Molecular and chemotaxonomic comparative study (based on Jaccard


M6 and Kulczynski coefficients) of the genus Saccharomonospora

ABSTRACT RESULTS
A comparative study was conducted on 15 validated species/subspecies
belonging to the genus of Saccharomonospora, and the evolutionary distances
in relation to S. piscinae were deduced. Jaccard and Kulczynski similarity
indexes were used to construct the numerical chemotaxonomic dendogam Via
PAST tool, while MEGA11 tool was used to obtain a 16S rRNA phylogenic
dendogram. Surprisingly, major differences were observed between the two
statistical coefficients, with a complete accordance between Jaccard coefficient
and 16S rRNA Neighbor Joining method in MEGA11. Further comparison Fig 2. Saccharomonospora Sp. phylogenetic tree based on Jaccard index
taking into consideration the whole genome, and biosynthetic clusters, among
others, rather than the partial RNA 16S gene, is recommended. Even the
molecular tool is the systematics’ gold standard, there will be always a necessity
to a multifaceted approaches, and the chemotaxonomy is still a useful tool and
can go hand in hand with the molecular approaches.

INTRODUCTION

The genus Saccharomonospora was proposed by


Nonomura and Ohara in 1971 for monosporic
actinomycetes that contained meso-diaminopimelic acid,
arabinose, and galactose in the peptidoglycan [1].
Subsequently, additional chemical markers were found
to be characteristic [2].
Up to date, 19 species are designated to this genus, and
only 15 are validated . In the context of filling the
necessity to determine the taxonomic structure of this

compare it with the molecular 16S rRNA .
The order of similarity is the same only for the first 4 species
(PI>OC>XI>HA), and differs significantly for the rest, according to the Jaccard
and Kulczynski’s coefficients; probably due to lack of information about many

method in Mega11, (not in MEGA4) both give a remarkably identical between.
It is worthy to note that the similarity order for the molecular methods is

CONCLUSION
This comparative study on the genus of Saccharomonospora, shows that the
molecular taxonomic methods are the gold standard of systematic and
classification, due to the rapidity, efficiency, and ease of use; and the rapid
advances in the field of bioinformatics and technology make it even more
pertinent, and made the chemotaxonomy fell out of favor. even that, the
chemotaxonomy still a useful tool at least to delineate groups of related species
