Batch Culture:

Batch culture is a type of suspension culture where the cell material


grows in a finite volume of agitated liquid medium.

For instance, cell material in 20 ml or 40 ml or 60 ml liquid me-


dium in each passage constitute a batch culture. Batch suspension
cultures are most commonly maintained in conical flasks incubated
on orbital platform shakers at the speed of 80-120 rpm.
Dedifferentiation:

1. It is the process in which the differentiated cells regain their property to divide.
2. The fate-committed cells revert to a state where they re-enter the cell cycle under
cellular divisions.
3. It is widely noticed in plants.
4. Plants undergo dedifferentiation of parenchymal cells to form meristem tissue.
5. As a result, the differentiated parenchymal cells form cork cambium and
interfascicular cambium.

6. Indirect Androgenesis:
Microspores undergoes division in unorganised fashion to give
rise to callus and by embryogenic or organogenic induction the
haploid plantlets may be obtained.

Thawing
Remove the cryovial containing the frozen cells from liquid nitrogen storage and
immediately place it into a 37°C water bath. Quickly thaw the cells (< 1 minute) by
gently swirling the vial in the 37°C water bath until there is just a small bit of ice left in
the vial. Transfer the vial it into a laminar flow hood.

Plating efficiency
Plating efficiency ("PE") is a measure of the number of colonies originating from
single cells. It is a very sensitive test and is often used for determining the nutritional
requirements of cells, testing serum lots, measuring the effects of growth factors,
and for toxicity testing.
