Aim:
Total leucocytic count.
Introduction:
Leukocyte or WBC count is determined to decide whether an infectious activity is
present and should average between 5000 and 10,000 cells / mm3.3 An increase in the
number of WBCs, called leukocytosis, is essential for phagocytosis (cell death of
microorganisms) and may indicate the presence of an acute infectious process.
Leukocytosis may also be associated with inflammation and may arise during treatment
with postoperative inflammation. Decreased WBC count from baseline, called
leukopenia, may indicate altered immunity or the presence of infection that exhausts the
supply of certain WBCs. Decreased WBC count relative to a previously high count (i.e.
becoming more within the norm).
Materials:
1- Microscope
2- Haemocytometer
3- Pipette

Discussion and Calculation:

20 μl blood was inserted into an eppendorf. Mixed pipetting has been applied to 380 μl of
WBC count solution. This solution was moved to the sheet of the haemocytometer.
Dilution number : (20/(380+20))=0.05
Average cells number : 7
