Testing and Quality control. LABORATORY ACTIVITY FOR SET A WEEK 10-17 Introduction Germination percentage is an estimate of the viability of a population of seeds. The equation to calculate germination percentage is: GP = seeds germinated/total seeds x 100 . The germination rate provides an measure of the time course of seed germination. Plant hormones, mainly abscisic acid (ABA) and gibberellin (GA), are the major endogenous factors that act antagonistically in the control of seed dormancy and germination; ABA positively regulates the induction and maintenance of dormancy, while GA enhances germination. Gibberellins control different stages of plant development, including seed germination, seedling growth, stem elongation, root extension, leaf size and shape, flower and fruit development, pollination (Garcia-Martinez et al., 1997; Yamaguchi, 2008; Heden and Thomas 2012) Gibberellin is the other major phytohormone with an important role in the regulation of seed dormancy and germination (Finch-Savage and Leubner-Metzger, 2006).

Job sheet no. 4 Title: RICE SEED GERMINATION TESTING Performance 1. Prepare a demonstration of Seed Germination using Gibberellic Acid Objectives (GAA3) 2. Measures seed germination and rate Materials GAA 3, 3 L water, 16 polypropylene box, 2 Filter paper or Tissue paper Facilities/Area Home/ School Step and Procedure 1. Prepare 500 grams filled rice seeds (good seeds) and at least 2 liters of clean water. 2. Clean the seeds with tap water thoroughly in two (2) times if necessary to avoid contamination. Seeds that are floated in the water should be removed. 3. Air dry the seeds until it is good to transfer into the polypropylene box. Count 50 seeds per box. 4. Submerge the seeds in 500 ml of water at various GAA 3 Concentration at a different length of time. See table 1: Various Concentrations at different length of time. 5. Moisten the filter paper or Tissue paper with pure water only. Spread the seeds on a layer of filter paper or tissue paper in the polypropylene box. 6. Observe the process of germination for the Monocotyledon seeds. 7. Record the date of planting, breaking the seedcoat and from the emergence of the radicle and plumule. 8. Determine the germination rate of each treatment as submerge to different length of time. 9. Make your activity report using the following. - Observation - Findings and Result - Conclusion - Documentation 10. Complete the data needed in the table 2 11.
