What is plasmid?
• They are extra chromosomal DNA fragments present in the cell
• They are double stranded structures (exceptions are the linear plasmids
in bacteria Streptomyces spp and Borrelia spp)
• They can replicate independently
• The absence of a plasmid in the cell does not affect cell functioning,
but the presence of a plasmid in the cell is usually beneficial.

Small circular DNA molecules that replicate independently of the


host chromosomes
Plasmid Characteristics
• Ukuran plasmid bervariasi 2 kb - 200 kb.
• Plasmid berukuran kecil umumnya berjumlah sedikit, sedangkan
plasmid berukuran besar jumlahnya lebih banyak. 
• Kebanyakan plasmid mengandung gen TRA , yaitu gen yang penting
untuk mentransfer plasmid dari satu sel ke sel lain

Structure of Plasmids
Unsur penting pada plasmid:
• Origin of replication (OR) – Tempat dimana proses
replikasi dimulai
• Selectable marker site – Daerah dimana terdapat gen
yang resisten terhadap antibiotic (berguna saat akan
identifikasi dan seleksi bakteria yang mengandung
plasmid)
• Promoter region – Daerah tempat mengawali proses
transkripsi
• Primer binding site – urutan DNA pendek untai
tunggal yang bermanfaat dalam amplifikasi
(penggandaan) dan sekuensing DNA
• Multiple cloning sites/Restriction sites – Mengandung
berbagai sekuen tempat enzim restriksi terikat dan
memotong struktur rantai
Functions and Applications of Plasmids

• Dapat digunakan sebagai vektor untuk menyisipkan gen tertentu dan


mentransfernya ke sel lain. Sering digunakan untuk penghantaran
obat dan produksi hormon pada sel lain
• Mengandung gen yang resisten terhadap antibiotik
• Plasmid degradative dapat mendegradasi bahan kimia industri yang
membahayakan lingkungan
• Sumber penting untuk transfer gen secara horizontal (antar sel).

plasmids are purified from bacteria.
We are using plasmids that have been termed shuttle vectors,
because they can be propagated in either bacteria or yeast

non-pathogenic strain Saccharomyces cerevisiae


of Escherichia coli deletion strains

Plasmids are propagated in Plasmid-encoded genes


bacteria, which grow quickly are expressed in yeast,
and maintain multiple copies and phenotypes are
of the plasmids analyzed

Shuttle vectors have origins of replication and selectable markers


for propagation in both bacteria and yeast
