1.0. PURPOSE:
To lay down the procedure for Cleaning of Microbiology Laboratory.
2.0. SCOPE:
This sop is applicable for cleaning of Microbiology Lab.
3.0. RESPONSIBILITY:
Lab Attendant, House keeper.
4.0. ACCOUNTABILITY:
Microbiology Lab Incharge, Microbiologist.
5.0. GENERAL INSTRUCTIONS:

a. Perform the cleaning operation in Microbiology lab which includes:
 Sterility Testing Area and Change Rooms

 Microbiology Lab (Documentation Room)
 Bio-Assay room.
 Autoclave room
b. All disinfectant solutions shall be prepared freshly.
c. Use Purified Water for preparing disinfectant solutions.
d. Use clean dusters for cleaning of Microbiology Testing area and its change room,
Autoclave room and Bio-Assay room.
e. Record the preparation of disinfectant solutions.
f. Maintain records of daily and weekly cleaning in cleaning record of Respective areas.
6.0. PROCEDURE:
6.1. DISINFECTANT SOLUTIONS:
 Dettol 2.5%.
 Descocid 01%.
 Triacid 03%.
 IPA 70%.

6.2. PREPARATION OF DISINFECTANT SOLUTIONS:

6.2.1. Dettol 2.5%:
Take 500ml of purified water add 25ml of Dettol and make-up volume of1000ml
with autoclaved purified water. Mixwell.
6.2.2. Descocid 01%:
Take 500ml of purified water add 10ml of Descocid and make-up volume of
1000ml with autoclaved purified water. Mix well. Use within a week.
6.2.3. Triacid 03%:
Take 500ml of purified water add 30ml of Triacid and make-up volume
of1000ml with autoclaved purified water. Mix well. Use within a week.
6.2.4. IPA 70%:
Take 100ml of purified water add 700ml of IPA and make-up volume of 1000ml
with autoclaved purified water and then filter through 0.45µm filter
paper. Mix well. Use within a week.
6.3. DAILY CLEANING PROCEDURE:
6.3.1. STERILITY ROOM:
 After completion of day’s work Laminar Air Flow, SS rack, SS trolley should be
clean by using Triacid 3% and IPA 70%.
 Clean the floor using a non-shedding duster/mop.
 Mop the area in uni-direction.
 Subsequently mop the floor using non-shedding mop dipped in the Descocid
01%.
 Maintain records of daily cleaning in cleaning record of Sterility room.

6.3.2. BIOASSAY ROOM:
 Before starting or after completion of day’s work clean the Laminar Air Flow
using IPA 70%, on each operating shift in Laminar.
 Subsequently mop the floor using non-shedding mop dipped in the Dettol 2.5%.
 After every moping clean the non-shedding wiping mops in Purified water.
Then again dip in the Dettol 2.5% and use.
 Maintain records of daily cleaning in cleaning record of Microbiology
department.
6.3.3. MICROBIOLOGY LAB (DOCUMENTATION ROOM):
 Before starting or after completion of day’s work clean the floor using a non-
shedding duster/mop.
 De-dust the instruments, windows and tables with clean dry duster.

6.3.4. AUTOCLAVE ROOM:
 Before starting or after completion of day’s work clean the floor using a non-
shedding duster/mop.

6.4. WEEKLY CLEANING PROCEDURE:
6.4.1. STERILITY ROOM:
 After completion of weekly operations Laminar Air Flow, SS rack, SS trolley,

door should be clean by using Triacid 3% and IPA 70%.
 Clean the walls and floor using a non-shedding duster/mop.
 The pre-filters of Laminar Air Flow should be washed or maintained on bi
monthly basis.
 Subsequently mop the floor using non-shedding mop dipped in the Descocid
01%.
 Make sure that complete surfaces, walls, floor has been cleaned by trained
supervisor.
 Fumigation should be done on bi monthly basis in order to control the area.
 Maintain records of weekly cleaning in cleaning record of Sterility Room.
6.4.2. BIOASSAY ROOM:
 Before starting or after completion of day’s work clean the Laminar Air Flow,
SS Table and the incubator using IPA 70%.
 Maintain records of weekly cleaning in cleaning record of Microbiology
6.4.3. MICROBIOLOGY LAB (DOCUMENTATION ROOM):
 Before starting or after completion of day’s work clean the Tables, Incubators
using IPA 70%.
 Incubators should be clean from inner side on weekly basis with IPA 70%.

6.4.4. AUTOCLAVE ROOM:
 Before starting or after completion of day’s work clean the sink using
detergent and bleach.
 Autoclave should be clean from inner side on weekly basis.
 After every moping clean the non-shedding wiping mops in Purified water, then
again dip in the Dettol 2.5% and use.

7.0. PRECAUTIONS:
7.1. Perform the area cleaning when there is no activity.

7.2. Use proper safety apparels such as gowns, rubber hand gloves and safety goggles
during preparation and use of Disinfectants.
7.3. Always add disinfectant to water while diluting.
7.4. Do not use expired disinfectant solutions.
7.5. For sterility area cleaning use freshly prepared 0.45µm filtered disinfectants and use
within one week.
7.6. Wherever suitable use vacuum cleaner. Ensure the vacuum cleaners are in clean and
dry condition before use.
7.7. Use separate lint free mop and fresh solution for cleaning of different area to avoid
contamination of mop and solution.
