MICROORGANISMS IN THE ENVIRONMENT

I. OBJECTIVE:
To provide the techniques that will prevent organisms from contaminating
materials they work with such as media, sterile solutions and sterile
equipment.

II. EQUIPMENT / MATERIALS NEEDED:

3 petri dishes
1 water bath
1 incubator
1 stirring rod
1 beaker (150 mL)
1 graduated cylinder (50 mL)
1 colony counter
Nutrient agar
Distilled water

III. PROCEDURE:

1) Melt the nutrient agar and cool to 45 degrees Celcius.

2) Sterilized the media using the autoclave.

3) Pour 15-20 mL nutrient agar into each of the petri dishes.

4) Upon solidification of the medium, inoculate the petri dishes.

5) Incubate plates (inverted) at 35 deg. Celcius until the next laboratory


period.

6) Count the colonies in each petri dish using the colony counter.

7) Tabulate the result and describe the representative colony as to its


color, shape and elevation Preparation of agar solution (2-3%).
