Gene therapy, bone marrow stem cells manipulation and pre-clinical animal models. Cell/ Molecular techniques: Generation of recombinant DNA constructs and chimeric lentivirus particles, shRNA, sample preparation for microarray, PCR, Real Time PCR, Immunoprecipitaiton, pull down assay, Western Blot, Reporter assays, FACS, Packaging recombinant lentivirus particles, Propagation of recombinant adenovirus, cell cytotoxicity assays and anti-cancer efficacy assays. Pathology: Histology, Histomorphometry, quantitative Immunohistochemistry. Cell culture: Cancer cells, chondrocyte, osteoblast, stem cells, their differentiation and staining. Surgical skills- bone marrow ablation model, fracture healing model, injection of cells or lentivirus particles into tibia by intra-tibial injections, surgical implantation of cell graft, alginate beads or osmotic pump subcutaneously. Imaging- Advanced computer aided imaging system as IVIS 200 imaging system, fluoresce and confocal microscopy. Animal work- mice handling with breeding colony of transgenic, wild type and immune-deficient mice Local delivery of recombinant proteins and its stabilized formulations in animals. Experience with various routes of drug administration iv, ip, and sc.
