Activity No. 6: Stability of Proteins


Objectives
At the end of the experiment, student should be able to:
 Identify the physical and chemical properties of proteins.
 Identify denaturants and describe their effects on proteins.
General Procedure:
Perform the following tests using 10ml solutions of the following as samples:
casein, egg albumin and gelatin. Label samples. In these activity, coagulation
or precipitation will indicate denaturation.
A. Heat Denaturation
1. Heat 2ml of each of the sample in different test tubes then label.
2. Observe if it coagulates.
3. Add 2 drops of 1.0% acetic acid in each test tube and observe the effect.
4. Record the result

B. Isoelectric Precipitation
1. Obtain 2mL of each of the sample in different test tubes then label.
2. Add 0.1N HCl drop by drop until maximum precipitation occurs in each sample.
3. Continue the addition of HCl, shaking the tube after each addition, and note that the proteins
redissolve.
4. At this point, add 0.1N NaOH drop by drop in each test tube. Note whether the protein is
precipitated and redissolved in an excess alkali.

1. Obtain 1ml each of the sample in different test tubes then label.
2. Add 2 drops of hydrochloric acid on each sample.
3. Observe the effect

Concentrated mineral acids Observation

D. Organic Solvents
1. Obtain 2mL of the each of the sample in different test tubes then label.
2. Add 1 drop of 1.0% acetic acid and 4mL of 95% ethyl alcohol in each test tube.
3. Mix thoroughly.
4. Observe the characteristics of the precipitate.
