Microplate Reader is composed of light source system, monochromator system, sample
chamber, detector and other components.
1.1.4 Application scope
PKL PPC 142 can read microplates used in Enzyme-Linked Immunology Assay (ELISA) for the
qualitative and quantitative analysis, which is used widely for immunoassay in clinic lab, blood
center, hospital, university, institute or other academic organization.

1.3 Warranty and service


Free repair service since installation can be enjoyed for one year. However, free repair service
will be not available for the following conditions:
1) Working environment does not meet requirements of the specification;
2) Damage caused by unspecified power supply or abnormal power;
3) Man-made damage caused by improper operation.
4) Damage caused by unauthorized man's repair work.
5) Other irresistible natural factors, such as earthquakes, fires, war damage.

2 . Safety and precaution

2.1 Safety classification


a. Microplate Reader is class I for anti-electric shock, class Ⅱ for overvoltag
ordinary desktop contaminated equipment;

2.2 Safety
a. Biological contamination: All test samples, control materials, calibration materials, etc.,
should be regarded as contagious, and any components contacted with sample shall be
regarded as contagious as well, you should wear gloves when contact ;
b. High temperature: You can change the lamp after you turn off the device more than 10
minutes when lamp becomes normal temperature.
c. Physical injury: Don’t put hands to the moving mechanism when PKL PPC 142 is working.

2.3 Notes
a. If the device does not work in the way specified by the manufacturer, it may damage the
protective function in device itself;
b. Do not place the device where it is difficult to turn it off.

3.1 Installation condition


The Installation Microplate Reader can be only done by PARAMEDICAL srl. Appropriate
environment and space should be provided by customers during installation.

3.2 Damage check


It was strictly checked by QC Team before shipping. When you open the packing, please check
carefully. Please note if there is following damage or not:
(1) There is deformation for outside carton.
(2) There is obvious flood damage for outside carton.
(3) There is obvious hit damage for outside carton.
(4) Cartons seem to be opened before.
If you find any damage as above listed, please immediately notify local agents.
If packaging is intact, please open the box, and check together with distributor all the parts
(1) Check all parts according to packing list.
(2) Check carefully if there are broken, bruises or deformation damage for device shell
appearances or not;

3.3 Packing List


Check it with packing list in the box.

3.4 Installation requirements


(1) Place: Dry, clean, stable working desk, where there is clean air and enough space around
device to ensure air circulation. And there is NO dust, shock, strong magnetic fields, the impact of
interference voltage, and direct sunlight.
(2) Temperature: 5 ~ 35°C;
(3) Humidity: ≤ 85%;
(4) Atmospheric pressure: 86 ~ 106Kpa;
(5) Ventilation: It can exchange air with the outside world, and it can flow air smoothly;

3.5 Installation
Open box and take out device from the box, and keep it away from water and unstable desk;
Open box and take out the accessory kit and power adapter cable. Connect one end of the
power adapter back into the instrument's power jack and the other end with the power cord;
Check whether parts in device are loose or not;
Connect the power cable into plug.

5.1 Turn it on
Press the red button in the front panel, which will make the indicator lamp light.
After Built-in computer starts, it will enter windows XP and show following interface.
Double-click lnk HSIMM as below to start data analysis software for microplate reader.
Software had already been installed in model PKL PPC 142 in factory.
Instrument will automatically do self-test to light source, filter and movement mechanism
respectively. If there is anything unusual, screen will show that there is an error. The instrument
would be used better after start up 15 minutes.

5.2 Password, User right and Main menu


a) Password: Normally there is no password in software. Sometimes factory password is 123.

b) User right: Admin is the manager who can create, delete other user. Admin can operate
some system operation as below shows:

c) Main menu:
After starting HSIMM software,it will show the main menu as follows:
Title :It shows the name of software is “Micro-plate system(v2.1)”.
Menu: Pull-down menu to execute the main function

5.3 Test
Click Test button to input/edit sample information, measuring sample in different ways: Rapid
test, Single Test, Multi-test.

5.3.1 Sample information

Sample information can be inputted, modified, checked and deleted. After you click sample
information, it shows as below:

When you open Sample Information window, it only displays today’s sample information by
system default. Same sample No. is not allowed in the same day, if date is different, same
sample No. is allowed, which means you can’t name same sample No. in the same day. All
samples information in the same day are displayed on the right side, while each sample details
are displayed on the left side.
1) Save
Input sample information (such as name, Sex, Age, specimen type, blood type, ID No. and
submission doctor, etc.) in the left. You can also click drop-down menu in the chosen item, and
then press ENTER to complete input. After inputted, click Save button. Sample number will be
automatically added, and current sample information will be automatically displayed on the
right.
2)Delete
If need to delete sample information, you can click Delete after the sample No. had been
selected in the right list. Prompt will show as below:

Click yes to delete what you have selected, and click No to cancel deletion.
3)Modify:
Select the sample you want to modify in the left column or the right sample information list.
When you single click modify, sample number and reading date become gray which indicates
that it can’t be operated since then. But all other functions can be modified. In this case, click
save S can save modified information. sample number and reading date will be restored.
4) Check(Find)
Choose starting date and ending date. Click Find, it can show all sample information during
selected time. When sample No.or name is shown, click check, all information you selected will
be shown.

5.3.2 Rapid test


Rapid test is a quick detection for whole plate, it supports single-wavelength and
double-wavelength detection. OD values and results can be directly printed out , when a blank
hole is set up, it will automatically subtract OD value of the blank hole . It is a fast way for
reading optical density value and there are various calculations according to optical density,
such as A1-A2, A2-A1, line 1 - line 2, line 2 - line 1. 1)Click Rapid test, it will show as follows:

2) Plate No.:It can be set by users according to date and project. The plate No. will be
increased automatically.
3)Plate style: Plate style is the positional relation of each well in the microplate. In the right
drop-down list, style can be chosen. If it changes, you can modify it through the read kinds
setting (details refers to sample type setting in the manual), in order to get the plate type you
want. Regular plate style can be saved for future usage.

4) Choose wavelength: When you choose single wavelength, WL1 should be selected. When
you choose double wavelength, both WL1 and WL2 should be selected, and they can not be
the same wavelength.
5)Judgment formula:According to Cutoff calculation formula in reagent manual, input blank
value 、negative control , positive control and their calculation relationship with Cutoff value.
To choose Positive value or Cutoff value by single clicking < or > . Proportions between
Little Positive and CUTOFF can be set by inputting percentage parameters.

6)Sample style setting:


You can also use start No. starting from 1. You can input the starting number as you want. Use
mouse to drag it to the column. If drag the mouse horizontally, it will be as follows:

If drag the mouse vertically, the sample number will be arranged vertically. After complete one
drag, it will show next number.

Click above NC (negative), PC (positive), HP(high positive), LP(little positive), QC( quality
control) and BLK (blank). Select one you want, and then drag it into main interface. In this
process, it should be corresponding with the real position of reagent in microplate.
7) Click Test, device start testing according to project parameters. Later measurement data is
immediately displayed in the data display. If you click Close Window before start measuring, it
will cancel measurement and return to the main interface.
8)Click Save to store the results, Export, Print can be used only after results had been saved
successfully.
9)If you need to test another new plate, please click Test new plate to enter initial Rapid test
interface, repeat same steps as above.
10) Go back main interface: After Rapid test, please click <close window>to go back the main
interface.

★Note:Rapid test can’t do quantitative testing, whose testing results can only be
printed out or exported.

5.3.3 Single Test:


When using Single-item Test , you need edit the sample style in the Edit Menu in advance,
Please place a standard material especially when it is a quantitative test. Otherwise it will show
following error.

In this case, you should click Edit in the main menu, select Project Parameters. In the
parameter setting interface, set the project parameters ( refers to 5.5 Edit on page 27). Click
single test and it will show following interface.

1)【Test Item】
a. Plate No: Use system default number.
b. Plate Style: in the right drop-down list, style can be chosen. If it changes, you can modify it

through the read kinds setting (details refers to sample type setting in the manual), in order to
get the plate type you want. Regular plate style can be saved for future usage.

c. Single click to select the project horizontally and vertically, if select the project horizontally,
the colors corresponding to the line A-H. If vertical row is selected, the corresponding item
on the color shown in column 1-12.
d. Project selection: project module is selected by clicking, in the data list. It is displayed by the
color module.

2)【Test Parameters】
a. Click < Test Parameters >, enter test parameters interface.

b. Choose wavelength: All should be set in the Edit menu instead of here. When you select
double wavelength, WL1 and WL2 should be selected separately. When you select single
wavelength, WL1 should be selected.
c. Cutoff, positive judgment can only be displayed but can’t be modified here. If you need
change it, you can return the Edit menu, to change the project parameters.
d. Testing type settings: Select sample, input Start No. Same project can’t use same sample
number in different projects. When the panel has multi-well blank or and multi-well negative
control, software will automatically calculate the average value.
Drag the mouse to select the test area, if horizontally drag the mouse, the sample number will
be horizontally arranged as below:

If you vertically drag the mouse, sample number will be vertically arranged.
Click NC (negative), PC (positive), HP(high positive), LP(little positive), QC( quality control)
and BLK (blank). Check their boxes below to add what you want. Click the appropriate location;
you can set the corresponding negative, positive, strong positive, little positive, quality control,

blank holes. In this process, it need be corresponding with the microplate status.
Note: Blank and the control of projects involved in the calculation (including Positive,
Negative, Standard, QC) should be set completely.

3)Click Test, instrument panel executes on ELISA optical density test. And then current
measurement data will be calculated, together with optical density result of holes in the data
display. If click "Close Window" before you start measuring, it returns the main interface,
and it will cancel the measurement.
Note: If setting is incomplete, please click Test, instrument will prompt parameter is
set incompletely. You can recall historical records or re-enter the missing holes
in the OD value of control. The method is same as “Recalculate."
4)Click Recalculate. If it is qualitative test, it will show following Standard

Historical value in the same item is shown in the left list, including BLK, NC, PC, LP, HP, etc. In
the right side will show the current value in the control wells. Users can click some historical
control data to modify, or control thoroughly the data in the optical density of a well. Click Select
to use modified control data, to recalculate the current board test results on the optical density.
Current plate result together with revised control value can be saved.
5) Click Recalculate. If it is quantitative test, it shows following standard curve as follows:

Users can change the curve type or the corresponding standard optical density to re-fit the
curve. You can also select the historical curve to modify current project type and standard
optical density. And then click Select to recalculate the optical density in current test results.
Results are kept together with the modified curve.
6)Click Save, test data can be saved, click Export (Note: it can export to EXCEL ), can be
saved in the data into EXCEL, and also you can directly click Print. Once it is completed,
click Close Window to return the main interface.

5.3.4 Multi test


Before multi-test, you need place sample style in the Edit menu, especially name QC for
quantitative test. Otherwise it will prompt the following error.

In this case, you should click Edit in the main menu, select Project Parameters. In the
parameter setting interface, set the project parameters up there as 5.5 Edit in page 27 says.

Click multi-test and it will show following interface.

1)【Test Item】
a. Plate No: Use system default number.
b. Plate Style: in the right drop-down list, style can be chosen. If it changes, you can modify it

the plate type you want. Regular plate style can be saved for future usage.

3)Click Test, instrument can identify the wavelength of all items in the current microplate. And
it execute absorbance detection at all wavelengths, same wavelength detects only one time,
whose test result data and the absorbance data will be shown at the data area. If click Close
Window before you start it, it return the main interface, which means it can’t be measured.
4)Click Save, test data can be saved, click Export (Note: it can export to EXCEL only when
you save), can be saved in the data into EXCEL, and also you can directly click Print .Once it is
completed, click Close Window to the main interface.

5.3.5 Quit and turn it off


Click Quit, it will quit the software. And then you can turn it off.

5.4 Report
In the main interface, click drop-down menu Report:

5.4.1 Plate report


Click the drop-down menu to enter the interface of plate report. Click the screen below the
key “ ”,“ ”to select different microplate data, click on the interface below the “ ”,
“ ”select the starting and ending of microplate data respectively.
Select microplate to be printed, and click the "Print" to print data.

Report by plates is a uniform reporting of all specimens for single rapid test absorbance,
multi-tests for microplate, negative and positive, quantitative value and the S / CO values, etc
on the same microplate, the specific formats as follows,

5.4.2 Combinatory report


Click the "Report” of drop-down list in the "Combination Report" into the interface of
combined Report.
Combinatory report is to collect the same specimen number’s summary of all test items in the
same report. Each project settings in the project can be ordered in project settings.

5.4.3 Standard curve


It can be used to print standard curve in quantitative test as below:

5.5 Edit
Click drop-down menu Edit, select Project Parameters to enter the program parameter setting
interface.

1) Qualitative measurement
Click Project Parameters as below:

【Qualitative parameters settings】


It is an existing project in the left. You can click the "+" to add new items, and then input English
name, project name, result unit, test wavelength, reference wavelength, project color, and
project code in the detail parameters. Choose measurement modes between qualitative and
quantitative. Qualitative measurements need set up critical value, judgment conditions, scope

of validity and suspicious setting. Quantitative measurement of parameters should be same as


reagents requirement.
Judgment conditions means that cutoff value range will be related to instructions. Such as the
reagent instructions, "If negative control OD value is less than 0.05, we calculate it as 0.05, If
higher than 0.05, we calculate it with real terms". In this case, judgment conditions should be
set to NC=MAX (0.05, NC).
Validity means that whether reagents and reaction conditions of the experiment meet the
requirements or not. For example, reagent manual says "positive control OD value greater than
0.8, the negative control OD value is less than 0.12, the experiment is effective", the
effectiveness should be set to "(PC> 0.8) |(NC <0.12.)”
Suspicious range refers to the range of absorbance in qualitative experiment values at cutoff
specimens near the floating range of optical density, based on cutoff value centigrade
boundaries.
Cutoff value refers to determine the absorbance value of positive samples. Calculated method
is according to reagent instructions, the instrument provides the PC, NC, LP, HP, BLK of the
four arithmetic operations, and also provides two logical judgment: MIN (A, B) and MAX (A, B).
By clicking positive method mode, "> Cutoff values is positive" and "< Cutoff values is
positive" to achieve the mutual conversion.

2)【Quantitative parameters settings】


Quantitative measurement need set up the result unit, normal reference range, standard
sample’s quality, concentration and the standard curve type, etc.
(1) Results reporting units: The list contains commonly used software in the concentration of
units, such as mol / L, mmoL / L, umoL / L, etc.
(2) Normal reference range: Units should be consistent with units of the project report, the
settings will be printed in the data report;
(3) Standard quantity and concentration: It should be set according to the instructions provided
by the standard reagent.
(4) Standard curve type: This software provides a powerful curve fitting function, the Y axis
represents OD of standard curve or sample, and X axis represents concentration value of the
standard substance or samples.
The curve types are the following:
a. Linear
The linear function fits the best straight line to the data. The equation for this fit has the form of:
Y = A * X+B, Where B is the y-intercept of the line and A is the slope.
A linear fit should be used whenever the standard values appear to lie on or are scattered
around a straight line.

b. Quadratic

The quadratic function fits the best parabola to the data. The parabola is a curved line based on the
equation:
y = (A * x^2) + (B * x) + C, where C is the intercept, B is the slope of the curve at the intercept, and A
is the measure of the curvature of the parabola.
The quadratic fit is most appropriate when the standard curve has a tendency to curve up or down.
c. Semi-Log
The semi-log function fits the best straight line to a set of data for log(X) plotted against Y. The
resulting curve displayed will be a straight line with the X axis drawn in logarithmic scale. The
equation for a semi-log fit is:
Y = B + [A* log10(X)], where B is the y-intercept of the line and A is the slope (note: log in this
equation is the common or base-10 logarithm).
d. Log-Log
The log-log function fits the best straight line to the set of data which consists of the logarithm of the
readings on the Y-axis (the response) and the logarithm of the dose on the X-axis. The resulting
display will be a straight line with both axes drawn in logarithmic scale. The equation for the log-log
fit is:
log10(Y) = B + [A * log10(X)], where B is the log10Y-intercept of the line for log10(X) = 0 and A is the
slope (note: log in this equation is the common or base-10 logarithm).

e. Point-to-Point

A linear equation is fit to each pair of data points. Fit parameters are not given. The point-to-point
curve fit is a linear fit composed of pieces that assume a linear relationship between each pair of
data points. The line segment defined by each pair is used to interpolate data between those points.
Since there are multiple line segments, fit parameters are not shown on the graph.

f. Four-Parametric

The Four-Parametric function fits the best straight line to the data. The equation for this fit has
the form of:
Y = (A-D)/(1+(X/C)^B) + D,
Most of reagent reaction should use Four-Parametric function according to some researches.

5.6 System
1)Click the "System"

2) Click "Initialization", reset the instrument components, including light bulbs, filters and return
the initial tray position.
3)Filter: Click the "System" drop-down menu, select "Optical Filter" and click into the filter
screen, showing the four filters microplate reader corresponding wavelength are 405nm,
450nm, 492nm, 630nm, according to user’s requirements, different wavelength filters can
be added, you can fill in the 5,6,7,8 positions to add the wavelengths, click the "Save" to
save settings, click "Close" the filter interface, and then back the main interface.

4) Absorbance correction: Click the "System" drop-down menu, select "Absorbance correction"
and click into the absorption correction interface, where the manufacturers engineers need to
adjust the password to enter the program, mainly for calibration when there are large difference
between the wavelengths of light density values and the standard value of standard substance.
This function is not available to users. New equipment is not to be calibrated when it is send
out.

5)Operating parameters: Click "System" drop-down menu, select <operating parameters> and
click to enter the operating parameters of interface

Operating parameters include the setting name of the units, connecting port settings and
sampling parameters. Name of the users’ organization will be printed on the results of the report;
connecting port is parameters for two-way communication between external control computer
equipment and the instrument. Sampling parameters is amplification needed to process the
original signals received by the software
WARNING: For changing, it requires setting parameters of the manufacturer’s engineers

to ensure proper use.


6)Clean Database: Only clear "sample information" and "project testing" of the record, when
the data is larger than usual, in order to ensure the system work efficiently, please backup
database before bulk delete historical records.

Select time period for records need to be cleared, when the project list is not marked with"√",
the record, can’t be deleted. Click "Confirm", the system pops up the following dialog box.

Click "Confirm", the system pops up, that record deleted successfully.
7)Backup Database: In order to ensure data security, you can regularly back up the system
database to other disks of the computer

Click the button to select the backup to backup directory and its name, click "confirm (S)"
instrument tips Backup successfully.
8)Recovery Database: When users need to query the historical records, just back up and
restore the database, the system software can query, print historical records.
Note: When restoring the database, the system will coverage the data currently being
used, so make sure to back up the database before restoring.

you, “Are you sure to recovery”. Click "Yes" to recovery successfully.

9)Commissioning Panel: Click the "System” in drop-down menu, select " Commissioning Panel
" and click to enter the external pass test interface, mainly for engineers in the
maintenance of equipment after the equipment used in debugging. This function is not opened
to the user, only used by the commissioning engineer debug.

6. Daily Maintenance

6.1 Maintenance
1) In order to ensure continued stability and accuracy of PKL PPC 142 Microplate Reader, It
should be avoided interference any part of the optical system. Optical path of adjustment error
can affect measurement results.
2) Maintaining the optical system clean to ensure the accuracy of the results of normal function
and should avoid liquid flow into the equipment. Prevent dust completely from endogenous
substances , do not touch the lens surfaces, filters, photoelectric detector with fingers.

6.2 Clean equipment


1) Turn off equipment and unplug the power plug.
2) Use disposable gloves, wet disposable cloth with water or mild detergent to clean equipment
outside, rail and carrier aircraft.

6.3 Clean the optical system


1) Remove the outer cover retaining screws, remove the lower cover, use above method to
clean the tracts in the measurement system.
Note: Do not touch the measurement circuit board.
2) Use a neutral cloth and wet it with mild detergent or 70% ethanol, clean the eight condensing
lens at the back of optical fiber bundle, such as using 96% ethanol, and then using the lint-free
wet cloth to wipe it. The upper lens is fixed under the detecting bracket, take off the bracket of
photoelectric detector from the fixed frame with a gasket, lift the bracket of photoelectric

detector with hands, clean upper lens with the above methods.
Note: Do not use allethrolone to clean plastic lens, softly clean.
3) After clean the clean upper lens, replace the bracket of photoelectric detector.
Note: Must replace the pillar sensor into the slot.
4) Replace the fixed frame with a gasket of the bracket of photoelectric detector, check the
alignment of edge of the fixed frame, ensure the fixed frame of the bracket of photoelectric
detector well replaced.
Note: Do not fold the wires of circuit board.
5) Put the bottom wires back in place. Turn the cover screws well. Screws connect cover and
the bottom of device to cut off the circuit interference.
6) Use lint-free cloth or lens paper to clean the filters, note that when you wipe the filters, do not
use liquid.
6.4 Replace spare parts
Equipment replacement and purchase of spare parts are required to provide and change
by manufacturers or their agents, the following details of the replacement parts:

A) Replace a single filter


1) Turn off equipment and unplug the power cord, open the cover.
2) Rotate the filter wheel to replace the filter at the disassembling end of the holder; do not
touch the filter surface.
3) Rotating the female screw of the fixed filter, remove the filter and the neutral gray film.
4) Insert a new filter; pay attention to the right direction of the filter.
5) Fixed the female screw of the fixed filter.
6) Close the cover and tighten all screws.
B) Replacing the Lamp
If the lamp is broken, according following steps to dismantling:
1) Turn off equipment and unplug the power plug, wait 10min, after the cooling, unscrew the
two screws on each side of the instrument cover, open it.
Note: The lamp and around may be hot, to guard against burn your hand.
2) Remove out the lamp together with the socket, and then remove the lamp from the socket.
Do not touch the lamp and the light parts.
3) Install the new light lamp (6V10W)with the socket, replace the new lamp.
4) Close the instrument cover and tighten the screws, connect the power, turn on the
instrument.
C) Replace the neutral gray film
1) Turn off equipment and unplug the power cord, open the cover.
2) Do not touch the neutral gray surface.
3) Rotating the female screw which fixing the neutral gray film, remove the neutral gray film.
4) Insert a new piece of neutral gray film.
5) Fixed the female screw which fixing the neutral gray film in its place.

6) Close the cover and tighten the screws.


D) Replacement power adapter
Turn off the instrument power, and disconnect the power plug, unplug the power adapter, then
replace the power adapter, the plug back.
