A microscope is a tool which can help you see tiny objects and living organisms. It comprises of three major
parts based on their functions: magnifying, illuminating and mechanical parts.

Lesson 2: Focusing Specimen using the Compound Microscope
Since discussions on magnification and how the microscope enlarges a specimen was done, it is time to learn
the steps in focusing specimen using the Compound Microscope so that when you will be in an actual laboratory,
you’d be able to familiarize yourself on how to manipulate it. Familiarity of its parts and function is necessary in order
for you, learner, to utilize the microscope’s efficiency to the fullest. So, the big question for this module is, “How are
you going to focus specimen using a compound microscope? It’s easy. Just follow the steps presented below.
Step 1: Determine the specimen you are going to focus on.
In the class, your teacher would provide you what specimen to focus on depending on the topic being discussed. Say,
the topic is all about animal cell, and then the teacher may opt to require you to scrape a tiny bit of sample from your
cheek using a clean toothpick or if its plant, then your teacher may tell you to bring an onion skin.
Step 2: Preparing a Wet Mount
A SPECIMEN is a sample part of any material such as plant, animal or even paper and mineral for examination
under the microscope.
Step 3: Viewing a Specimen
FIELD OF VIEW is the bright circle of light under the microscope. Remember not to use direct sunlight as a source
to view a specimen because it can permanently damage the retina of the eyes.

Using the Microscope Properly


1.Carry the microscope with one hand on the arm and one hand on the base. Carry it close to your body.
2.Place the microscope on a table with the arm toward you.
3.Do not touch the lenses with your fingers. Use lens paper to clean the lenses.
4.Rotate the course adjustment knob to raise the body tube.
5.Revolve the nosepiece until the low power objectives lens clicks into place.
6.Adjust the diaphragm. Look through the eyepiece and adjust the mirror until you see a bright circle of light.
7.Place the slide over the opening on the stage with the specimen directly over the light. Use the stage clips to hold the
slide in place.
8.Carefully rotate the coarse adjustment knob to lower the body tube until the low power objectives (LPO) lens nearly
touches the slide. Look at the stage from the side of the microscope while doing this.
9.With both of your eyes open, look through the eyepiece and slowly rotate the coarse adjustment knob until the
specimen comes into focus.
10.To switch to the high-power objectives (HPO) lens, revolve the nosepiece carefully until the high-power objective
lens clicks into place. Look at the stage from the side of the microscope while doing this to make sure that the lens
does not hit the slide.
11.Look through the eyepiece and turn the fine adjustment knob very slowly until the specimen comes into focus.

Levels of Biological Organization
Organisms are categorized according to the number of cells in the body, either unicellular or multicellular. A
unicellular organism depends upon just one cell for all of its functions while a multicellular organism has cells specialized
to perform different functions that collectively support the organism. As they grow and develop, they become highly
organized and specialized.
The different levels of biological organization are described and arranged below from the highest to the lowest
or in a descending order.
