1. What are the different viruses to be cultured and how many mL of inoculum is needed using
the following sites:
VIRUS INOCULUM (ml)

2. When is it suitable to use the intravenous and intracerebral site in chick embryo?
ANSWER: Embryonated chicken eggs are inoculated by the allantoic route at approximately the
middle of the 21-day embryonation period, at 8–10 days of embryonation; they are inoculated by
the amniotic route late in the incubation period, at 14–16 days of embryonation.
3. Describe the different signs that you will observe in the inoculated chick embryo that marks
an infection.
ANSWER: At embryonic day 12, β-galactosidase-expressing PRV strains were injected into the
vitreous humor of one eye, and virus replication and spread from the eye to the brain were
measured by β-galactosidase activity and the recovery of infectious virus from tissues. The wild-
type PRV strain, Becker, replicated in the eye and then spread to the brain, causing extensive
pathology characterized by edema, hemorrhage, and necrosis that localized to virally infected
tissue.
