OBJECTIVES
At the endPresentations
of an 25 mins discussion, learners
are communication
toolsto:
must be able that can be used as lectures

A define spectrophotometer,

B identify the components of

spectrophotometer and;

C cite the importance of

spectrophotometer in analytical

READY?
Direction: The students will be divided into 3 groups, they will
decode the given number that is equivalent to letters to be able
to know the hidden word behind it with the help of hints
provided. The words that they will get will be needed in the next
activity.

FILL ME
UP!
Direction: Using the same group. Arrange and
name the core components of the
spectrophotometer and present your work by
answering the guide question.
GUIDE QUESTION:
1. Based on the arrangement, What is the primary function of a
spectrophotometer?
2. How do you think spectrophotometers measure the
concentration of substances?
SPECTROPHOTOMETER
Spectrophotometer
measures how much light
is absorbed by, reflected
off, or transmitted
through a chemical
substance by measuring
the intensity of light as
the beam passes through
a sample.
SPECTROPHOTOMETER

CORE COMPONENTS:
1. Light source: An internal lamp that is the origination point for measurement light.
2. Collimator: Can be a mirror or a lens, depending on instrument configuration. Bends
light so that photons travel parallel to each other.
3. Prism: Prisms and diffraction gratings refract light to separate out the different
wavelengths. Rotating the prism changes the wavelengths that fall over the hole of
the slit card, allowing the user to select a wavelength for use.

4. Slit or Aperture: In some instruments, the size of the hole through which light
passes can be controlled ('slit width' value). Only light that falls over the hole will be
permitted to fall on the sample.
5.Sample holder: It's where the cuvette goes!
6.Detector: Most often, these are photovoltaic cells. Such components convert
the intensity of light falling upon them to a change in voltage in a circuit, thus
converting a light signal to an electrical signal that can be interpreted by software.
