Gram Stain

Methodology:
 Isopropil
 Safrain
 Destilled Water
 Lugol
 Crystal Violet
 Inmerssion Oil
Experiment description:
We wanted to use a staining method, to classify negative and positive bacterias with pink
and purple (violet).
Step by step:
1. First, one teammate give us a saliva sample of her (Dana).
2. Then in the sample, we put 1drop of crystal violet, and carefully we spread the drop
and wait 1 minute.
3. We wash the sample with destilled water and let it dry.
4. Then ,we put Lugol so the crystal violet stick to the sample.
5. We wash again with destilled water and put isopropyl and wait some seconds.
6. Then we put safrain, we put one drop, and repeat the same process of the crystal
violet. (2-3)
7. The we put inmersion oil and carefully put it in the microscope.
Expectations:
I expect to find positive and negative grams, and dye them on our sample.

Observations results:
We could see a little bit of the color purple, we didn’t see any pink.
Conclusions:
We suppose that when one teammate let fall the sample and dry it accidentally, so that’s
our explanation for why the purple we didn’t see it correctly, and for the color pink, we
suppose there weren’t any positive grams.
