OBJECTIVES

 Understand the basic principle of doing the aseptic technique.


 Be able to learn and master the basics of aseptic technique.

INTRODUCTION
Inside the laboratory, it is important that all tools and equipment are cleaned sterilized, and
sanitized, especially if we are going to handle microorganisms or microbes because they can
easily spread in your work area. One technique that you can do to maintain the cleanliness of
your environment is the aseptic technique.
The aseptic technique is a technique used to stop the spread of undesirable organisms in
space. Any technique used to sterilize and keep an area or object sterile, such as a laboratory, is
referred to as this. It also aids in preventing the spread of microbes found in a container. The
aseptic technique is a strategy used to lessen contamination from germs that incorporates target-
specific activities and processes under adequately controlled settings.
This technique prevents contamination of cultures while transferring them into a fresh media
such as a beaker, test tube, or petri dish as well as the laboratory and the tools and equipment
inside it. It also prevents possible health hazards that a person inside the laboratory can get if the
spread of microbes is not prevented.
MATERIAL

PROCEDURE
a. Wire Loop
 Open the Bunsen burner.
 Hold the wire loop and pass it at an angle from its base to the top and then top to
base in the blue flame of the burner until the entire length of the wire loop
becomes orange from the heat.
b. Transferring Culture Media to Beaker
 Open the Bunsen burner.
 Hold the wire loop and pass it at an angle from its base to the top and then top to
base in the blue flame of the burner until the entire length of the wire loop
becomes orange from the heat.
 Open the culture media container using your ring and pinky finger in an upward
motion.
 Let the wire loop cool down before inserting it in the culture media to avoid a
reaction. After cooling down the wire loop use it to get a culture in the culture
media container.
 Flame the opening of the culture media container at a 360 degrees angle and then
close it with the lid.
 Flame the opening of a separate beaker at a 360 degrees angle, place the culture
media in the wire loop onto the beaker, and then flame the opening of the beaker
at a 360 degrees angle and close it with the lid.
 Flame the used wire loop from base to top and then top to base on the blue flame
of the Bunsen burner.
c. Transferring Culture Media to Test tube
 Open the Bunsen burner.
 Hold the wire loop and pass it at an angle from its base to the top and then top to
base in the blue flame of the burner until the entire length of the wire loop
becomes orange from the heat.

motion.
 Let the wire loop cool down before inserting it in the culture media to avoid a
reaction. After cooling down the wire loop use it to get a culture in the culture
media container.

 Place the culture media in the wire loop in a test tube flamed at a 360 degrees
angle and then flame the opening of the test tube at a 360 degrees angle and close
it with the lid.
 Flame the used wire loop from base to top and then top to base on the blue flame
of the Bunsen burner.
d. Transferring Culture Media to Petri Dish
 Open the Bunsen burner.
 Hold the wire loop and pass it at an angle from its base to the top and then top to
base in the blue flame of the burner until the entire length of the wire loop
becomes orange from the heat.
 Open the culture media container using your ring and pinky finger in an upward
motion.
 Let the wire loop cool down before inserting it in the culture media to avoid a
reaction. After cooling down the wire loop use it to get a culture in the culture
media container.

 Slightly open the petri dish and flame the opened area, then put the culture media
in the wire loop inside the Petri dish in a zigzag pattern.
 Close the petri dish and flame the area where you insert the wire loop.
 Flame the used wire loop from base to top and then top to base on the blue flame
of the Bunsen burner.
OBSERVATION
In this laboratory activity, the aseptic technique was practiced. The aseptic technique is a
technique that is used to prevent the contamination of the culture media in a container as well as
the culture that will be transferred to another fresh medium for example in this laboratory
activity we used an Erlenmeyer flask, beaker, petri dish, and test tube. Furthermore, it also
prevents the contamination of the laboratory space as well as the tools and equipment in the

laboratory and avoids and prevents the possible health hazard that can be acquired by the
personnel inside the laboratory.
The aseptic technique is considered any kind of activity that helps avoid the spread of
microorganisms either pathogenic or not. In this laboratory activity, I observe that when going
inside the laboratory and handling microorganisms it is important to wear proper attire such as a
laboratory gown, hairnet or hair cap, face mask, gloves, and closed shoes to protect ourselves
from possible hazards inside the laboratory.
During the laboratory activity, we used heat to sterilize the tools and glass wares that we
used. Every time we open culture and transfer some to a new container using a wire loop, we
make sure that the opening of the containers as well as the wire loop was flamed in the hottest
flame of the Bunsen burner. This is done to avoid the possible spread of microorganisms.
Additionally cleaning and sanitizing the laboratory workspace as well as the tools and glass
wares before and after the experiment should always be practiced.
CONCLUSION
The aseptic approach was used in this scientific experiment. The aseptic technique is a
technique that is used to prevent the contamination of the culture media in a container as well as
the culture that will be transferred to another fresh medium. Additionally, it eliminates and
inhibits any potential health risks that may be acquired by the laboratory staff as well as the
contamination of the lab environment and its tools and equipment.
In this lab activity, we utilized an Erlenmeyer flask, beaker, petri dish, and test tube as a
new media where we transfer the culture. The aseptic technique is important in transferring
culture from one media to another to make sure that we prevent the spread of microorganisms. It
is important to treat all used microorganisms as pathogenic even if they are not to make sure that
we avoid touching them without proper personal protective equipment such as a laboratory
gown, glove, hairnet, face mask, and closed shoes.
Whenever you went inside a laboratory especially when you are dealing with
microorganisms always remember the phrase “do not touch or avoid touching” to ensure that you
can avoid holding the things inside the laboratory and spreading microorganisms from one object
to another as well as avoid possible health hazards that we can acquire inside the laboratory. That
is why we have aseptic techniques that we can practice ensuring that our safety will always come
first when using the laboratory.
