1 Definition
‟QuEChERS" (Quick、Easy、Cheap、Effective、Rugged、Safe) is a multi-residue analysis m
ethod of pesticides, veterinary drugs and mycotoxins in food and agricultural products.
This procedure uses a single extraction in acetonitrile and requires a very small (10-15
g) sample.
Compared to traditional sample preparation techniques, QuEChERS solutions offer:
1. Decreased time spent on sample preparation
2. Efficient and cost-effective sample preparation
3. Reliable, high-quality product in a convenient format

2 Methods
Today, there are 2 commonly used buffered methods.
European Committee for Standardization (CEN) Method 15662 uses a citrate buffer for e
xtraction. The second method uses acetic acid buffer and was adopted as the AOAC off
icial method 2007.01

3 Operation steps
Step 1. Prepare the sample for extraction
Step 2. Transfer the sample to a 50ml Centrifuge tube
Step 3. Add extraction solvent and shake
Step 4. Add buffering salts and Centrifuge
Step 5. Transfer supernatant to dispersive SPE cleanup tube and shake
Step 6. Centrifuge cleanup tube
Step 7. Obtain final extract and place into autosampler vial for analysis

3.1 Prepare the sample for extraction

A: The sample shall be homogenized. For
dry commodities such as cereals, it is nece
ssary to either homogenize the sample in
water or hydrate the commodity by soakin

3.2 Transfer the sample to a 50ml Centrifuge tube

Q: What’s the sample size?

A: 15g sample for AOAC method While 10g sample For CEN method.

3.3 Add extraction solvent and shake

Q: What's the ratio of additions?

A: Add acetonitrile by dispenser on the ratio of 1:1.

3.4 Add buffering salts and Centrifuge

Q: How to choose buffering salts?

A: AOAC 2007.01: For base sensitive pesticides many researchers prefer the AOAC aceta
te buffered extraction. The acetate buffered method yields extracts with a nominal pH n
ear 5.
CEN 15662: To obtain consistent and reliable recovery of base insensitive pesticides, usi
ng the CEN method. The citrate buffered extract has a nominal pH near 8. which is hig
her than the extract obtained from the acetate buffered method.

3.5 Transfer supernatant to dispersive SPE cleanup tube and shake

Q: How to choose a purification tube?

A: The clean-up tubes are available in the standard 2 mL size as well as a 15 mL size
for sample enrichment.
Q: How to remove impurities?
A: C18 removes very non-polar interferences, such as fats;
PSA removes polar interferences, including sugars and organic acids;
Graphitized carbon black (GCB) effectively removes pigments such as chlorophyll and car
otenoids;

Sorbents including graphitized carbon black and C18 are also available in bulk for meth
ods requiring the removal of excess fat or high amounts of chlorophyll.
