Introduction:
Recombinant DNA technology or genetic engineering is a method that allows scientists to study, improve, or manipulate the genetic material of a cell to create
genes of interest or transgenes. The technology allows the transfer of a specific gene from different organisms to produce a new trait in an organism. The DNA
combined from different cells is called recombinant DNA and the organisms produced by the technology are called GMO’s or transgenic organisms. Genetic
engineers can now employ the molecular tools and techniques that allow manipulation of DNA sequences in the laboratory to identify genes that control a
desired trait, extract them, copy them, and insert them into another organism.
Direction: The steps involved in the process of genetic engineering of microorganism are
shown in the figure below to illustrate the introduction of the human insulin
gene into bacteria. Analyze and interpret the figure below and answer the
following questions:
1. What are plasmids and tell how they are used in genetic
engineering? _______________
____________________________________
4. Read different reference materials to gain more information about recombinant DNA
technology. From your readings, supply the missing word to complete the basic steps
Step 1. The _____ of interest which is human insulin, is isolated from the human DNA
and cloned. The _____ of DNA means making multiple exact copies of DNA or
gene.
Step 2. The human insulin gene is inserted into a _____, a plasmid at specific sites cut by
2
an enzyme called restriction enzyme. A plasmid which can carry a DNA fragment
contains the promoter elements and terminator sequence that will work for the
bacterial cells.
Step 3. The recombinant plasmid is introduced into bacterial cell by physical means. The
_____ are subjected to heat and then put an ice and this will result into
formation of pores in the cell membrane that allow the DNA to get inside the cell.
This process is called transfection or transformation. The _____ plasmid is
multiplied into many copies in the cell.
Step 4. This process results in the production of many copies of the inserted human insulin
gene. This is called _____ of the human insulin gene by bacterial cells.
Direction: Select the steps involved in the genetic engineering of corn with the Bt gene to
produce insect-protected corn in the proper order.
1. 2. 3. 4. 5.
Direction: Transgenic organisms are organisms that have been transformed by the insertion of one or more foreign gene(s) obtained from another organisms.
“Foreign” means the donor organisms belong to different species. Here are some of the genetically modified organisms, also called transgenic organisms.
Write the peculiar traits they acquired through recombinant DNA technology.
