Dear Dr. Editorian:

I am pleased to submit the article entitled “A Hybrid Approach Based on R-CNN ResNet-50 Pre-trained and Image
Segmentation Algorithm to Detect Glaucoma Using Fundus Image” by Marlene Puchaicela, Ana Andrade, Dayanna
Baque, Carolina Cadena, Andrés Tirado, Lenin Ramírez, Diego Almeida-Galárraga, Jonathan Cruz and Fernando
Villalba-Meneses for consideration and publication in the Journal of Advances in Information Technology.

This work aims to use a hybrid method for glaucoma fundus image localization with pre-trained R-CNN ResNet-50
and cup-to-disk area segmentation. For which the ACRIMA and ORIGA databases were used that contain fundus
images. As a result, evaluating the ResNet50 shows average confidence of 0.879, which means that the
convolutional neural network is a reliable alternative for glaucoma detection. The final accuracy of 95% was
obtained. In addition, the validation loss plot indicates that for five epochs, ResNet50 achieved an appropriate fit
due to the application of the transfer learning concept.

We believe this manuscript is appropriate for publication in the Journal of Advances in Information Technology, as it
provides a new trustworthy approach for the safe diagnosis of glaucoma using fundus images. Here also, the cup-to-
disc ratio is calculated by using Gradient-color-based optic disc segmentation. The algorithm coincided with the
Resnet50 results in 80% of cases, having an average confidence score of 0.84. Therefore, considering both results, it
can be determined whether there is glaucoma or not.

This manuscript has not been published and is not under consideration for publication elsewhere. We have no
conflicts of interest to disclose.

In addition, we declare that all authors have made substantial contributions to all of the following: (1) conception
and design of the study, (2) drafting of the article or critical revision for important intellectual content, (3) final
approval of the version to be submitted.

Thank you for your consideration!

Sincerely,

Luis Zhinin-Vera
