1 Introduction
This report summarises a study performed to assess the antibacterial performance of paint
formulation either without or with an antibacterial additive against Escherichia coli,
Staphylococcus aureus, Pseudomonas aeruginosa, Methicillin-resistant Staphylococcus aureus,
Klebsiella pneumoniae, Salmonella typhimurium, Listeria monocytogenes, Streptococcus
pyogenes, Enterococcus faecalis, Shigella sp, Legionella pneumophila SG 1 and the Spores of
Bacillus subtilis using the method described in the standard ISO 22196 : 2011.
2 Test Materials
Liquid paint was supplied by Jotun U.A.E. Ltd.. Flexible test panels (Leneta scrub resistance test
panels) were coated (300 µm wet film thickness) with the paint formulation prepared with an
antimicrobial agent.
A sample of unfortified paintwas supplied by IMSL to act as a reference material. All samples
were held in the dark at 20°C prior to testing and the coated panels were equilibrated at 20°C for
7 days and then statically leached for 24 hours to eliminate any volatile components from the
paint film.
3 Methods
Antibacterial activity was determined using the method described in ISO 22196 : 2011 (Ref 1).
An aliquot (225µl) of a log phase cell suspension of either Escherichia coli (ATCC 8739),
Staphylococcus aureus (ATCC 6538P), Pseudomonas aeruginosa (ATCC 15442),
Methicillin-resistant Staphylococcus aureus (MRSA) (NCTC 12493), Klebsiella pneumoniae
(ATCC 10031), Salmonella typhimurium (ATCC 14028), Listeria monocytogenes (ATCC 7644),
Streptococcus pyogenes (ATCC 19615), Enterococcus faecalis (ATCC 29212), Shigella sp
(ATCC 12022), Legionella pneumophila (SG 1) (ATCC 33152) or the spores of Bacillus subtilis
(ATCC 6633) prepared using the method described in ISO 22196 (no nutrient broth was used for
the preparation of the spores of Bacillus subtilis and a higher concentration of nutrient broth was
employed in the carrying medium for Legionella pneumophila) were held in intimate contact with
each of 3 replicates of the test surfaces supplied using a 30 x 30 mm polyethylene film (cut from
a sterile Stomacher bag) for 24 hours at 35°C.
The size of the surviving population was determined using the method described in ISO 22196.
The viable cells in the suspension were enumerated by spiral dilution on to Trypcase Soya Agar
or other appropriate media and by the pour plate method described in ISO 22196 , where
possible. These plates were incubated at 35°C for 24 hours or up to 96 hours for Legionella
pneumophila and then counted. The method is described schematically in Figure 1 below.
All data were converted to colony forming units (CFU) cm-2 and then transformed (Log10) to
provide a data set that conformed to a Gaussian distribution. Potential outliers were tested using
Dixon’s Q-test (P = 0.05).

It can be seen from the results above that the population of Escherichia coli held in contact with the
IMSL paint increased in size by 1.9 orders of magnitude during the 24 hour contact interval. This is
considered a normal response for this species on an inert surface under the conditions imposed by ISO
22196. The populations of Escherichia coli exposed to the surfaces of Fenomatic Hygiene Matt
(Blank) declined by 0.2 orders of magnitude and the populations of Escherichia coli exposed to the
surfaces Fenomatic Hygiene Silk (Blank) increased by 0.2 orders of magnitude after 24 hours
compared to the initial population.
In contrast, the population of Escherichia coli exposed to the surfaces of Fenomatic Hygiene Matt
(Preserved) and Fenomatic Hygiene Silk (Preserved) declined by > 4.0 and > 4.1 orders of magnitude
respectively to below the limit of detection after 24 hours compared to their relevant controls.

It can be seen from the results above that the population of Staphylococcus aureus held in contact
with the IMSL paint declined by 67.89% during the 24 hour contact interval. This is again considered
a normal response for this species on an inert surface under the conditions imposed by ISO 22196.
Similarly, the population of Staphylococcus aureus held in contact with the surfaces of Fenomatic
Hygiene Matt (Blank) and Fenomatic Hygiene Silk (Blank) declined by 78.85% and 91.17%
respectively during the 24 hour contact interval. In contrast, the population of Staphylococcus aureus
exposed to the surfaces of Fenomatic Hygiene Matt (Preserved) and Fenomatic Hygiene Silk
(Preserved) declined by > 99.97% and > 99.93% respectively to below the limit of detection after 24
hours compared to their relevant controls.

It can be seen from the results above that the population of Pseudomonas aeruginosa held in
contact with the IMSL polypropylene increased in size by 2.2 orders of magnitude during the 24
hour contact interval. This is considered a normal response for this species on an inert surface
under the conditions imposed by ISO 22196.
Similarly, the population of Pseudomonas aeruginosa held in contact with the samples of
Fenomatic Hygiene Matt (Blank) and Fenomatic Hygiene Silk ((Blank) increased in size by 1.7
and 2.2 orders of magnitude respectively during the 24 hour contact interval. In contrast, the
population of Pseudomonas aeruginosa exposed to the surfaces of Fenomatic Hygiene Matt
(Preserved) and Fenomatic Hygiene Silk (Preserved) declined by 2.8 and 3.3 orders of magnitude
respectively after 24 hours compared to the appropriate control.

It can be seen from the results above that the population of MRSA held in contact with the IMSL
paint remained relatively constant during the 24 hour contact interval. The population of MRSA held
in contact with the surfaces of Fenomatic Hygiene Matt (Blank) and Fenomatic Hygiene Silk (Blank)
declined by 83.32% and 84.54% respectively during the 24 hour contact interval.
In contrast, the population of MRSA exposed to the surfaces of Fenomatic Hygiene Matt (Preserved)
and Fenomatic Hygiene Silk (Preserved) declined by 99.89% and > 99.95% respectively after 24
hours compared to their relevant controls.

Fenomatic Hygiene Silk (Preserved) declined by > 5.8 and > 4.9 orders of magnitude respectively
to below the limit of detection after 24 hours compared to the appropriate control.

Fenomatic Hygiene Matt (Blank) and Fenomatic Hygiene Silk ((Blank) increased in size by 0.9
and 0.8 orders of magnitude respectively during the 24 hour contact interval. In contrast, the
population of Salmonella typhimurium exposed to the surfaces of Fenomatic Hygiene Matt
(Preserved) and Fenomatic Hygiene Silk (Preserved) declined by > 5.1 and > 5.0 orders of
magnitude respectively to below the limit of detection after 24 hours compared to the relevant
control.

It can be seen from the results above that the population of Listeria monocytogenes held in contact
with the IMSL paint remained relatively constant during the 24 hour contact interval. The population
of Listeria monocytogenes held in contact with the surfaces of Fenomatic Hygiene Matt (Blank) and
Fenomatic Hygiene Silk (Blank) declined by 74.19% and 36.81% respectively during the 24 hour
contact interval.
In contrast, the population of Listeria monocytogenes exposed to the surfaces of Fenomatic Hygiene
Matt (Preserved) and Fenomatic Hygiene Silk (Preserved) declined by > 99.98 and > 99.99%
respectively after 24 hours compared to their relevant controls.

It can be seen from the results in Table 8 that the population of Streptococcus pyogenes held in
contact with the IMSL paint declined by 46.43% during the 24 hour contact interval. Similarly, the
population of Streptococcus pyogenes held in contact with the surfaces of Fenomatic Hygiene Matt
(Blank) and Fenomatic Hygiene Silk (Blank) declined by 79.94% and 87.35% respectively during the
24 hour contact interval.
In contrast, the population of Streptococcus pyogenes exposed to the surfaces of Fenomatic Hygiene
Matt (Preserved) and Fenomatic Hygiene Silk (Preserved) declined by > 99.95% and 99.89%
respectively 24 hours compared to their relevant controls.

in contact with the samples of Fenomatic Hygiene Matt (Blank) and Fenomatic Hygiene Silk
((Blank) increased in size by 1.0 and 0.2 orders of magnitude respectively after 24 hours
compared to the initial population.
In contrast, the population of Shigella sp. exposed to the surfaces of Fenomatic Hygiene Matt
(Preserved) and Fenomatic Hygiene Silk (Preserved) declined by > 5.1 and > 4.3 orders of
magnitude respectively to below the limit of detection after 24 hours compared to the relevant
control.

and Fenomatic Hygiene Silk (Preserved) declined by 27.38%, 30.72%, 26.42 and 52.14%
respectively after 24 hours compared to the initial population.

1 Introduction
This report details a study performed to assess the resistance of paint prepared either with or without
biocide to fungal growth using BS3900 Part G6.
2 Test Materials
Liquid paint was supplied by Jotun U.A.E. Ltd.. Flexible test panels (Leneta scrub resistance test
panels) were coated (300 µm wet film thickness) with the paint formulations. A sample of unfortified
paint was supplied by IMSL to act as a reference material.
All samples were held in the dark at 20°C prior to testing and the coated panels were equilibrated at
20°C for 7 days and then statically leached for 24 hours to eliminate any volatile components from
the paint film.
3 Methods
Anti-fungal activity was determined using BS3900 Part G6. Individual panels were inoculated with
a consortium of fungi (see Table 1) and then incubated under humid conditions to stimulate their
growth.
Three replicate panels (each ca 100 mm x 50 mm) of each test system were inoculated (1000 µl per
sample) with a mixed spore suspension (> 105 spores ml-1 of each species) of the fungi described in
BS3900 part G6 (see Table 1 and Ref. 1 for details).
The samples were then allowed to stand at ambient room temperature overnight then transferred to
humid chambers (each containing 200g sterile vermiculite moistened with 800g sterile water) and
incubated in the dark at 20°C for 20 Weeks.
After incubation the surface of each test panels was inspected for growth and rated in accordance with
BS3900 Part G6 and the appearance recorded photographically.
The ratings of the test to BS3900 Part G6 are shown in Table 2. The gross appearance and
microscopical appearance of the test specimens are shown in Plates 1 - 4.

It can be seen from the results above and Plate 1 below that the IMSL control coating supported
growth of a range of fungi following incubation for 12 Weeks.
The sample of Fenomatic Hygiene Matt (Blank) showed 30% - 70% coverage of fungal growth after
12 weeks incubation. In contrast, the sample of Fenomatic Hygiene Matt (Preserved) showed no
growth on the test surface after 20 weeks incubation indicating that the addition of an antifungal
additive to paint has demonstrated an antifungal effect under the conditions of BS3900.
The sample of Fenomatic Hygiene Silk (Blank) showed 1% - 10% coverage of fungal growth after
12 weeks incubation increasing to 10% - 30% coverage of fungal growth after 20 weeks incubation.
The sample of Fenomatic Hygiene Silk (Preserved) showed some germination of the spores on the
surface and hence was given a rating of trace to 1% growth on the test surface after 12 weeks
incubation, this gemination did not produce mycelial growth across the surface in the following 8
weeks indicating that the addition of an antifungal additive to paint has demonstrated an antifungal
effect under the conditions of BS3900.
