PRACS IH (45 mins procedure & 15 mins extract)

1. Extract — clot (wag masyado matagal sa paghintay ng clot)

• *If wala ka kasabay* gawa ka extra tube for balancer sa centri

2. Centri 10 mins (if not clear-ring-centri) or yung tinuro ni ma’am na teknik

• Habang nagcecentri maglabel ka tubes & lagay antisera at known A & B
• Kuha ka na rin 1-2mL donor blood para sa donor RCS
• I-ready mo na rin yung 1.9mL NSS each in 2 tubes

3. Get 12 tubes and label

1. Patient (Initials):
1. Forward (A, B, & D): 3 TUBES
• 1 drop Px RCS each + 1 drop Antisera A, B, & D
2. Reverse (A & B): 2 TUBES
• 1 drop Px Serum each + + 1 drop Known A & B (from Donor RCS)

2. Donor (4 numbers):
1. Forward (A, B, & D): 3 TUBES
• 1 drop Donor RCS each + 1 drop Antisera A, B, & D

3. Middle tube (X + Full Name)

• For forward and reverse: centri 30s-1minute | dislodge | MAMAYA KA NA

4. (Practice mo muna) Use pipette to drop Px RCS, Px Serum, and Donor RCS

5. Make RCS (for patient and donor)

• Sabay mo gagawin yung patient and donor RCS – no need for balancer sa centri
1. SEPARATE SERUM
2. Wash Red Cells
• Fill 3/4 tube with NSS (wag punuin sa 3/4 agad mix mo muna)
• Cover with parafilm
• Centri 1 min
• Decant NSS
• Repeat 3 times
3. On 3rd wash after decanting, 100uL washed red cell + 1.9ml/1900uL NSS (both
Px and Donor)
CROSSMATCHING:
GET X TUBE
1st phase (Immediate Spin Phase):
1. Get X Tube
2. 2 drops px serum
3. 1 drop donor RCS
4. centri 1 min
5. Read if may agglu/wala (Sa tube)

2nd phase (Thermo Phase):

1. Get same X Tube
2. Put 2 drops LISS
3. Water bath: Incubate @37C for 15 mins
4. Centri 1 min
6. Read if may agglu/wala (Sa tube)

3rd phase (Anti-Human Globulin Phase):

1. Wash with nss 3x (decant-centri-wash)
2. 3rd wash-decant all nss
3. Add 2 drops of AHG rgt
4. Centri 30s-1minute-dislodge
5. Check in microscope (must: no agglutination)
1. Have agglutination : not compatible
2. No Agglutination: proceed next step
4. Agglutination: add 1 drop of check cells
1. Centri 30s-1min
2. Read microscope (must: agglutination)

FOR INTERPRETATION:
Ex. Pag O blood type ng patient – Known A and B lang may agglutination sa iba wala na kaya
wag mo na icheck yung iba kasi sayang sa time
• May grading yung sample ng patient at donor, ng blood type, ng IS, ng Thermo, ng AHG,
at CC tapos microscopy
