SpecPlex® Gloss endows skin beauty not only from outside in but also from inside out !

SpecPlex® Gloss is a water-soluble compound product with six skin brightening/lightening


ingredients. It can smooth skin through remove dead horn cells reduce the production of
melanin, removing the formed pigment and reducing pigmentation; it contains the
ingredients strenthening extracellular matrix (ECM) though promoting the production of
collagens and glycosaminoglycans (GAGs); Thus, it has the function of plumping/enriching
skin texture and lightening skin tone, delaying aging, moisturizing and nourishing skin, etc.

Conclusions:
• According to SN/T 2328-2009 and MTT method, the IC50 of SpecPlex ® Gloss is 2.6% on
the NIH/3T3 cell line.SpecPlex ® Gloss had no toxicity on NIH/3T3 cell line when the
concentration was not more than 1.56%.
• If converting above value into the acute oral toxicity LD50 of mice theoretically, then the
acute oral toxicity LD50 of SpecPlex ® Gloss was 4645mg/kg, so SpecPlex ® Gloss was
classified as grade 5 (non-toxic) according to GHS acute toxicity classification standard.

According to the experimental results,SpecPlex® Gloss at the above different concentrations


did not significantly increase the release rates of allergy marker β-hexosaminidase in mast
cells, contrary to the positive control (Compound 48/80,allergen).
Therefore, SpecPlex® Gloss has no allergenicity under testing concentrations (up to 20%).

• Components of the ECM are produced intracellularly by resident cells and


secreted into the ECM via exocytosis.
• Once secreted, they then aggregate with the existing matrix. The ECM is
composed of an interlocking mesh of fibrous proteins and glycosaminoglycans
(GAGs).
• Collagens are the most abundant protein in the ECM. In fact, collagen is the
most abundant protein in the human body and accounts for 90% of bone matrix
protein content.
• Collagens are present in the ECM as fibrillar proteins and give structural support
to resident cells.
• Elastins, in contrast to collagens, give elasticity to tissues, allowing them to
stretch when needed and then return to their original state. This is useful in skin,
and these tissues contain high amounts of elastins.
• Elastins are synthesized by fibroblasts and smooth muscle cells.

Promoting ECM-related gene expression-SpecPlex™ Gloss

According to the results of PCR test, compared with blank control (BC) group,
SpecPlexTM Gloss can obviously promote the gene content of collagen I,
collagen VII and Laminin, i.e. the expression of ECM-related genes, to
enhance the ECM function.

According to the results of cell tyrosinase activity test and cell dopa staining
experiment, SpecPlex® Gloss can inhibite the activity of tyrosinase in melanocytes.
The inhibition rate is 58.5% at 0.1% and IC50 is 633μg/mL(≈0.0633%).
In addition, it also has a significant inhibitory effect on the production of melanin in
B16F10 melanocytes, the inhibitory rate reached 41.4% when the concentration was
