1. INTRODUCTION

Thank you for choosing Hospitex Diagnostics SCREEN MASTER equipment.


This new instrument was designed to have powerful features in a compact unit for biochemistry,
enzymes, immunoturbidimetrics and drugs tests.
These characteristics make the Screen Master a perfect laboratory instrument.

2. DESCRIPTION OF THE ANALYSER

The Screen master technical features are:


- Integrated pump and flow cell.
- Internal pre-adjusted peltier element at 25°C, 30°C, 37°C.
- 37°C incubation block with eight positions.
- Easy to use with step by step procedures.
- MORE THAN 200 PROGRAMMABLE TESTS.
- Multilingual capability.

It performs:
- End Point
- Kinetic
- Fixed Time
- Multistandard assays

Results are shown on graphic display and printed on paper by in-built printer.

2.1 Technical specifications

Light source Halogen lamp –12V, 20W

Wavelength 340nm-700nm.
Wavelength selection Automatic via 8-position filter wheel;
6 standard interference filters: 340nm, 405nm, 505nm, 546nm, 578nm, 630nm.
Two free positions for optional filters.
Photometric Range: 0-2.5 O.D.
Flow cell system 32L micro-flow cell with 10mm light path, interchangeable
with 80L micro-flow cell or standard cuvettes (macro or semi-
micro, disposable or special optical glass).
Reading time 1s-300s.

Incubation time 10s-600s.

Temperature control Peltier elements, 25°C, 30°C and 37°C.

Reaction volume 500 L per test.

3. MEASUREMENT PRINCIPLES

The concentration of a solute that absorbs light at a determinate wavelength can be determined with
a spectrophotometer. The fraction of light absorbed is proportional to the concentration of the
absorbing solute (Beer’s low):

(1) I / I0 = e-kc

Where:
I0= Incident light intensity before going through to sample.
I= Transmitted light intensity through to sample.
K= Constant that depends upon the length of light path, nature of the substance and the
light wavelength used.
C= Substance concentration in solution.

Equation (1) can be changed into:

log( I0/I) = kc

The quantity log(I0/I) is called Optical Density (OD) or Absorbance (ABS) and is proportional to
concentration.
A spectrophotometer measures the intensity of a known wavelength (measured in nanometers, nm)
of light passed through the sample: OD = log( I0/I)

By using specific reagents in solution for the substance that we want to measure, it is possible to
estimate the constant ( k) for a fixed wavelength, in this way transmitted light intensity becomes
concentration function.

The instrument has a reading point, kept at 25-30-37°C temperature, and in order to perform a
measure it will be necessary to have:

 Analysis with flow cell using a peristaltic pump.


 Keeping manual cuvette at 37°C temperature using external incubator.

4. INSTALLATION

4.1 Packing contents

Inside the package you will find a starting kit composed as listed here below, which will allow you
to correctly install the instrument.
If any of the following items is missing or damaged, please contact us for help.
- Power supply cable.
- two fuses.
- Waste bottle and silicone tube.
- One hundred macro cuvette.
- Two paper rolls.
- Instrument cover lining.
- Two cuvette adapters:

4.2 Keyboard

Correct use of the keyboard is very important to perform the analyses and to make operations fast
and accurate.

KEY FUNCTIONS
- To insert ‘0’ and to set parameters to zero in ABS mode.
0

from 1 to 3 - To insert numbers and to choose between the options.

from 4 to 9 - To insert numbers.

- Print of Kinetic and Fixed Time Dynamics in the ‘Sample


? Measurement Screen’.

- To move on main screen.


Right and left arrows - To move on values.
- To move between several menu options.
- To move on main screen.
- To move between several menu options.
Up and Down arrows - To scroll up and down the method list in ‘EXE’, ‘PRG’ and
‘PRT’.

OK - To confirm your choices.

Del - To exit from the current Menu.

4.3 Rear panel setting

Located in the rear panel (from left to the right as shown in the picture) you will see:
- Fan for internal cooling (another similar is located under the instrument).
- The power supply switch.
- The polysnap (for the power supply).

Before switching on the instrument, remember to connect the plastic outlet connector
(blue) to a waste tank by means of a silicone tube.

4.4 Location of the instrument

The instrument should be located in a clean environment, placed on a stable surface, away from dir-
ect sunlight which could affect the operating temperature and the quantity of light measured by the
instrument.
The following points should be taken into consideration.
- Ensure that it is on one level.
- Avoid positions subject to jerks or vibrations.
- Make sure that the instrument is not placed close to air conditioning or heat sources.
- For the long life of the instrument these temperature conditions should be followed.
5°C - 50°C for instrument storage.
15°C - 30°C for instrument use.

4.5 Power supply connection

- Please check the setting of power supply switch according to your country’s electrical net-
work.
- Connect the power plug to a grounded AC wall outlet, preferably not shared with other
electric appliances and with low fluctuation of the line voltage compared to the standard
voltage specified (10-15%).

- Keep the instrument away from other appliances that generate high frequency electrical
noises (e.g. radiological instruments).
- Before connecting the power cord, check that the AC power supply is of the same value as
the one stated on the label stuck on the instrument.

Caution

Do not connect to an AC power supply different to the value indicated on the label.
- Before connecting the power and finishing the installation section, make sure that the
instrument is switched off (check the polysnap located on the rear part of the instrument).
- Make sure that your AC main line has an efficient ground line. A bad ground line
connection may compromise analysis results and damage the instrument.
- After switching on the instrument, do not go near the fan with liquids or micro solid
substances.
- Pay attention not to spill liquids and micro solid substances on the surface around the
instrument.
- Keep the instrument away from young children.

If the above procedures are carefully followed up, it will be possible to SWITCH ON the
instrument by using the switch located on the polysnap block.

4.6 Reset and switching off

To reset the instrument:


 turn off the switch.
 wait for 5 seconds.
 turn on the switch.

Remember that the instrument must only be reset if the software does not work.

4.7 Measurement lock settings

Open the measurement lock cover. It is placed on the left part on the top of the instrument.
You will find:
- a peristaltic pump that allows the solution to enter the instrument for measurement.
- two hollows: one, below, for reading and one for the flow cell service place.
- an incubator which keeps the cuvette at the temperature of approximately 37°C.
- a inlet pipe.

To set the pipe in reading mode, rotate it by about 90 degrees clockwise as displayed in
the figure below, until the tube fits perfectly onto the hole in front of the instrument.

4.8 Tube setting


Pump type A: Make sure that the transparent tube (the one that connects the steel inlet pipe with the
internal hydraulic circuit) is not twisted or squashed.
Place the red tube around the peristaltic pump and make sure that the white plastic connections are
set as shown in the figure.

Pump type B (from serial number 1131559): Make sure that the transparent tube (the one that
connects the steel inlet pipe with the internal hydraulic circuit) is not twisted or squashed.

Under the measurement block there is a lever (the PUSH button). Press the lever for no more than
half a second to pump samples into the flow cell.

4.9 Printer settings

To insert the paper roll, proceed as follows:


- Switch on the instrument.
- Remove the printer cover and locate the green lever.
- Raise the little green lever located on the right side of the printer.
- Take a new paper roll and pass the edge of the paper under the black rubber cylinder. The
cylinder will start its rotation so that the paper will set itself automatically.
- Pull the lever down again, pass the edge of the paper through the aperture and place the
printer cover in its right position.

4.10 Incubator

The incubator temperature is brought up to 37 °C by the software and it will be kept until the
instrument is turned off.
This function is very important to heat macro cuvettes for performing the analysis at the correct
temperature.

4.11 Optical group

 Light pulse is performed by an halogen lamp, that covers wavelength range (340630 nm)
with its emission.
 Wavelength is selected using optical filters.
 Iigth beam is focus on sample by a proper lens system.
 Receiver is a photobattery that changes light signal in tension signal.

4.12 Amplification

Amplification chain consists in these parts :

 Preamplification is performed in a preamp board on optical group, and it has a 12V dinamics.
 Differential on main board erases on line noises (giving no further amplification).
 Amplification on main board supplies gain over a 5V dinamics.
 A/D conveter works on 5V dinamics.

5 SERVICE FUNCTIONS

In order to enter service function :

 Switch on instrument and the chess board on display will appear:

 at the same time press several times the following keys depending on which menu you want to
enter:

In this screen you can monitor (after having filled water) tension coming from optical group (OPT
TENS) and tension coming in A/D converter (A/D TENS) , using a particular filter (FILTER)
WATCH TENSION SCREEN is very useful to verify lamp calibration , filter / lamp / lens /
photodiode / preamp board quality , position sensor working.
Moreover in this screen you can test correct pump motion.

As you enter this menu:

 you have to select one filter using 1…6 key

1…. 340nm
2…. 405nm
3…. 505nm
4…. 546nm
5…. 578nm
6…. 630nm

 Press PUSH BUTTON to fill water.

 Look at OPT TENS and A/D TENS values.

 Press any other 1…6 key if you want to change filter, press PUSH BUTTON if you want to fill
again water.

If you have a correct calibrated optical group, when you use the flow cell, the OPT TENS values
are between 250 mV (for 340 nm) and about 3.5 V (for 630 nm) for all filters.

If you have a correct working of digital potentiometer , you will see A/D TENS value in this range :

 3.5V +/- 10% for OPT TENS < 3.5 V

 OPT TENS + /- 5% for OPT TENS > 3.5 V

Function FILTER HOME is useful to monitor position sensor activity.

Each time you set a filter position program is showing you number of step sensor has recognized
wheel crack :

If recovery position system is working well:

 for every filter this number must be similar.


 this number has to be <=4.
 this number has to be >0.

This screen allows you monitoring temperature gradients of incubator and cuvette according to
temperature displayed in user program and digital level acquired by microcontroller (both current
and target).

As you enter in this menu you will see MEASURE POINT case (cuvette):

 you have to select one of three temperature using 1...3 key:

1 …….. target temperature 25°C, target digital level 132.

2 …….. target temperature 30°C, target digital level 158.

3…….. target temperature 37°C, target digital level 190.

 you will see increasing / decreasing current temperature and digital level towards targets.

 when program considered reached target values (with a consequent decrease of temperature
gradient op to zero) software is showing you current values in reverse mode.

Use DEL to exit this screen

Use NUMBER 6 to pass INCUBATOR case.

This case is similar to other one:


target temperature 37°C, target digital level 190.

Use DEL to exit this screen

WATCH TEMPERATURE SCREENS are important for:

 test good working of temperature acquirement circuit.

 calibrate temperature control.

There is one primary circuit :

0 – 220 V ( +/- 10% )

There are 2 secondary circuits :

1) 12 – 0 – 12 V for 0. 2 A

2) 7 – 0 – 7 V for 5 A

Trasformer type is toroidal with temperature sureness switch inside .

6.2 Hydraulics (for pump type A)

1) Silicone 2X4, length cm 4.

6.3 Control and calibration

1) Connect JP 18 . NOTE JP19 and JP 20 disconnected.


2) Disconnect J1 - J2 - J19 - J20 - J12 - J18 - J15.
3) Verify transformer. With disconnected J3 verify tensions on female connector coming from
trasformer according this table :

1 = 0 V
2 = 7 Vac
3 = 14 Vac
4 = null
5 = null
6 = 12 Vac
7 = 0 Vac
8 = 12 Vac

4) Connect J3 again e verify following test point:

Tp7 = GND
Tp6 = 12 Vcc
Tp8 = - 12 Vcc
Tp12 = 12 Vcc
Tp14 = 5 Vcc

5) Regulate following tensions:

Lamp tension

Connect Voltmeter Tp10 (GND) and Tp1 and regulate 12 Vcc by RV1
Connect lamp (J1) and control tension again.

Printer tension
Disconnect J15
Connect Voltmeter Tp10 (GND) and Tp9 and regulate 5,6 Vcc by RV5
Connect power printer J15 and control tension again.

Stand by motors tension

Connect Voltmeter Tp10 (GND) and Tp2 and regulate 7.5 Vcc by RV2.

6) Temperature calibration.

Zero offset
-Connect Jp24.
-Connect Voltmeter Tp10(GND) and Tp15 and regulate least tension by RV8.
-Disconnect Jp24.

-Connect Jp25.
-Connect Voltmeter Tp10(GND) and Tp17 and regulate least tension by RV10.
-Disconnect Jp25.

Temperartures calibration

Cuvette
-Connect J19.
-Switch on instrument and press 4 times 2 key during chess board on display.
-On menu tes select watch temperature.
-Put termometer on test cuvette test and press 3 (37°), trim digital level 190 by RV7 in
order to have 37.5°C on termometer. When balance is reached the digital level will fluctuate
around 190.
- Switch off instrument.

Incubator
-Connect J20.
-Switch on instrument and press 4 times 2 key during chess board on display.
-On menu test select watch temperature.
-Put termometer on incubator, press OK and 3 (37°), trim digital level 190 by RV9 in
order to have 38.3°C on termometer. When balance is reached the digital level will fluctuate
around 190.

7) Analog calibration.

Offset
- Disconnect J2.
- Connect JP5.
- Connect Voltmeter between TP5 and pin1 of JP6 (near TP4).
- Make zero tension by RV4.
- Disconnect JP5.
- Connect JP6 by a grabbe cable.
- Connect voltmeter between other head of grabber cable and TP4.
- Make zero tension by RV3.
- Disconnect JP6.

Preamplification calibration

- Connect Voltmeter between pin 1 and pin 2 in J2 (extract contacts from connetctor and
connect them at voltmeter).
-Switch on instrument and press 4 times 5 key during chess board on display
-On menu test select watch tension.

- Select 630 nm filter and regulate tension 3,5 Volt by trimmer on preamp board ( using
full of distilled water cuvette ).
- On this table sign up tensions of each filter for at least 3 times.

Various repeatitions must be similar.

N.B.
For each filter you have to see a value not greater than 3.5 V.
If you find a tension greater than 3.5 V, control if a correct diafram is present, if not, change it and
calibrate preamplification gain.

7. TROUBLESHOOTING

1. PROBLEM : pump doesn’t work.

POSSIBLE CAUSES :

 Disconnected PUSH (J10).


 Connector has no correct contact on board side (J5) or on motor side.
 Motor is broken.
 Regulator U20 on board is broken.
 Driver U23 on main board is broken.
 Mechanical problem on motor shaft.
 Too stretched pump red tube.

2. PROBLEM : incubator doesn’t work.

POSSIBLE CAUSES :

 Power connector has no correct contact on board side( J17 ).


 Sensor connector has no correct contact on board side( J20 ).
 Temperature sensor broken.
 Problems on incubator power supply (transformes , capacitor C43).
 Thermic resistors disconnect or connect in a wrong way.

Right way : 10

 OPA U30 broken on board or some other broken components.


 Not enough conductive stuff on sensor or on thermic resisto.r
 Calibration problem.

3. PROBLEM : instrument is switched on but it doen’t display anything.

POSSIBLE CAUSES :

 Some false contacts on keyboard – display joint board.


 Broken display.
 Broken chip controller on main board.

4. PROBLEM : program seems to not read keyboard.

POSSIBLE CAUSES :

 Keyboard disconnected on keyboard – display joint board.


 Some problem on RR1 on main board.
 Broken keyboard.

5. PROBLEM : printer doesn’t work.

POSSIBLE CAUSES :

 Disconnected J16 (data) or J21 (power).


 Broken regulator U19.
 Broken pinter.

6. PROBLEM : instrument doesn’t measure.

Cases :

1. Wrong ABS values inside significative range (for example little ripples in ABS values):

POSSIBLE CAUSES :

 Micro / macro bubbles on cuvette caused by dirty cuvette or old fluidic.


 No earth connection on board.
 Problem on power supply of lamp (look at transformer or power capacity (C43) connections).
 Lamp broken : change with a new lamp following the procedure illustrated in Appendix 1.

2. Out of significative range (from 3.00 to 5.00) ABS values for every reading session:

 Not well fitted cuvette.


 Problem on lamp.
 Problem on filter wheel motor.
 Problem on preamplification board.
 Disconnected J2.
 Problem on main board (broken U14,U15,U16).
 Preamp board needs to be calibrated again.

3. Problems on Kinethics analysis:

 Not calibrated cuvette temperature.


 Broken peltier cell.
 Broken temperature sensor.
 Problem on Peltier power supply on board.
 No earth connection on board.

7. PROBLEM : PC serial port doesn’t work.

POSSIBLE CAUSES :

 Problem on serial cable and connections.


 Problem on serial section on board (U25 , C60 , C61 , C62 , C63).

Appendix 1: Changing lamp

This is the procedure to change lamp :

 Open case of the instrument, pulling out four screws down the equipment and three ones on the
back side .
 Pull out two screws at right and left sides of cuvette site, and move away pump support (the one
with pump rotor).
 Move away the little tongue that fix the lamp, by pulling out the only screw .
 Extract old lamp, and set carefully the new one, bringing it up to the bottom of the site (pay
attention to not touch lamp glass with finger skin).
 Set up again the little tongue, and check the top of the lamp being inside the little hole.
 Set up again pump support and equipment case.

Appendix 2: The flow cell does not aspire correctly:

The red tube of peristaltic pump ( pump type A) does not aspire well.

The white tube of peristaltic pump (pump type B) does not aspire well. To check this operate as
follow:

 Switch on the instrument and when chessboard appair press number 7 several times
 The following screen will appair:
5100 1150

 5100 is the default pump step number for pump type A


 1150 is the default pump step number for pump type B
 Prepare 500 l of water in a cuvette set the aspiration tip in an bottom angle of
manual cuvette and press (.) key to aspire
 If the pump works correctly, the water should be aspired completely from the
manual cuvette, and also the aspiration tip should be empty from water:
 Repeat operation several times

If water stops at beginning of aspiration tip or stops to near the cuvette the red tube has a problem
and you have to adjust aspiration by changing the aspiration pump steps.

proceed as follow:

 Press up arrow keys to increase aspiration pump steps (the liquid will stop nearer to cuvette)
 Press down arrow keys to decrese aspiration pump steps(the liquid will stop further to
cuvette)
 Prepare 500 l of water in a cuvette set the aspiration tip in an bottom angle of manual
cuvette and press (.) key to aspire
 If the pump works correctly, the water should be aspired completely from the manual
cuvette, and also the aspiration tip should be empty from water:
 Keep adjusting until the aspiration is correct

Press right arrow key to return at default pump step number

Appendix 3: Red tube changing

1. Disconnect the red tube from peristaltic pump by pulling one of the white holder.
2. Free the red tube from holders by pulling the two white pieces apart:

3. Measure the red tube with a ruler, the length must be 10 cm, if not cut the exceeding of the
tube with sharp cutter and set the tube back. Then check aspiration as appendix 2.

Sometimes red tubs go longer than 10cm especially in hot wheather
