1. POLICY STATEMENT:
All Medical Technologists shall be oriented and have skills in performing blood
crossmatching procedure using gel method to achieve precise and reliable results prior to blood
transfusion.

2. PURPOSE:
To help the Medical Technologist in performing the procedure and determining whether
donor blood is compatible (or incompatible) with recipient blood.

3. SCOPE:
This covers from the receipt of sample and compatibility testing request up to the release of
result.

4. EQUIPMENT/REAGENT/SUPPLIES:
3.1 Matrix card warmer
3.2 Matrix card centrifuge
3.3 Patient sample (EDTA)
3.4 AHG Gel card
3.5 Matrix LISS
3.6 Marker
3.7 Test tube (12x75mm)
3.8 Centrifuge
3.9 Plastic Pasteur pipette
3.10 Scissors/
3.11 Ball pen
3.12 Mechanical pipette

4. TECHNICAL PROCEDURE:
All Medical Technologists shall wear the prescribed Personal Protective Equipments (PPEs)
such as laboratory gown, gloves and mask in performing the crossmatching procedure.

4.1 Centrifuge patient sample at 3400rpm for 5 minutes.


4.2 Cut segment from donor unit blood bag

4.3 Copy and record blood bag number and date of expiry.
4.4 Prepare test tube needed for blood typing and for red cell suspension, label poperly
4.5 Prepare gel suspension. Using mechanical pipette, pipette1000uL matix LISS to a each
clean test tube for Donor’s RCS and Patient”s RCS
4.6 Prepare gel card and label properly.

4.7 Add 10uL PRBC from patient’s sample to 1000uL LISS for Patient;s RCS
4.8 Add 10uL PRBC from donor’s sample/segment to 1000uL for Donor’s RCS
4.9 From Patient’s RCS and Donor’s RCS perform blood grouping same with tube method
4.10 From Donor’s RCS pipette 50uL into MC (Major Crossmatch) PAGE:
well 2of- 2AHG gel card
4.11 From Patient’s RCS pipette 50uL into AC (Autocontrol) well of AHG gel card
4.12 Add 25uL patient’s plasma or serum into same MC and AC well of AHG gel card
4.13 Incubate AHG gel card @ 37C for 15 mins.
4.14 Centrifuge gel cards for 10 mins
4.15 Interpret results and record to blood compatibility worksheet
4.16 Encode result to blood compatibility template
4.17 Validate result including patient and donor record prior to printing of result
4.18 Counter check results by other Medical Technologist

5. INTERPRETATION OF RESULT
(Refer to printed image)

6. RELEASING OF RESULT
Endorsed result to the clerk on duty for endorsement to ward.
