The document lists 10 room requirements for a medical laboratory, including sample grossing, processing, staining, section cutting, doctor, store, reporting, electrophoresis, media preparation, and sample processing rooms. It also provides procedures for performing an ESR test using a Helico disposable ESR pipette and collecting and analyzing a blood sample. Reticulocyte counting methods are outlined for manual and automated methods, including expressing the count as a percentage, absolute number, or reticulocyte index.

Collect 1.6 mL of whole blood and add 0.4mL of Sodium citrate 3.8%
Procedure:
1. Mix the Blood gently in tube.
2. Gently insert lower end of ESR Pipette(having the vacuum plug) into the blood collection tube and using continuous force push the pipette down to the bottom of the tube. 3. The blood will automatically rise into the pipette and stop at “0” mark. 4. Place the unit (Tube+ ESR Pipette) absolute vertically in ESR stand. 5. Allow the blood cells to sediment for 60 minuites. 6. At the end of 60 minuites,the numerical results are used in millimeters directly from the imprinted scale on the pipette.
Disposal: discard the tubes, pipette as per biomedical waste procedure.
Corrected ESR= measured ESRx15/55 for hct <35%
Reticulocyte count For Manual method Reticulocyte = No. of reticulocytes /100 RBC Whenever a RBC count is available, express Reticulocytes in absolute numbers. Absolute Reticulocyte count = of Reticulocytes x Total RBC Reticulocyte Index- When RBC count is not available, it is recommended that Reticulocyte count be expressed as a Reticulocyte Index R.1. { R.1. = (Reticulocyte x PCV)/ 45.}
