MICROBIAL QUALITY EVALUATION OF FOOD CONTACT SURFACES

SAMPLE COLLECTION

Procedure:

A total of 100 square cm surface was swabbed from a plate in an eatery on the campus. The
sampling was carried out by measuring an area of 20cm square using a cardboard paper as a
guide. The swab stick was soaked in the test tube containing normal saline and used to swab
underneath the cardboard which was placed slightly above the plate and repeated 4 more times
on different portions on the plate. Before each repetition, the swab stick was thoroughly swirled
in the saline solution to elute any picked microorganisms.

The sterile swab was inserted into the test tube and brought back to the laboratory.

Aim: to evaluate microbiological standards of sanitation of food contact surface.

Materials used:

- 4 x 5 cm 20sq cm Cardboard paper

- Sterile swab stick

- Screw-cap test tube (containing physiological saline solution – as diluent)

INNOCULATION OF SWAB SPECIMENS

Procedure:

• Serial dilution was made by serially transferring 1ml from the sample-containing bottle into 4
different sample bottles containing 9ml of normal saline.

• The last dilution was made by inoculating 0.1ml on two separate plates of Nutrient Agar
medium, thus culminating to 5 serial dilutions(x105).

• The inoculum was then incubated at 37°C for 48 hours.

Result readings:

• Growth was observed on the medium showing slight raised whitish round colonies.

Total count on both plates = 35+23 = 58

Average count on plate =

58 ÷ 2= 29

=29.0 x 105

=2.90 x 106 CFU


The total count per square centimeter is obtained by dividing the total colony-forming units by
100

= 2.90 x 106 ÷ 100

= 2.90 x 106 ÷ 102

= 2.90 x 106-2

= 2.90 x 104 CFU/cm2

Interpretation: Bacterial count was 2.90 x 104 which was higher than 150 CFU/cm2 which
indicated inadequate sanitation.

Advise: The restaurant should improve their sanitary practices
