimetric methods. Some of the
methods of analysis are of great importance, particularly in the determination of glucose, ketone bodies,
occult blood, pH and protein in biological specimen. Interaction of matter and radiant energy also occur in the
other regions of the electromagnetic spectrum. Qualitative and quantitative methods are based on these
interactions

Materials: Clear colorless plastic cups (10 cups)


1 small sachet Coloring Dye
Water
Procedure:
1. Dissolve the small Sachet of the coloring dye into a plastic cup that contains water as a solvent Label the cup
as Cup A

2. Transfer half of the contents of Cup A into a new Cup, and then Add enough water to make it full, and then
label it Cup B.

3. Then transfer half of the contents of Cup B into another new Cup, then add enough water to make it full, and
then label it Cup C.

4. Repeat the Process until you can reach to Cup J.

5. Arrange the following Cups from Cup A - Cup J then place the picture below.

CUPS A - CUP J

1. Based in the Color scheme, which from the following cups has the strongest color?

Cup A has the strongest color.

2. Based in the understanding and procedure which of the following has the Highest concentration?

Based on the results and findings with proper understanding, Cup A has the highest concentration.

3. Based in the understanding and procedure which of the following has the Least Concentration?

Based on the results and findings with proper understanding, Cup J has the least concentration.

4. Base on your Observation what are the concepts stated in Beer’s Plot and Is it Applicable to the Observation
that you have?

According to Beer’s law, absorbance is directly proportional to the concentration of the absorbing
species. The higher the concentration of the mixture the higher its ability to absorb light that passes
through it, like how Cup A is the most concentrated and only allows a small amount of light to pass
through it while in Cup J it is the least concentrated and allows more light to pass through it.
