PRACTICUM OF EXPERIMENTAL PHARMACOLOGY AND TOXICOLOGY

Practicum title : Experiment I. Effect Of Administration Route On Drug Absorption

Name : leonardo demas krisna satria wijaya
Student number : 21/475052/FA/13028
Class, group : IUP/1
Practicum date : Wednesday, March 1st 2023

A) Practicum Aim :
To know, to practice, and to compare the techniques of drug administration to the speed of
their absorption, through analyzing the pharmacological data.

B) Method :
a) Equipment :
- Injection syringe and needle (1-2 ml)
- Blunt tipped needle/sonde (for oral)
- Gloves
- Stopwatch.
b) Material :
- Ketamine injection 100 mg/ml
- Xylazine injection 20 mg/ml
- Water for injection
- Alcohol 70%.

c) Workflow :

Preparation of a ketamine/xylazine cocktail for p.o, i.p, i.v., and s.c. Administration.
0.2 ml of ketamine 100 mg/ml are taken and put in a tube (sterile)

0.2 ml xylazine 20 mg/m is added.

4.6 ml water for injection is added.

Labeled " k/x encer"

Preparation of a ketamine/xylazine cocktail for i.m. Administration.
0.2 ml of ketamine 100 mg/m is taken and put in a tube (sterile)

0.2 ml xylazine 20 mg/m is added

4.6 ml water for injection is added

Drug administration to rats

Mice are weighed one by one

The drug cocktail (ketamine dose of 65 mg/kg bw and xylazine dose of 13 mg/kg bw)
is administered to the rats

Observed and recorded the time of loss of the reverse reflex of the body

d) Data analysis :
After the test animal has received treatment, observe carefully, and record the time when
the turning reflex is lost, marked by the loss of the test animal's ability to turn over from a
supine state. Calculate the onset and duration of sleep time for each experimental group
and compare the results using statistical test of unidirectional pattern analysis of variance
with 95% confidence level.
