1. Draw and label the parts of a micropipette.


Answer:
1. Plunger/Button: The plunger or button is used to aspirate (draw in)
and dispense liquid. By pressing the plunger, you can create suction to
draw liquid into the pipette tip.
2. Tip Ejector: A tip ejector button is used to eject used pipette tips
safely after use, preventing contamination and ensuring accurate
measurements in subsequent uses.
3. Volumes Window: Some micro pipettes feature a display window
where the set volume is shown. This allows users to verify the volume
before dispensing.
4. Shaft: The shaft connects the plunger/button to the tip ejector and the
tip cone. It houses the internal mechanisms responsible for volume
adjustment and liquid aspiration/dispensing.
5. Tip: The tip cone is the part of the pipette where the disposable pipette tip
is attached. It ensures a secure fit of the tip and prevents leakage during
pipetting.

2. Illustrate the following steps:


A. Drawing up liquid
Answer:
 Attach a disposable tip to the micropipette.
 Set the desired volume using the volume adjustment.
 Immerse the tip into the liquid.
 Depress the plunger slowly to draw up the liquid into the tip.
B. Dispensing liquid
Answer:
 Position the tip over the target container.
 Depress the plunger to expel the liquid.
 Release the plunger slowly to avoid introducing air into the liquid.
3. List five Do’s and five Don’ts in using a micropipette:
Do’s:
 Use the correct size of disposable tips for the micropipette.
 Calibrate the micropipette regularly to ensure accuracy.
 Hold the micropipette vertically when drawing or dispensing liquid.
 Prime the pipette by drawing and expelling liquid before use.
 Use a pipette aid or pump for efficient and comfortable operation.
Don’ts:
 Over-tighten the disposable tip; it may damage the pipette.
 Use excessive force when depressing the plunger; it may damage the pipette or introduce errors.
 Lay the micropipette on its side when it contains liquid.
 Pipette by mouth; always use the appropriate equipment.
 Use a micropipette beyond its specified volume range.

C. P200: 73.0 µL
D. P20: 1.3 µL
