Preparation and Characterization of Highly Stable and Substrate


Diffusible Cross Linked Enzyme Aggregates of Maltogenic
Amylase from Bacillus lehensis G1

Abstract
Introduction: Maltogenic amylase from Bacillus lehensis G1 (MAG1) is a potential
biocatalysts for oligosaccharides synthesis. However, poor stability and lack of reusability of
the free enzyme presents a significant barrier in its industrial application. To overcome these
problems, formation of cross linked enzyme aggregates (CLEA), a carrier-free enzyme
immobilization technique was applied on the free MAG1. However, the substrate diffusion
problem is the main bottleneck in CLEA technology thus cause low product formation.
Formation of porous structure on CLEA particle (P-CLEA) is one of the strategy to improve
the accessibility of the substrate to the immobilized MAG1. Methods: The effect of porous
agent concentration and conditions for preparation of P-CLEA on the properties, stability and
reusability of MAG1 was examined. Results: The P-CLEA of MAG1 exhibited improved
catalytic efficiency and stabilities compared to free MAG1 and CLEA without addition of
porous agent against harsh condition of thermal and pH stress as well as high reusability.
Conclusion: The characteristics of P-CLEA of MAG1 highlight their great potential in
industrial application for oligosaccharides production.
