Bài 3: Key aroma compounds in fermented Forastero cocoa beans and changes induced by
roasting

Abstract
The infuence of the roasting procedure on the composition of key aroma compounds in fermented
Forastero cocoa beans was investigated. For this purpose, the volatile fractions from the unroasted,
fermented and the roasted cocoa were isolated by extraction/SAFE distillation, and the odour-active
constituents were characterised by gas chromatography–olfactometry in combination with an aroma
extract dilution analysis. A total of 41 aroma compounds in the favour dilution (FD) factor range of 2–
8192 were identifed in the unroasted and 42 compounds with FD factors of 2–8192 in the roasted cocoa
beans. Qualitatively, the set of aroma compounds was nearly identical in both samples; however,
diferences in the favour dilution factors were observed. 2- and 3-Methyl butanoic acid (sweaty) and
acetic acid (vinegar-like, sour) showed the highest FD factors in the unroasted beans, while 3-
methylbutanal (malty), 4-hydroxy-2,5-dimethyl-3(2H)-furanone (caramel-like) and 2- and 3-methyl
butanoic acid (sweaty) were detectable by GC/O in the highest dilutions in the roasted seeds.
Quantitation of the most odour-active compounds by means of stable isotope dilution assays, followed
by calculation of odour activity values (OAV, ratio of concentration to odour threshold), revealed
concentrations above the threshold level for 20 compounds in the unroasted and 24 compounds in the
roasted beans. The roasting procedure led to a strong increase, in particular, in the concentrations of the
two Strecker aldehydes 3-methylbutanal (malty) and phenylacetaldehyde (honey-like) as well as 4-
hydroxy-2,5-dimethyl-3(2H)-furanone (caramel-like).

Abtract:
Application of a comparative aroma extraction dilution analysis on unroasted and roasted Criollo cocoa
beans revealed 42 aroma compounds in the flavor dilution (FD) factor range of 1-4096 for the
unroasted and 4-8192 for the roasted cocoa beans. While the same compounds were present in the
unroasted and roasted cocoa beans, respectively, these clearly differed in their intensity. For example,
2- and 3-methylbutanoic acid (rancid) and acetic acid (sour) showed the highest FD factors in the
unroasted beans, while 3-methylbutanal (malty), 4-hydroxy-2,5-dimethyl-3(2H)-furanone (caramel-
like), and 2- and 3-methylbutanoic acid (sweaty) were detected with the highest FD factors in the
roasted seeds. Quantitation of 30 odorants by means of stable isotope dilution assays followed by a
calculation of odor activity values (ratio of the concentration/odor threshold) revealed concentrations
above the odor threshold for 22 compounds in the unroasted and 27 compounds in the roasted cocoa
beans, respectively. In particular, a strong increase in the concentrations of the Strecker aldehydes 3-
methylbutanal and phenylacetaldehyde as well as 4-hydroxy-2,5-dimethyl-3(2H)- furanone was
measured, suggesting that these odorants should contribute most to the changes in the overall aroma
after roasting. Various compounds contributing to the aroma of roasted cocoa beans, such as 3-
methylbutanoic acid, ethyl 2-methylbutanoate, and 2-phenylethanol, were already present in unroasted,
fermented cocoa beans and were not increased during roasting.

Fine flavor cocoa is worldwide renowned to produce origin chocolates with special aromas,
e.g. fruity or floral, in addition to its chocolate aroma. This research aims to elucidate fine
flavor dynamics during fermentation by analyzing the sugar, free amino acid (FAA) and
volatile profile. Ecuadorian Trinitario beans (Sacha Gold) were sampled after 0, 18, 24, 48 and
66 h of spontaneous fermentation. The unfermented beans contained significant sucrose,
glutamic acid and asparagine amounts while the fermented beans (66 h) contained more flavor
precursors, e.g. glucose, fructose, hydrophobic and other FAA. Forty-one volatiles were
identified, including 13 fruity- and 12 floral-like, derived from various metabolic pathways.
Whereas the level of fatty acid-derived fruity volatiles decreased, the amount of amino acid-
derived fruity and floral volatiles increased and floral terpenes remained stable. Some fine
volatiles were assumed to be pulp-derived (e.g. linalool, β-myrcene, 2-heptyl acetate) or
intrinsic to the bean (e.g. 2-heptanol, 2-heptanone, 2-pentanol), while others were generated
during fermentation by microbial synthesis (e.g. 2-phenylethanol, isoamyl alcohol).
Multivariate analysis clustered samples according to fermentation time and quality. These
findings demonstrate that cocoa fermentation is essential for the formation of flavor precursors
and the development or preservation of important fine aroma compounds. Trinitario (or
hybrids), one of the cocoa varieties with fine flavor potential, is cultivated all over the world
and hence, care should be taken during post-harvest to fully exploit this fine flavor character
and deliver high-quality cocoa beans with fine sensory characteristics.

Bài 1: Most of the oxygenated aliphatic compounds and aromatics were potentially
derived via an acetic acid and shikimic acid pathways, respectively. Untreated cocoa beans
contained 12 sesquiterpene hydrocarbons potentially derived through the mevalonic acid
pathway and contributed to harsh notes.
Bài 2: Fine aroma precursors produced in the pulp during the fermentation phase will
migrate into the bean when it’s permeability is improved and then retained during the drying
phase.

Các tiền chất của mùi hương trong hạt cacao sinh ra sau quá trình lên men là đường khử
(glucose và fructose) và các acid amin tự do kỵ nước (Castro-Alayo et al., 2019; Rottiers et al.,
2019; Saunshia et al., 2018). Từ những tiền chất hương này, các hợp chất chính có vai trò tạo ra
mùi hương đặc trưng cho hạt cacao pyrazine và aldehyde (Castro-Alayo et al., 2019; Saunshia
et al., 2018), ester, alcohol, acid, ketone, hydroperoxide, hydrocarbon (12 sesquiterpene)
(Saunshia et al., 2018) được tổng hợp bên trong của hạt cacao và được giữ lại sau những quá
trình xử lý kế tiếp.
Nhiều nghiên cứu đã kết luận rằng quá trình rang không ảnh hưởng lên thành phần hợp chất
