Compartment Model-Based Analysis
Multiple Compartment Model
Multi-compartment Models explains the observation that after a rapid IV injection the
plasma level-time curve does not decline linearly as a single first-order rate process. In a multi-
compartment model, the drug distributes at various rates into different tissues. The tissues with
the highest blood flow may equilibrate with the plasma compartments. The highly perfused
tissues and blood make up the central compartment.
Cp = Ae-at + Be-at
Where
A = 12 µg/mL
B = 0.17 µg/mL
a = 13 hour -1
b = 0.06 hour -1
Calculate:
a. The volume of the central compartment
2. Mitenko and Ogilvie (1973) demonstrated that theophylline followed a two compartment
pharmacokinetic model in human subjects. After administering a single intravenous dose
(5.6mg/kg) in nine normal volunteers, these investigators demonstrated that the equation best
describing theophylline kinetics in human was
