The document outlines the requirements for the Biology practical exam for Class XII CBSE students. It includes 3 sections: a major experiment involving isolating DNA from plant material, a minor experiment using the quadrat method to study plant population density or frequency, and preparing microscope slides showing pollen germination or mitosis in onion root tip cells. Students will also need to identify specimens in a spotting question and be evaluated on their practical records, project work, and viva voce.

Isolation of DNA from given plant material separate it by spooling. Write observations, preparation of solution and precautions taken by you while performing an experiment.
2. MINOR EXPT. : (4 marks)
Study plant population density by using Quadrat method. Write requirements, observations and two precautions taken by you while performing an experiment. Calculate density of given plant species and discuss/write the vegetation composition of the area and comment on the dominant component species. OR Study plant population frequency by using Quadrat method. Write requirements, percentage frequency formula, observations and two precautions taken by you while performing an experiment. Calculate percentage of frequency of given plant species. Write comments on observations taken by you.
3. SLIDE PREPARATION : (5 marks)
Prepare a temporary mount showing “Pollen germination “on a cavity slide. Draw well labelled diagram and write constituents of nutrient medium used. Also write two precautions taken by you while preparing the slides. OR Prepare a temporary mount of onion root tip to study Mitosis. Show any one stage of Mitotic division and draw a well labelled diagram. Write observations (any two features of the stage) and two precaution taken by while preparing the slide. 4. SPOTTING : (7 marks) Identify the spot A to G and comment.
