Preface
Thank you for purchasing BK-EL10C Elisa Microplate Reader.
Intended use
The instrument is designed according to the vertical 8-channel optical path, which can
perform single and dual wavelength measurements, and provides multiple modes such
as absorbance test, qualitative test, quantitative test, and inhibition ratio test. This
instrument is widely used in scientific research laboratories, quality inspection bureaus,
agricultural and animal husbandry, feed enterprises, food enterprises and other testing
fields.
Using object
This manual is intended for the clinical laboratory technician who operates the
instrument. Before operation, please read the contents of this manual carefully and use
the product correctly. Please keep this manual properly for easy viewing at any time. If
you do not comply with the precautions described in this manual, the instrument will
not be subject to warranty.

Safety Regulation
In order to use this instrument safely, please read the following safety regulations
carefully. Any operation that violates the following may cause personal injury or
equipment failure.

Safety Precautions
Prevent electric shock
To prevent electric shock, please observe the following precautions.
Warning:
 Do not plug or unplug the power supply with wet hands. It may
cause electric shock.
 Do not use damaged wires and connecting cables, as electric shock
or fire may occur.
 Do not use wires and cables other than the design requirements,
and do not use power cords or sockets that are not certified by 3C,
which may cause electric shock or fire.
 When replacing fuses, bulbs, filters or opening the housing, please
turn off the power and unplug the power plug.
 Do not put reagents, water, etc. on the surface of the instrument to
avoid the leakage of liquid into the instrument and damage to the
instrument.

Prevent biological infection


In order to effectively protect against biological hazards, please observe the following
precautions.
Biological infection risk:
 All biological samples, quality controls and standards should be
considered potentially infectious. Do not directly touch samples,
quality control products, calibrators, mixtures and waste liquids
with your hands. Please be sure to wear gloves and overalls during
operation to prevent infection, and wear protective glasses when
necessary. Strict compliance with GLP (Good Laboratory Practice)
regulations can ensure personal safety.
 If the sample accidentally touches the skin, please handle it
immediately in accordance with the user's working standards and
consult a doctor.
 When cleaning the instrument, please wear gloves. Do not use
chemical reagents such as turpentine oil or benzene to clean
external stains, because it may cause changes in color and shape.
Use a soft or damp cloth to scrub. For serious stains, use detergent
or 75 % Alcohol clean. For spilled samples at risk of infection,
clean them with 75% alcohol.

Prevent punching
To prevent personal injury caused by moving parts when the instrument is working, please
observe the following precautions.
Warning:
 When the instrument is working, the access switch is potentially
dangerous. The operator must be professionally trained and must
follow the instructions to ensure that it operates in a safe area.
 After the instrument is powered on, do not put your fingers or hands
into the open parts.
 After the access compartment is opened, please place or remove the
enzyme label plate in time, and do not leave your fingers in the
access compartment for a long time to prevent the instrument from
being accidentally touched and closed.

Emergency handling
When the following unexpected situations occur, please observe the following precautions
to prevent secondary damage to the instrument.
Warning:
 When there is abnormal movement or sound, the operation should
be stopped immediately and the power should be turned off.
 Although the housing of the instrument is made of metal and
flame-retardant ABS material, when smelling scorching odor or
abnormal sound is heard, turn off the power immediately, unplug
the plug, and be prepared for fire extinguishing.
 If screws or metal objects falling into the instrument, stop the
operation and unplug the power plug immediately. Please ask
professional maintenance personnel to take out the metal objects
before starting the operation, otherwise it may cause instrument
failure.

Protection against chemical hazards


To prevent personal injury caused by chemical dangerous goods, please observe the
following precautions.
Warning:
Some reagents (including but not limited to washing buffer, developer,
stop solution, etc.) may cause toxicity, irritation, burns or carcinogenesis.
Please use reagents carefully and avoid contact with skin and mucous
membranes. If you accidentally touch your hands or clothes, wipe them
immediately and rinse them with soap and water. If it gets into the eyes
accidentally, rinse immediately with plenty of water and consult an
ophthalmologist.

Waste treatment
To prevent waste from causing environmental pollution and personal injury, please observe
the following precautions when disposing the waste.
Biological infection risk:
 Pipette tips, reagent bottles, test strips and specimen containers should
be properly collected and autoclaved at 121°C for no less than 2 hours
or treated with 10% sodium hypochlorite for 30 minutes before
processing. Note: Do not autoclave solutions containing sodium
hypochlorite.
 Some substances in reagents, quality control products, calibrators,
washing buffer, cleaning fluids, and waste fluids are subject to
pollution regulations and emission standards. Please comply with local
emission standards and consult the relevant reagent manufacturer or
distributor.
 When disposing of waste liquid, be sure to wear gloves and overalls to
prevent infection, and wear protective glasses if necessary.

Prevent fire and explosion


To prevent fire and explosion, please observe the following precautions.
Warning:
Alcohol is flammable, please be careful when using it.

Processing instrument
In order to reduce or eliminate the risks involved in the process of stopping use (such as
maintenance or transportation) or scrapping of the equipment, please handle the equipment
according to the following precautions.
Warning:
 All equipment parts in contact with biological samples, patient
samples, positive quality control samples or hazardous materials
should be considered as potentially infectious areas.
 Before removing the instrument from the laboratory or before the
instrument is repaired, all surfaces and microplate carriers must be
disinfected, and a safety certificate must be issued. The disinfection
procedures and disinfectants should comply with relevant national
laws and regulations. If no safety certificate is provided, the customer
service center will refuse to accept the device.
 Some substances in discarded instruments are subject to pollution
regulations. Please follow local waste disposal standards when
disposing of discarded instruments.

Chapter 1 Main Introduction


Elisa Microplate Reader (hereinafter referred to as Elisa Microplate Reader or this
device, this instrument), which is designed according to the principle of enzyme-linked
immunoassay technology, adopts the design of vertical 8-channel optical path, and can
be used for single and dual wavelength measurement, providing absorbance test,
qualitative test, quantitative test, inhibition ratio test and other modes. The instrument
adopts 10.1-inch industrial-grade color LCD display, touch screen operation, and
external thermal printer. The test results can be displayed, stored and printed in the form
of a whole microplate. Elisa Microplate Reader can be configured with computer-side
Elisa Microplate Reader analysis software.
This instrument is widely used in scientific research laboratories, quality inspection
bureaus, agricultural and animal husbandry, feed enterprises, food enterprises and other
testing fields.
1.1 Configuration and Structure
Elisa Microplate Reader is shown as below, with 10.1-inch LCD screen, touch screen
operation, optional thermal printer. that can work independently, or it can be equipped
with a computer to form a workstation mode.

1.1.1 General Accessories


The following table shows the general accessories and tools equipped with Elisa
microplate reader before leaving the factory. Please refer to the accessories list in the
box for the specific accessories and quantity.

Note:
TP for technical parameters, DP for Device parameters, EP for Environmental parameters.
When the storage and transportation conditions (temperature, humidity) of the instrument exceed the
range of normal working conditions, the instrument should be placed under normal working
conditions for more than 24 hours before it is powered on. Do not power on immediately.
1.3 Filters
The standard filter wheel of this instrument has 10 filter configuration positions, and the
factory default configuration has 4 narrow-band interference filters with fixed
wavelengths (405, 450, 492, 630nm). Other locations support the selection of more than
20 wavelengths. For specific optional wavelengths, please contact your local agent or
our pre-sales engineer.
The filters of the standard filter wheel are installed as follows:

When the wavelength is selected for measurement, the specific filter is brought under
the beam by rotating the filter wheel to the corresponding position.
Notice:
 If the customer chooses the wavelength of the filter before leaving
the factory, the engineer will define and initialize the position of
the filter according to the customer's needs before the instrument
leaves the factory. If the customer adds or replaces the filter
wavelength after the instrument leaves the factory, it is necessary to
install the filter assembly according to the engineer's instructions,
and redefine and initialize the filter wavelength in the software. For
specific operations, see section 5.5 Adding or replacing filters.
 Do not disassemble or assemble the filter privately to avoid
confusion about the position and wavelength of the filter, resulting
in inaccurate measurement data.

1.4 Microplate
All 96-well microplates with transparent bottom (including microwell strips, flat, C, U
and V) that meet the following standards can be used in this instrument.

The installation operation of the microplate is as follows:
 Click [OP/CL] in any measurement interface to open the access door, and the
microplate carrier will automatically pop up.
 Place or remove the microplate only when the microplate carrier is fully opened (as
shown in Figure 1-3) and the motor is not moving.
 Align the hole A1 of the microplate with the hole A1 of the microplate carrier as
shown in the figure, put the microplate in the carrier steadily, and use the two spring
leaves shown in Figure 1-3 to clamp the microplate live.

Warnning:
 Do not use external force to open the door or push or pull the
microplate carrier.
 When handling the microplate, be sure to wear disposable gloves,
and if necessary, wear protective clothing and goggles.
 Since scratches and stains can interfere with reading, do not touch
the bottom of the well during the handling of the microplate.
 Before placing the microplate, make sure that the bottom of the
plate is dry and there are no bubbles in the wells.
 Before placing the microplate, remove the plate cover.

1.5 Function
1.5.1 Measurement Mode
The instrument has the following three measurement modes:
STD The light source is in a dormant state before the test. When the test
starts, the light source is turned on, and the light source is turned off
immediately after the test.
ACC1 The light source is automatically turned on before the test, the test
starts after 60 seconds of preheating, and the light source is turned
off after the test is over.
ACC2 After the instrument is powered on, the light source is turned on, and
the default is to continue to warm up. The test can be performed at
any time without waiting time.
1.5.2 Shake Mode

Notice:
 When the instrument is in ACC2 mode, the main interface will
continue to flash and display a small light bulb (see 4.2.1.1 for
operation details) to remind the user that the halogen lamp is on.
 The light source is a long-life halogen tungsten lamp with a
lifespan of more than 2000h. Long-term preheating of the lamp
will reduce the service life of the tungsten halogen lamp. It is
recommended that you do not select the ACC2 mode when it is not
in use for a long time.
The shake amplitude and shake frequency of the three shake modes are as follows:

1.6 Transportation
The Elisa Microplate Reader adopts three-layer packaging, in which the inner packaging
is moisture-proof plastic film, the middle packaging is rigid shock-proof foam and the
outer packaging is carton or wooden box.
When the Elisa Microplate Reader is transported, we need to use the special matching
packaging provided by our company. The Elisa Microplate Reader with complete
packaging can adopt transportation modes such as steam, iron, air and sea. The Elisa
Microplate Reader is a precision instrument. During the transportation process, it should
be protected from rain and snow, strong vibration, excessive heavy stacking pressure,
excessive temperature and humidity.

Chapter 2 Installation
Considering that the installation and operation of this equipment are easy and simple,
there is no need to send engineer for installation under normal circumstances.
The instrument is debugged and calibrated by professionals before leaving the factory.
After receiving the equipment, the customer can install and operate according to the
requirements of this chapter.
2.1 Requirement of Installation
Before the installation, user and engineer must check and confirm that the lab meets the
requirements of space, power supply and working environment, etc.
2.1.1 Space Requirements
To ensure enough space for releasing heat, repairing, maintenance, keeping the pipeline
not squeezed and ensure the fluid can flow freely, the space must meet the requirements
as follows:
 keep the instrument not less than 150mm distance from wall and other objects for
each side(left, right and back).
 There should be enough space where the power cord plug is inserted into the socket
to ensure that the power plug can be unplugged from the power socket quickly and
smoothly in an emergency.
2.1.2 Power Requirements
 Power supply: 100-240V~, 50Hz/60Hz
 Good grounding socket within one meter of the device.
Attention:
 The power socket should be within one meter of the analyzer to ensure that the plug
can be unplugged in time when an accident occurs.
 Check whether the instrument voltage is consistent with the grid voltage.
2.1.3 Environmental Requirements
The Elisa Microplate Reader should pay attention to dustproof, shockproof, away from
strong electromagnetic interference and corrosive places. The instrument should avoid
direct light, and should not be operated in an environment with excessive humidity or
large temperature changes. The instrument requires operation under the normal working
conditions specified in the environmental parameters in Section 1.2 above.
2.2 Unpacking
After the instrument arrives, please carefully check the packaging of the instrument to
see if there is any damage, such as broken or wet or polluted. If there is, please contact
our company. After confirming that there is no external damage, open it by following
steps:
 Open the box and according to the packing list to check whether the objects are
complete, if missing, please contact our company.

 Carefully check the appearance of the instrument, if damaged, please contact our
company timely.
 Check whether the serial number is in accord with packing list, outer package.
2.3 Installation Steps
1. Take out the Elisa Microplate Reader carefully after standing for 24 hours and place
it on a horizontal working platform (the instrument weighs about 11Kg, and the work
surface must be able to bear more than 50Kg).
2. Take out the ground cable to connect on the red grounding screw.
3. Connect the computer or printer according to the following chart.

Note: If the customer chooses a printer, the printer configuration baud rate before

4. When the conditions of storage or transportation (temperature, humidity) of the
instrument exceed the range of normal working conditions, the instrument should be
placed under normal working conditions for more than 24 hours before it is powered
on, and it is not allowed to be powered on immediately.
5. In the workstation mode, the computer operation part is composed of the computer
host, monitor, keyboard, mouse and printer. The requirements of computer:
 It must be a branded computer with a CPU frequency of at least 2.8GHz, a hard
disk capacity of at least 80G, at least 2 partitions (C, D...), at least 2GB of memory,
and a stable USB interface.
 Install Windows7 or higher version of the operating system, 32-bit or 64-bit version
is available, with and install Microsoft.NET FrameWork 4.0 and microplate reader
special software, it is recommended to install Microsoft OffceAccess software.
 Configuration requirements of display: 17 inches or more, resolution not less than
1366*768.
6. Under the premise of meeting the above requirements, connect the power cords of
the Elisa Microplate Reader and the printer to the network power supply.
Attention :
 This instrument is suitable for standard 96-well microplate for measurement.
 Protective grounding: The protective grounding of this instrument adopts the
method of connecting the power cord plug of the instrument to the protective ground
wire of the network power supply, so the power cord plug must be plugged into the
network power socket with reliable protective grounding when the instrument is
working.
 The instrument should not be placed close to the wall, and a space not less than
15cm should be left to ensure air circulation. There should be enough space where
the power cord plug is inserted into the network power supply to ensure that the
power plug can be quickly and smoothly in an emergency Unplug from the power
outlet.
 Unplug the power plug when the instrument replaces fuses, bulbs, maintenance or
long-term non-use.
 During the experiment, the tested samples and reagents may be infectious and
corrosive. Operators should pay attention to the use of effective protective tests,

and the waste after the experiment should be discarded in a dedicated waste box for
unified treatment. If you accidentally spill samples or reagents on the instrument
during the experiment, you should disconnect the power supply of the Elisa
Microplate Reader in time, and use the relevant methods in the cleaning section to
deal with it. If the instrument fails and needs to be repaired, it should be disinfected
before packaging , Transportation and maintenance.
2.4 Software Installation
For the workstation mode, the Elisa Microplate Reader analysis software and driver
need to be installed.
2.4.1 Installation of Analysis Software
This software is suitable for WIN7, WIN8, WIN10 operating system, open the software
installation folder, double-click the Setup.exe file to enter the installation program.
When installing the software, you must log in to the computer as the system
administrator to install, and it is recommended to turn off the antivirus software, or
remove the related block, allow all installation requirements of the program, and trust
the installed programs and files.
Note: If Microsoft.NET.Framework 4.0 is not installed on the computer, the software
supplement program license terms will pop up, click [Accept], if you have installed 4.0
or above framework, you will directly enter the interface of the installation program .

2.4.2 USB-RS232 Driver Installation
1. Connect the 9-pin COM end of the USB cable to the "computer" interface on the rear
panel of the Elisa Microplate Reader when the power is off, and plug the other USB
end into the USB port of the computer.
2. For Windows WIN7 operating systems or above, please click "
PL23XX-M_LogoDriver_Setup_v205_20210129.exe" in the "USB-RS232 Data
Line Driver Software" folder to install the driver, if not succeed, try
"pl2303_prolific_DriverInstaller_v1160.exe" to install the driver again.
3. After finishing the installation, plug in the USB cable again to complete the program
update.
4. Right-click on "This PC"-"Manage"-"Device Manager" to view the newly installed
Prolific USB-to-Serial Comm Port port number COM.

Chapter 3 Software Operation


This chapter is applicable to the operation of Elisa Microplate Reader in station mode
with computer.
Click the icon of the Elisa Microplate Reader software on the desktop, a login interface
pops up, enter the login password "1000" to enter the Elisa Microplate Reader software
interface as shown below:

The software system is divided into several functions: test, entry information, result
processing and basic settings, among which:
[Information entry]: Add, modify or delete the basic parameters of test items, samples or
QC products.
[Test]: Design the layout of samples, standards, and blanks for the entered items, start
the test. Users can store layout methods as templates, and directly call the layout
templates next time.
[Results processing]: You can view the test results, calibration and QC curves according
to the sample information or microplate information, and edit, delete or print the results.
[Basic settings]: Adjust the mechanical position and absorbance of the instrument, the
parameter information commonly used in preset reports, software user account
management and software version viewing.
3.1 Item Information
Click the item information can add, modify or delete items. Click [Add] on the right to
add a new item.
Note: [Item Information] function only allows senior users to edit, ordinary users have
no editing authority, the buttons on this interface are grayed out.

3.1.1 Qualitative Test Items

As shown in the figure, select the item in the items list on the left, and click [Modify] to
modify the item information.
Click [Add] to add a new item, select [Qualitative] on the item type, edit the project
name, unit, main wavelength, sub-wavelength, reference upper and lower limits in
sequence, the shake time and shake classification can be set, and the microplate move
mode defaults to stepping.
Qualitative test needs to add the Cut-Off calculation formula according to the reagent
specification. The user can input the judgment condition by himself, or after selecting
the "positive judgment condition", manually set the boundary value of the gray zone.
The boundary value is 100% of the Cut-Off valve by default. Click to modify it.
The reference valve is the reference range of the item. Select the reference valve, and
after the test, you would see the comparison between the reference value and the test
result in the sample result interface.
In the Cut-Off formula:
NC: Represents the average value of the negative control value.
PC: Represents the average value of the positive control value.
BLK: Represents the average value of the blank control value.
Cov: Represents the average value of the critical control.
After editing the item parameters, click [Save] to entry this item.

For quantitative test, input the standard concentration and select the calculation formula
according to the standard concentration value given in the reagent specification. This
instrument supports multiple types of curve fitting such as linear, semi-log, natural
logarithm, log-log, point-to-point, four-parameter equation and cubic spline.
After editing the item parameters, click [Save] to entry this item.
3.2 Sample Information

When the software is connected to the LIS system, you can directly select the entered

Click on the [QC information] to display the entered QC information, you can select or
modify or delete the entered QC in this interface.
Click [Add], enter the QC name and QC lot number, enter the corresponding target
value and SD value according to the QC product manual.

Choose the item name, select the Hole Category, and click or frame select on the
microplate layout on the left to layout the microplate. The completed layout can be
saved as a available layout by inputting the name of the illustrated position and clicking
[Save], which is convenient for direct recall next time.
Hole type
Sample For the sample to be tested, the Sample number and Retest times
must be set before the layout of the sample.
Blank For single-wavelength measurement, blank wells should be set as
control wells to calibrate the absorbance, and the solution in the
blank wells should be added in strict accordance with the amount
required in the kit instructions.
PC Positive control, used for qualitative measurement.
NC Negative control, used for qualitative measurement.
Critical Critical control, used for qualitative testing.
QC Quality control products are used to ensure the validity of each
experiment. If the test results do not fall within the acceptable
range established by the quality control product data, the
experimental data will be deemed invalid.
STD Standards, used to establish a standard curve.
Notice:
 The number will automatically increase during the layout process. When the [hole
type] is sample or SD, you can set the repetition number in the check box after the
[Retest number], the hole and its repeated holes will be used in the same number.
 When the hole type is selected as "QC", you need to select [QC Lot Number]
before arranging the QC. After the layout is finished, click [OP/CL] to open the
microplate carrier, place the microplate well and then close the carrier, and click
[Run] to test this microplate.
3.5 Results Processing

processing interface. After selecting the start and end time of the test, click [Query], and

the sample information column will filter the list of samples in the time period. Select
the sample, you can see the test results of the sample on all microplates in the [item
Information], including the OD value, S/CO value or concentration value, Cut-Off value,
and item reference value and qualitative results.
When the software is connected to the LIS system, you can select a sample in the
[Sample Information] column and click [Upload to LIS] to export all the test results of
the selected sample information.
If the print preview function is checked in advance, click [Print Current] to preview the test result
report of this sample, click [Print All] to preview the test result report of all samples filtered out in
the sample information column, click the upper left corner of the preview interface Can print the
report. If the print preview function is not checked, click the [Print] button to directly report the
corresponding report.
3.5.2 Plate Results Processing

Click on the [Plate result] on the left to enter the interface of microplate results. Select
the test time and microplate number, click [Query] to filter the results in special
condition.
After selecting the test result of a microplate, the test results of all samples on the same
microplate are displayed on the right, including OD value, S/CO value or concentration
value, item reference value and qualitative result. After selecting the microplate result,
you can right-click to delete the result. This method can delete the entire plate result or a
single test result on a microplate.
Click the left mouse button to select the test result. The upper right corner will calculate
the average value of the concentration value, the sample standard deviation SD and the
repeatability CV of the selected holes in real time.

[Print] button to preview the microplate test report as shown in the figure below, and

Click [Add] to manually add QC results. After clicking [Save], the QC curve and QC
result list on the left are automatically updated.
After selecting the quality control result, right-click to delete the result. After selecting
the quality control result, click [Upload to LIS] in the lower right corner to successfully
export the selected quality control result.
Click the [Print] button with the right mouse button to preview the printing effect of the
QC report, and click the [Print] button with the left mouse button to print the QC report.

Select the item and test time in the [Settings] column on the right, you can view the

as the current calibration. Right-click the test time to delete the calibration curve, and
right-click the curve part to copy, save or print the curve.

managed in user management. User accounts are classified into senior users and
ordinary users.
Select user information, senior users can click [Delete] to delete the user or click
[Modify] to modify the user name, user password or user level, and click [Add] to add a
new user account. For ordinary users, only new ordinary users are allowed to be added,
not allowed to modify, delete or enter a new senior user.

Click [Software Settings] in the user settings to perform basic software settings.
The parameter information commonly used when entering samples can be preset in the
setting list, such as the name of the commonly used sender, the name of the tester, the
sample type and the unit.
In the print order column, you can adjust the print order of the items by adjusting up,
down, and on top.
Check the icon [Print Preview] option, and click [Settings] to complete the print
preview function settings. All reports will be printed first to jump to the print preview
interface, and then print after confirmation. If this function is not checked, the test result
will be printed directly after clicking Print.
In the language setting bar, you can choose four modes: Chinese, English, Ukrainian
and Spanish, and click the setting to save.
The theme color can be selected in the theme setting bar, click the settings to save.
You can enter or modify the print title in the other setting column, and click the settings
to save.

select the connection method of LIS, and support network port and serial port
connection. After checking the icon [Auto upload], the sample results and quality
control results will be automatically uploaded to LIS after online interpretation and
acquisition of the results, without manual selection. After entering the connection
parameters, click [Connect] to connect to the LIS system.

Chapter 4 LCD Screen Operation


This chapter is only applicable to Elisa Microplate Reader in independent working
mode. For operation of Elisa Microplate Reader in workstation mode, please refer to
Chapter 3 Software Operation.
4.1 Boot Operation
Turn on the power switch, and check whether the LCD screen displays the menu or icon
normally. When the Elisa Microplate Reader is turned on, it will be initialized for about
one minute. Do not operate it during this period.
During operation, please note that after each click of the button, the instrument will
complete the action. Clicking the button before completion is invalid.
4.2 Main Menu

The instrument has function menu buttons such as absorbance test, qualitative test,
quantitative test, inhibition ratio test, data management, basic settings, etc. Click the
icon button on the main interface of the instrument to enter the corresponding function
submenu. Note: When the instrument is in ACC2 mode, a small light bulb will continue
to flash on the upper right corner of the main interface (see 4.2.1.1 for operation
details).
4.2.1 Settings
Setting functions include the settings of wavelength, shake and basic settings. In the
main interface, click [Settings] to enter the following [Settings] interface:

[System settings]: You can preset the wavelength, test mode, language, and see the data
storage capacity and software version.

[Shake Settings]: You can preset the shake time and shake classification.
[Print Test]: After the printer is online, you can click the [Print Test] button to print a
blank test page to check whether the printer is normally online.
[Light Inspection]: Users can perform self-inspection on the 8 channels of each filter
through the light inspection function.
[Light Positioning]: Including the adjustment of the mechanical position of the filter and
the microplate carrier, the adjustment of the absorbance and the
adjustment of the control circuit before leaving the factory. Note: This
function needs to be operated under the guidance of engineers or trained
professionals.
4.2.1.1 System Settings
Click [System Settings] in the settings interface to enter the interface as follows:

[Wavelength]: The filter No. 01 to No.10 in the figure represents the 1st position to 10th
position on the filter disc. Click the number at the wavelength position,
and enter the actual wavelength at the corresponding position through the
soft keyboard. The effective value is from 400 to 999. The number must
be 3 digits. For unused positions, you can enter 000 to mark. The default
wavelengths of the instrument on the 1st to 4th filter discs are 405, 450,
492, and 630 nm, click and enter the corresponding wavelength.
Note: The default wavelength of filter wheel 1~4 of this instrument is 405, 450, 492,
630nm respectively. If the customer needs to add or modify the wavelength of the filter
in the later stage, he needs to disassemble the machine to replace or add a new filter
under the guidance of the engineer, and manually click and store the new wavelength in
the system setting interface.
[BLK mode]: The blank test mode is divided into two test modes, 0 and 1. When it is 0,
you need to manually enter the BLK value before reading the plate. After
reading the plate, the screen display is the result of the OD value-the
BLK value. When the blank mode is 1, you can select a group of test
results in the data storage Set to blank, after reading the plate, the
displayed value on the screen is the OD value - the blank value of the
hole. At this time, manually inputting the BLK value is invalid.
[Test mode]: The test mode is divided into STD standard mode, ACC1 warm-up mode
and ACC2 light-on mode.

STD mode: turn on the light during the test.


ACC1 mode: Automatically turn on the light before the test to warm up and wait
for 60s to start the measurement.
ACC2 mode: The light will be turned on after the instrument is powered on, and it
will continue to warm up by default, and there is no need to wait
during the test.
Notice:
Note: Long-time turning on and preheating will reduce the service life of the
tungsten halogen lamp. It is recommended not to select ACC2 mode when the lamp
is turned on for a long time and not in use.
[Current data / curve storage]: Set the amount of data stored in the current storage area.
If it is displayed as 3, then 3 sets of test results have been stored in the
database.
[Language]: The language defaults to two language modes, Chinese and English, and
the user can freely switch the language. The instrument supports
customization of small languages such as Ukrainian and Spanish before
leaving the factory.
4.2.1.2 Shake Settings

In this interface, you can input the shake time, the maximum time can be set to 255s.
The shake classification is divided into H (high), M (medium), L (low) and CL (closed).
Click [Shake] to preview the effect of the shaking. After the shaking is finished, the
carrier will open and close once, and the mechanical position will return to zero. Click
[Save] to reserve the settings.
4.2.1.3 Print Test
After the Elisa microplate reader is connected to the printer (for printer connection,
please refer to Chapter 2.3 Installation Steps), click [Print Test] in the setting interface,
and the printer will automatically output paper as shown in the figure below, which
proves that the printer is connected properly.

4.2.1.4 Light Path Inspection
Click [light Inspection] in the settings interface to enter the light inspection interface to
check the lights of each wavelength.

 Click [Microplate on/off] to open the carrier to prevent the microplate carrier from
blocking the light path.
 Select the target position of the filter wheel, click [Y], the filter wheel rotates to the
optical path to be tested.
 Click [Light on/off] to turn on the halogen tungsten lamp and automatically start the
light source test. At this time, you can observe the A/D readings of the A~H eight
channels to check whether the light path is normal.
 Click [Y] of the light source test to stop the acquisition of A/D readings. At this
time, you can re-select the target position of the filter disc, and repeat the above
steps to check the A/D values of other light paths.
 In the process of optical path inspection, if it is found that the A/D value of the
eight channels of a certain filter is overall low and lower than 30000, the user can
input the gain value of 48000 and click [Gain]. If the A/D value of multiple filters is
lower than 30000 and repeated gain fails, consider whether the life of the halogen
lamp reaches the upper limit, and the user can refer to Chapter 5.6 to replace the
halogen lamp.
 After the light inspection is over, click [Light on/off] to turn off the tungsten
halogen lamp, click [Microplate on/off] to close the carrier to reset to zero [Y] to
reset the microplate carrier, and click on filter wheel return to zero [Y] to reset the

filter wheel.
4.2.2 Absorbance Test
The absorbance test is applicable to items that only need to test the sample absorbance.
Set the wavelength or use the last default wavelength to test the item.
Click the [Absorbance] button on the main menu interface to enter the interface:

1. Use the or button to select the required wavelength for measurement,


support single-wavelength measurement and dual-wavelength contrast measurement.
2. Click the [OP/CL] button to open the microplate carrier, place the microplate
carefully to prevent liquid spillage, be careful that there should be no debris or water
droplets at the bottom of the microplate.
3. If the BLK mode is set to 0, you can click to manually enter the blank control value,
and press the [Enter] key on the small keyboard after the input. If the BLK mode is
set to 1, there is no need to modify the BLK before testing value.
4. Click the [Read] button to do test. During the testing, do not touch the LCD screen
for any operations.
5. The test-data will be displayed on the LCD screen automatically. The displayed
value is the result after the BLK value is automatically subtracted. Click [OD] to
clear the BLK value, and the displayed value will return to the original OD value.
6. Click the [Save] button to store the test-data.
7. Click the [Print] button to print the test-data this time.
8. Click the [Back] button to return to the main menu.
4.2.3 Qualitative Test
The software has open Cut-Off formulas, which can input various special critical values
in the test. Click the [Qualitative] button on the main menu interface to enter the
interface as below:

9. Click the [Print] button to print the test-data this time.
10. Click the [Return] button to go back to the main menu interface.

Unlabeled holes on 96-well microplate will default to sample holes.

1. Click the or button to choose the sample type, and click on the
corresponding position on the LCD screen in correspond with the actual position on
the microplate to mark the layout.

2. Click the or button to select judgement type, when the absorbance value of
the sample is greater than or equal to the Cut-off value (critical value), the result is
judged as positive (+) or negative (-).
3. According to the reagent specification, set the coefficients and correction coefficients
in front of PC, NC, and Cm in the Cut-Off formula. Click the 0.000 digits in front of
PC, NC and Cm, to enter to input the control value. If it’s not set PC in the test, the
default value is 0.000.
4. In some items, the reagent specification stipulate "when the NC is is less than ..., it is
calculated according to ...." At this time, you need to enter the corresponding value, if
there is no such regulation, then ignore it.
5. Click the [Empty] button to clear all the contents of the layout.
6. After finishing the layout, click [Sure] to return to the test interface for reading or
qualitative analysis after reading.
4.2.4 Quantitative Test
The software is equipped with a linear fitting equation, with the function of linear or
logarithmic coordinates for the absorbance axis and concentration axis, respectively.
With the function of zero absorbance-concentration logarithmic curve fitting, which is
suitable for the detection of aflatoxin and melamine.
Click the [Quantitative] button on the main menu interface to enter the quantitative test
interface:

water droplets at the bottom of the microplate.
4. Click the [Read] button to do test. During the testing, do not touch the LCD screen
for any operations.
5. The test-data will be automatically displayed on the LCD screen.
6. Click the [Historical Curve] button to select the previously stored curve to fit this
measurement data. The concentration value can be directly calculated without setting
standards. (Note: If there is any standard on the layout interface, the recall of the
historical curve will be invalid.)
7. Click [Curve calc] to calculate the curve and update and display the blank control
value at the same time according to the layout.
8. Click the [Conc] button to calculate and display the concentration value of the
sample to be tested according to the layout.
9. Click the [Save] button to store the test data.
10. Click the [Print] button to print the test-data this time.
11. Click the [Return] button to go back to the main menu interface.
4.2.4.1 Curve
Click the [Curve] button in the quantitative test interface to enter the previously stored
quantitative curve selection interface:

1. Click or button to select the curve to be recalled. When selecting, the curve
equation bar can display the corresponding curve equation.
2. Choose curve, click [Sure] to return to the test interface, and clicks the [Conc] button
to calculate the concentration value of the testing item. [Note: When there is any
standard on the layout interface, calling the historical curve will be invalid]
3.Click the [Cancel] button to cancel the curve selection and use the [Layout] function
for standards layout.

Polyline: When the Polyline mode is selected for both the abscissa and the ordinate, the
standard curve adopts a polyline regression curve, the abscissa represents the
concentration, and the ordinate represents the absorbance.
Attention:
 After selecting the coordinate axis, the calculation formula of the microplate is
confirmed. The instrument supports three types of curve fitting: one-time linear,
semi-log and log-log. After connecting to a computer to form a workstation mode,
you can select multiple types of curve fitting such as linear, semi-log, natural
logarithm, log-log, point-to-point, four-parameter equation and cubic spline in the
supporting software.
 Please select the coordinate axis correctly according to the Elisa kit instructions. If
you make a wrong choice, the curve equation will not be generated correctly.

1. Click or button to choose the sample type, and click the corresponding
position on the LCD screen with the position of the well on the microplate to mark
the layout.
2. Select the X and Y axes of the standard curve, X-axis defaults to ND (concentration),
Y-axis defaults to OD (absorbance).
3. Enter the concentration value of ST1 ~ ST8 according to the reagent specification,
click the number in "ST1 = 0.000" to entering the corresponding concentration value
through the soft keyboard.
4. Click the [Empty] button to clear all the contents of the layout.

5. After finishing the layout, click the [Sure] button to return to the test interface to do
reading or quantitative test after reading.
4.2.5 Inhibition Ratio Test
The Elisa Microplate Reader is specially equipped with the inhibition ratio test function
for the detection of pesticide residues in foods such as fruits and vegetables.
The detection method and judgment standard adopt the national standard by default GB
/ T 5009.199 Rapid Detection of Organophosphorus and Carbamate Pesticide Residues
in Vegetables.
In order to fit the standards promulgated by the Ministry of Agriculture, the default
values of the response time and the result judgment in the software can be modified.
Click the [Inhibition Ratio] button on the main menu interface to enter the inhibition
ratio test interface:

1. Use the or button to choose the wavelength.


2. Click the [Layout] button to carry out the layout of the standards in the inhibition
ratio test. This [Layout] function supports layout before test or layout after test. For
details, see the following inhibition ratio layout section.
3. Click the [OP/CL] button to open the microplate carrier, place the microplate to be
tested carefully to prevent liquid spillage, be careful that there should be no debris or
water droplets at the bottom of the microplate.
4. Click the [Read] button to do test. During the testing, do not touch the LCD screen
for any operations.After the first testing, the microplate reacts in the microplate
carrier. and the second measurement will be performed automatically after the
reaction time is over.
5. The test-data will be automatically displayed on the LCD screen.
6. Click the [Inhibition Ratio] button to display the inhibition rate value of the sample
and display the△A0 value of standards according to the layout.
7. Click [+/-] button to calculate and display the positive or negative results of the
sample according to the layout.
8. Click the [Save] button to store the test-data.
9. Click the [Print] button to print the test-data this time.
10. Click the [Return] button to go back to the main menu interface.

2. Set the reaction time according to the reagent specification.
3. Set the limit value of the inhibition rate result judgment according to the reagent
specification.
4. Click the [Empty] button to clear all the contents of the layout.
5. After finishing the layout, click the [Sure] button to return to the test interface and do
reading or the analysis of the inhibition ratio after the reading.
4.2.6 Data
In independent working mode, 2000 sets of microplate data can be stored independently
in the lower computer software of the instrument, a total of 190,000 sample test results
can be displayed, and historical test data storage records can be displayed and printed.
After the test is completed, click [Save] to enter the save interface, click to enter the
data name and curve name, click the back [Save] button, when a green √ appears after
the data name or curve name, it represents the result data or curve The data is stored
successfully.

Click or button to select and display the name of the stored data, the first two
digits are the serial number, and the following are the names, data names support
Chinese, English and numbers.
After selecting the stored name, click the [Test], [Conc] or [+/-] button to display the
corresponding values respectively. When the blank mode is set to 1, after selecting a
group of data, click [Set to Blk] to set the result as blank valves.
Click the [Print] button to print the current viewing data. Click the [Del] button to delete
the choosing data.

5.1 Overview
5.1.1 Introduction
In order to ensure the reliable performance, good working condition and life of the
system, please operate and maintain the system in strict accordance with the
requirements of this user manual. Even if you are an operator, it is very important to
understand the operation and maintenance of this chapter. In-depth study will make the
instrument achieve the best running state and exert the best performance in use.
For the unsolvable problems encountered during use, the maintenance or repair
problems not covered in this chapter, please contact the company's customer service
center or the local distributor in time.

Warning:
 Do not perform maintenance work that is not explicitly stated in this
chapter. Failure to do so may result in system damage and personal
injury.
 Do not touch parts that are clearly documented and can be operated and
maintained by the user.
 Unauthorized repairs to the system can result in system damage and
personal injury, and the terms promised in the repair contract are no
longer valid.
 After the maintenance work is completed, please confirm that the system
is working properly.
 Do not spill liquids such as water or reagents on the mechanical or
electrical parts of the system.

5.2 Instrument Cleaning


The frequently touched places such as the instrument housing, touch screen and
microplate carrier are very easy to get dirty. In order to keep the working environment
clean and reduce biological risks, the exposed parts should be cleaned in due time.

Remove dust or other contaminants for keeping clean.
Maintenance opportunity
Perform this maintenance when dust or other contaminants accumulate on the exposed

 Dip a small amount of distilled water or neutral detergent with a soft cloth to clean
the surface of the instrument and the microplate carrier. After wiping, please dry the
instrument with a paper towel in time to avoid liquid retention.
 The optical path system is designed as a closed optical path system, and there is no
need to clean and maintain the optical components during normal use.
 Use a damp cloth when cleaning the instrument. Do not use a dripping cloth to wipe
the instrument to avoid excessive humidity.
 After the instrument is cleaned, power on and the microplate carrier will
automatically reset.
5.3 Instrument Disinfection
After using infectious hazardous substances, the instrument needs to be disinfected.
Precautions

Warning:
 Do not spill liquid on the analyzer to prevent liquid from
immersing and causing damage to the instrument.
 Before disinfecting the instrument, disconnect the device
from the external power source.
 Do not manually move the microplate carrier during the
disinfection process.

 All parts that come into contact with biological samples,
patient samples, positive quality control products, or
hazardous materials should be considered as potentially
infectious areas.
 During maintenance work, be sure to wear gloves and
work clothes to prevent infection and wear protective
glasses if necessary.
 Do not discard the gauze used for wiping. Please dispose
of it in accordance with relevant regulations.
Operation Steps
 Click [OP/CL] in any measurement interface to open the microplate carrier and
remove the microplate.
 Turn off the power switch, and unplug the power plug of the Elisa Microplate
Reader to ensure that the instrument is disconnected from the network power
supply.
 Wear disposable medical protective gloves and disinfect the instrument with a
disposable cloth moistened with 75% ethanol (the cloth should be damp, not
suitable for dripping).

5.4 Fuse Replacement

The operator must use fuse of appointed specification.


Operation steps
 When the analyzer is powered off, unplug the power cord from the power socket
(when the Elisa Microplate Reader only turns off the power switch and the power
cord plug is not unplugged, the fuse is live and may cause electric shock!)
 Pull out the fuse holder shown and replace the fuses.

5.5 Addition and Definition of Filters

Customers need to disassemble the rear parts of the instrument when adding new filters

or replacing filters. After adding or replacing the filter, the customer needs to redefine
the filter in the program. The above operations must be performed under the guidance of
engineers after familiarizing this section
Instrument status
When performing the disassembly and assembly operations in this maintenance, make
sure that the instrument is in a power-off state.
Operation steps
 Turn off the power switch of the Elisa Microplate Reader, and use a 2.5mm external
hexagon wrench to unscrew the two M3*16 screws of the filter cover of the Elisa
Microplate Reader behind the instrument.
 Rotate the filter disc to find the serial number of the filter position that needs to be
added or replaced.

 Use a Phillips screwdriver to loosen the M3*16 screws that fix the filter assembly,
remove the old filter assembly, and fix the new filter assembly to its original
position.
 Install the filter cover of the Elisa Microplate Reader as soon as possible (because
the optical path system is fully enclosed, if the filter wheel cover is not installed, it
will affect the linear index measured by the instrument, and if the filter enters the
dust, it will affect the optical path performance).
Definition of filter
After replacing the filter assembly, restart the device to check and redefine the optical
path. Take the addition of a 546 wavelength filter at the position of the filter wheel 5# as
an example:
Filter definition in independent working mode:
 Power on the instrument, click [Settings]-[System Settings] on the main interface of
the touch screen to enter the system settings interface. Click the blank space behind
No.5 and enter the newly added wavelength 546. Click [Save], the interface will
display "System Self-Check" and store the new wavelength.
 Return to the [Settings] interface, click [Light Inspection], click [Microplate on/off]
to open the carrier to prevent the microplate carrier from blocking the light path.
 .Select the target position of the filter wheel as 5, click [Y], the filter wheel rotates

to the the No.5 light path.


 Click [Light on/off] to turn on the halogen tungsten lamp and automatically start the
light source test. At this time, you can observe the A/D readings of the A~H eight
channels to check whether the light path is normal.
 Click [Y] of the light source test to stop the acquisition of A/D readings.
Filter definition in workstation mode:
 Find the configuration file config.ini under the installation path of the Elisa
microplate reader special software (the default installation path is D:\Program
Files\Elisa microplate reader), double-click to open it, find the [Filter_Settings]
byte, and add “|546” after the wavelength, as the picture shows:

Warning:

 The order of the wavelength in the configuration file must correspond


to the installation position of the filter.
 Do not confuse the position of the filter with the wavelength of the
filter, otherwise the measurement data will be inaccurate.

5.6 Replacement of Lamp


If light source lamp aging, light energy will deviate from the light measurement range.
During the sample test, it won’t test correctly because of interfere.
Maintenance opportunity
When the AD value of all optical path are less than 30,000, click [Filter] on the main
menu to do gain. If the gain fails, consider replacing the halogen lamp.

When performing this maintenance operation, make sure the instrument is out of power.

Precautions

Warning:

Do not touch the surface of the halogen lamp, otherwise it will affect
the amount of light. If the surface is found to have smudges such as
fingerprints, wipe it with a gauze dampened with alcohol.
Operation steps
 Prepare a new halogen lamp.
 Turn off the power switch of the instrument, and unplug the power plug (note: the
bulb and surrounding parts may be very hot, wait for more than 10 minutes after
shutdown).
 Unscrew the 7 pcs M3 * 8 screws fixing the upper cover assembly at the rear of the
instrument, slowly move the upper cover assembly of the Elisa Microplate Reader
towards the rear of the instrument by 5 ~ 10cm, pay attention to the FFC cable, then
remove the upper cover assembly upwards and place it next to the instrument.

 Loosen the M3*6 screws, remove the fixed spring of the lamp holder on one side,
and take out the tungsten halogen lamp and the lamp holder (be careful of burns).

 Unplug the old halogen lamp from the lamp holder, replace the same specification
model with the new one, replace the tungsten halogen lamp assembly to the original
position and reinstall it with spring.

 Reinstall the upper cover assembly of the instrument, tighten the 7 pcs fixing
screws on the back normally, pay attention to the slot between the front panel and
the front panel when installing the upper cover, and then push it forward when it is

Chapter 6 Troubleshooting
This chapter explains all kind of malfunctions, which often happen in the routine
operations. Besides, it analyzes the related reasons about malfunctions and supplies
some methods against the malfunctions. Please take measures to eliminate the
malfunctions which occur in use or before use according to relevant troubleshooting.
If the malfunctions still exist, please contact the after service of our company or our
local franchiser as soon as possible. We are pleasure to serve you.
Warnings:
 You must turn off the analyzer, cut off the power, and then remove the power plug
from the socket. The repair work must be taken by professional trained staff.
 The analyzer must use suited power supply and voltage. Or else, the damage which
is caused against this order is out of our responsibility.
Caution:
 Analysis of samples may give incorrect test results in the case of instrument
malfunction. If there is a fault detected in the sample, be sure to troubleshoot before
use.
 Sample, QC product, STD, wasted liquid and so on have potential biochemistryrisk.
The operator must comply with the laboratory regulations about the safety operator
to wear personal protective device (like: laboratory protective clothing, gloves etc.),
and accordance with local government regulations to dispose the waste materials
generated by the instrument detection.
