Bromeline in pineapple

Task:
To investigate the effect of the enzyme bromeline on gelatines ability to coagulate.

Introduction:
Enzymes are greatly affected by their working conditions such as temperature and acidity. You will
test this by extracting enzymes from pineapple and treat these enzymes to different conditions.
After treatment their ability to work will be tested on a house-hold gel (a protein mixture).

Materials.
Beaker
Canned pineapple
Raw pineapple
Graduated cylinder 10 ml
Labels
Filter paper
Small glass funnel
Test tubes in holder (rack)
Bundsen burner
Matches
Gelatine in the form of "husblas"

Experimental:

1) Soften two gelatine plates in cold water in a beaker for 5 min. Afterwards heat them gently in
approximately 100 mL of water in the beaker using the Bundsen burner as a heat source, stir constantly
underway.
2) Once all of the gelatine has dissolved in the water turn off the heat and let the solution cool down for
~10 minutes.
3) While the solution is cooling down start preparing the fresh pineapple juice by cutting small pieces of
raw pineapple and crushing them in a handheld blender.
4) Once 6 mL of fresh juice has been achieved (through filter paper in the glass funnel) take out 3 mL
and boil it in a test tube for 10 seconds using the Bundsen burner to do so.
5) Distribute the gelatine solution evenly between four test tubes and label the tubes #1 to #4.
6) Ad the following to the labelled test tubes:
#1: 3 mL tap water
#2: 3 mL of untreated raw juice
#3: 3 mL of boiled, raw juice
#4: 3 mL of juice from canned pineapple
7) Put the tubes in your rack and place it in the refrigerator with your name until next class.

Results:
For your group PowerPoint presentation: slide 1) Create a hypothesis based on your knowledge of
enzymes. Slide 2) Define your variables (independent, dependent and controlled variables). Slide 3)
Collect the data and find a way of making a clear representation of your results. Slide 4) Give a valid
conclusion and evaluate the experiment.
