Activity # 03
Preparation of Citrated Plasma

I – Objective(s):
 To perform the proper procedure of collection using vacutainer method.
 To correctly separate the citrated plasma in the other tube using the automatic pippete.
 To centrifuge the blood sample in right speed and time in order to obtained the citrated
plasma.
II – Materials:
70% Alcohol Vacutainer set Automatic Pipette Gloves
Cotton balls Blue top tube Pipette tips TT rack
Lancet Plain tube Centrifuge

III – Procedure:
01. Wash/alcoholize hands, pad dry & put on gloves.
02. Prepare the necessary materials (vacutainer set).
03. Position the patient properly for easy & comfortable access to the site of puncture.
04. Apply tourniquet 3 inches above the site of puncture, let patient make a fist & palpate the
vein.
05. Disinfect the site of puncture w/ a cotton ball wet w/ 70% alcohol & allow to dry.
06. Obtain 5 mL blood by using blue top evacuated tube.
07. Release tourniquet, let patient open fist, place a dry cotton ball on top of the needle &
withdraw needle from the vein.
08. Dispose properly all used materials in designated biohazard container.
09. Mix tube by gentle inversion.
10. Centrifuge tube at 3500 pm for 5 mins.
11. Transfer the citrated plasma in another tube by using automatic pipettor.

IV – Observation & Drawing


- In this laboratory activity, we performed vacutainer method wherein we extract blood and
it is directly transfer to the tube. When the blood is in the tube, we centrifuged it and we
noticed the separation of plasma and blood. The blood in the centrifuged sample is the
RBC is packed, and the plasma is yellowish in color.
V – Conclusion:

The vacutainer method was used in this laboratory activity. The vacutainer method is
vital in modern medicine because it allows for reliable and safe blood collection for a variety of
diagnostic purposes.
In blood collection tubes, sodium citrate is commonly used as an anticoagulant to produce
citrated plasma. This is due to the fact that sodium citrate acts as a reversible anticoagulant,
preventing blood clotting by binding to and chelating calcium ions, which are required for
coagulation.
Citrated plasma is a type of blood sample that is commonly used in medical laboratories to
perform coagulation testing. It is obtained by mixing blood with citrate, an anticoagulant that
prevents clotting. The use of citrated plasma allows for precise measurement of clotting factors
and other coagulation parameters, aiding in the diagnosis of bleeding disorders, monitoring
anticoagulant therapy, and assessing thrombosis risk. Overall, citrated plasma is a crucial
component in coagulation testing and contributes significantly to the diagnosis and treatment of
various medical conditions.
