1. Overview
1.1 System structure
The appearance of the Erba Lisawash Elisa Washer is as shown in figure 1:

The system consists of pumps, controlling mechanism, wash head, plate tray, circuit,
and bottles. The pumps include a water pump for transporting detergent to wash head
and
a vacuum pump for evacuating the waste from the ELISA plate. The controlling mecha-
mech
nism is primarily used to control the up/down and left/right movement of wash head.
Wash head is available in 8 or 12 way manifold for 8×12 and 12×8 plates respectively.
The circuit is an advanced single chip system
stem composed of high speed CPU.

ation extremely convenient and easy.


The system includes a distilled water bottle, two wash bottles, and a waste bottle with
overflow sensors.

1.2 Basic Principles


The Elisa Washer is used for immunological determination ELISA.
Under control of the valve, the detergent is transported by the water pump to the ELISA
plate on the plate tray, through the wash head. The wash head moves up/down and
left/right to add the detergent into the plate wells. After a certain time of soaking, the
vacuum pump sucks the Microwell contents to the waste bottle. The ELISA plate can be
washed efficiently through repetition of this process.

3. Installation
3.1 Unpacking
Upon opening the package, check the main unit and accessories against the packing list.
If you find anything damaged or missing, please contact your supplier immediately.

3.3 Environmental Requirements


The environment should be free of dust, corrosive gases, strong direct electromagnetic
interference and sunlight. The working platform should be firm and reliable, and the room
should have good ventilation.

3.4 Connection
3.4.1 Connect the air pump tube (blue) on the waste bottle to the Air Pump port (blue) on
the back of the device, as shown in figure 2.

3.4.2 Connect the waste tube (green) on the waste bottle to the Waste port (green) on
the back of the device.

3.4.3 Connect the tube (yellow) on the wash bottle to the Wash port (yellow) on the back
of the device.

3.4.4 Put the duct next to the valve into the round hole of the valve by pressing the valve
core on the top of the valve.

NOTE: The duct must be led to the round hole.


3.4.5 Insert the overflow sensor wire (gray) on the waste bottle into the Overflow Sensor
plug on the back of the system.

3.4.6 Insert the two-wire cord (water pump supply lead) on the detergent bottle into the
Water Pump port on the back of the system.

3.4.7 Connect a proper ground wire to the Grounding on the back of the system.

3.4.8 Set the Power switch to off (O). Then insert the power cord into the Power socket
on the back of the device and switch on the power.

3.4.9 Unscrew the wash bottle cap (hold the cap with your left hand and rotate the bottle
clockwise with your right hand) and add sufficient detergent, then tighten the cap.

3.4.10 Add sufficient distilled water into the water bottle.

3.4.11 Tighten the waste bottle cap.

NOTE: When fixing the bottle cap, check if the white O-ring in the cap is
damaged or placed correctly.

3.4.12 Position the plate tray on the four poles, in the front base of the device, and push
it into its place.

NOTE: The Priming trough should be on the left.

3.4.13 Fix the 8-head (or 12-head) wash head on the wash arm. Sleeve the thinner tube
of the wash arm on the inlet port of the wash head and the thicker one on the outlet port.

4. Operation
4.1 Power-on
Switch on the system, it starts initialization and then the Main Menu is displayed on the
screen.

4.2 Operation Procedures

4.2.1 System Settings


This function is used to set the number of wash heads, start position of wash head and
pump speed.

4.2.1.1 Wash Head Settings


You should select corresponding wash head type according to the actual device config-
uration. Method: Touch System Settings (indicates being selected) and Wash Head
Settings in the submenu, then select “8” or “12” accordingly.

4.2.1.2 Start Pos. Settings


This function is used to adjust the wash head from the start position to the center of the
wells of the first row (Note: Since the start position of the 8- and 12-head wash heads is
different, it should be reset after replacing the wash head). Method: Touch Start Pos.
Settings and select the wash head type, then touch Increment or Decrease to set a
value (8-head 330 by default; 12-head 490 by default). Touch OK and go to “Demo”
menu. Touch Yes. Check whether the wash head stops at the center of the first row.
Increase or decrease the value until the required setting is reached. Then check it by the
demo program again. After the adjustment, touch OK and No to return.

4.2.1.3 Pump Speed


There are two flow speeds available for the pumps (you can control the prime volume
and prime pressure through the valve). Method: Touch Pump Speed and select 1 or 2 (2
is faster than 1), then touch OK to confirm and return.

4.2.2 Plate Settings


This function is used to set the V, U, or Flat bottom plate. Different plates are stored in
different No. (1-8), which is easy to refer when editing wash programs.
Method: Touch Plate Settings and set a plate Number in the submenu. In the Prime
Height screen, select a height (about 70), then touch Demo to check whether the longer
probes on the wash head reach the desired height. Increase or decrease the value till
desired setting is reached. After confirming with the demo program, touch No to proceed
to Aspirate Height. Select a height about 150 (you’d better increase from Low to High to
avoid damaging the wash head), then touch Demo to check if the longer probes on the

wash head reach the well bottom with a spacing of about 0.3-0.5mm. If not enough, in-
crease; otherwise decrease. Then touch No to proceed to Crosswise aspirate . The
aspiration of the fluid in the well bottom is done by moving the probes back and forth.
This function only applies to the flat bottom plates. Suppose plate No.1 is a flat bottom
plate, you can select Yes and then proceed to Range, which is used to set the range
setting to clean the well bottom. Set a value within 2-40 (about 30), then touch Demo to
check and make sure the probes will not touch the wells. When confirmed all the relevant
data will be saved in No.1 plate. Return to the main menu and proceed to settings of
No.2…8 plates.

4.2.3 Program Settings


Based on ELISA assay requirements, we must have different wash modes and proc-
esses, namely wash program. Program Settings is where you can edit various wash
modes and save them for future use.
Method: Touch Program Settings and select a program no. (1-40).
Select a plate Number to select a Microwell plate type. Proceed to Moving Cycle (1-9).
Set a value and touch OK to proceed to Row Cycle. To wash for 5 times, if you select
“Yes” for Row Cycle, then it will wash every row for 5 times and go to the next row; if you
select “No” for Row Cycle, then it will wash from the first row to the 12th (8th) row and then
repeat for another 4 times.
Select Yes or No . Proceed to Aspirate Cycle (1-9), set a value, and confirm.
Go to Prime Vol. (50μl - 700μl), which is commonly set to 300μl. Set a value and confirm.
In Prime Bottom setting, select Yes or No . Proceed to Prime Bottom Vol. (50μl - 700μl).
Set a value and confirm and go to Prime Bottom Cycle (1-9), which refers to number of
the well bottom washing. Set a value and confirm. For the Soak Time (0-255s), set a time
as per the assay protocol and confirm. Then this wash program will be saved. Return to
the main menu and proceed to settings of the next program.

4.2.4 Wash Program


You can start washing the plate when you finish the settings for the system and plate
program.
Method: Touch the Wash Program button and select a program No., you can select the
program you want from the programs set. Confirm and proceed to Rows No. (1-12). For
8×12 plates, the maximum is 12; for the 12×8 plates, the maximum is 8. Set a value and
confirm, then the system will start washing the plate. During washing process, touch
Pause to pause. Then select Stop to quit or Resume, for continue. If the waste bottle is
full during washing, the screen will display “Waste Overflow” and the washing will stop.

4.3 Shutdown
When the system is not used, the buffer salt in the detergent may be solidified, this
blocking the wash head. To avoid this situation, perform the distilled water prime.

4.3.1 Daily Shutdown


Before shutdown of the system, touch the Wash Program button and run the distilled
water prime. After washing, switch off the power supply and empty the waste bottle, then
fill the Priming trough on the plate tray with distilled water and soak the wash head.

4.3.2 Shutdown for Long Time
Wash the whole fluid system with distilled water prime for several times. Take out the
duct from the valve. Empty the detergent bottle and waste bottle, and then clean them
with distilled water.

4.4 Precautions:
4.4.1 Empty the waste bottle regularly.

Dispose of the waste in accordance with your hospital guidelines for


biohazard waste disposal.

4.4.2 Before starting the ELISA wash, check if the No. of heads of the installed wash
head is in accordance with the settings. If you only want to wash part of the plate, please
select corresponding No. of rows.

4.4.3 The selected type of plate should be consistent with the one you are using.

Note that the programs with Crosswise aspirate function can’t be


applied to the V, U bottom plates, for it may damage the wash head.

4.4.4 It is strongly recommended to run wash program #41 for 2 times after the daily
work.

5. Maintenance and Storage


5.1 Cleaning
The detergents used can be corrosive, so regular cleaning becomes very necessary.
Please take out the plate tray, clean the device with a soft cloth with neutral detergent
and dry it. Clean the cover of the device regularly.

5.2 Maintaining and replacing the duct


The duct is probably unable to restore as a result of long closing of the control valve.
However, you can press the valve core on the top of the valve and pull out one end of the
duct a little.
Replace the duct after being used for a long time.

5.3 Replacing the wash head


Refer to 3.4.13.

5.4 Unclogging the wash head


Remove the wash head and then unclog the probes of the wash head with the spare
needle.

5.5 Replacing the fuse


The fuse is located in the lower part of the power socket on the back of the device. Open
the fuse holder with a screwdriver and then replace the fuse with the spare 2A fuse.

5.6 Storage
The device should be stored in a room with good ventilation and relative humidity no
greater than 80% and without corrosive gas.

6. Troubleshooting

7. Warning
Erba Lisawash is a compact in vitro diagnosis instrument. It may bring risks to the pa-
tients indirectly, and accompanying risks to the operator and environment. To avoid
these risks, be sure to read the following instructions carefully.
7.1. This device adopts AC 220V/110V which may bring electrical harm to
human body. Don’t open the rear cover before switching off the power
supply. If you need to open the rear cover to repair the device, contact our service per-
sonnel or professionals.
7.2. Since the patient samples may contain germs or virus which will cause
disease, be sure to dispose of the waste in accordance with relevant
regulations of your hospital. Transport and store the patient samples and
handle the sample tubes strictly as instructed in relevant regulations of your hospital.
7.3. Don’t operate the device unless you have read this Operation Manual
carefully.
