Six years ago, we published a volume on Imaging Flow Cytometry in the Methods in
Molecular Biology series [1] and would now like to extend the presentation of the capabilities
of modern cytometry development in this series from a broader perspective.
In recent years, flow cytometry has demonstrated significant progress in technology
(data acquisition) and data analysis. Improvement of flow cytometry is now directed to
overcoming limitations of the traditional flow cytometry technology in the number of colors
(labels) that could be detected simultaneously and the inability to capture images of each cell
(object) during analysis. Two branches are emerging: imaging flow cytometry (IFC) is
gaining popularity, and spectral flow cytometry (SFC) instruments became commercially
available just several years ago. This volume aims to present an overview of spectral cyto-
metry, recently developed protocols in the IFC area, and several protocols developed by
using FlowCam – a special imaging cytometer whose potential in basic research is still
underestimated.
Multi-laser flow cytometers made it possible to detect up to 15–18 colors [2–4];
however, the compensation tables, in this case, become critically large, and detection is
usually limited only to relatively bright populations. It means that despite the many colors
used, not all subpopulations, for example in bone marrow probes, could be distinguished
unambiguously. This limitation has recently been overcome by introducing spectral instru-
ments equipped with PMT arrays (altogether, it has up to 186 PMTs), with each detector
assigned to the narrow part of the spectrum. The development of special spectral unmixing
algorithms for data analysis improved the discrimination of the populations stained with
dyes having similar fluorescent spectra compared to the spectral compensation algorithms in
conventional cytometry. Spectral cytometry allowed to extend of the multicolor panel
beyond 40 colors [5]. The most prominent advantage of SFC is its ability to analyze
autofluorescent spectra in detail. This feature gives new capabilities to the analysis of
heterogeneous populations of blood cells [6–8] and will become an indispensable tool for
the analysis of algae and cyanobacteria.
Another problem of conventional flow cytometry is the inability to take pictures of
individual cells. Information about objects in the flow cell of a conventional cytometer is
limited by its light scatter properties. For example, doublets cannot always be resolved from
singlets making analysis of the rare events extremely difficult. Visualization of individual cells
in the brightfield mode and in fluorescent channels during population analysis was resolved
by introducing Imagestream-100 and later Imagestream X and Mark II models (Amnis)
with the time-delayed camera(s). Imagestream instrument (Amnis) was developed for the
analysis of relatively small cells similar to those in the conventional flow cytometry and used
the cuvette 250 μm in diameter and 875 μm in depth. It allows to obtain high-resolution
images of cells during flow experiments in multiple fluorescent channels with high sensitiv-
ity. The image-enabled intelligent high-speed cell sorting of single cells is under develop-
ment [9, 10].
A separate line of evolution of the flow instruments resulted in the introduction of
FlowCam, designed for the analysis of relatively large objects. FlowCam was initially
developed for the analysis of phytoplankton and some zooplankton and later became a
useful instrument for the evaluation of aggregated particles in pharma industries. Specific

features of the FlowCam are interchangeable objectives with different magnification and
flow cells of different diameters allowing analysis of planktonic organisms up to 600 μM.
However, FlowCam is mainly addressed to the analysis in the brightfield mode using a color
camera, and its fluorescent capabilities are limited by not more than two channels. The
image gallery is created by this instrument offline using special software.
The basic knowledge and techniques of IFC have been well documented in our previous
volume (2016). Two aspects are considered in the current volume – the development of
rapidly emerging spectral cytometry and some applications of imaging flow cytometry.
This new volume is organized into three parts. The first part provides an introduction to
state-of-the-art spectral cytometry. In the first chapter, the authors review a relatively short
history of spectral cytometry development and discuss its advantages compared to conven-
tional cytometry. The second chapter demonstrates the possibility of discriminating differ-
ent phytoplankton species based on cell autofluorescence and provides a detailed protocol of
virtual filtering. At first glance, the absence of chapters on a detailed description of the new
SFC techniques may seem like an oversight for a volume having “Spectral Flow Cytometry”
in the title. However, given the rapidly evolving nature of SFC and the recent introduction
of new instruments, we expect special volumes dedicated to this novel technology will likely
be forthcoming in the next 2–3 years. The second part describes several novel applications of
imaging flow cytometry using Imagestream instrumentation for semi-quantitative and
quantitative analysis in different experimental models. Chapters reflect ongoing IFC
advances in quantitative analysis of pathogens (Legionella pneumophila), analysis of multi-
nuclearity, quantitative biodosimetry, and autophagy protocols. Moreover, methods for
quantification of specific organelles, such as Golgi complex and inflammasomes have been
added to the current volume. The third part contains detailed protocols for handling and
using the FlowCam imaging flow cytometer from the supplier and research protocol for the
studies of phytoplankton communities.
We are extremely grateful to all authors who provided chapters for this volume during
the difficult time of the COVID-19 epidemics. Last but not least, we would like to thank
Professor John Walker, Editor of the Methods in Molecular Biology series, for his unlimited
guidance and help.
We believe that this volume will be a valuable source for a wide audience looking for new
approaches in cytometry. The development of methods that will become instrumental in
spectral cytometry is continuing, whereas the IFC is becoming a matured cytometry
method. This is truly a fascinating time to be involved in cytometry, as spectral and imaging
cytometry continues to evolve at an amazing speed.

Abstract
Spectral flow cytometry is a new technology that enables measurements of fluorescent spectra and light
scattering properties in diverse cellular populations with high precision. Modern instruments allow simul-
taneous determination of up to 40+ fluorescent dyes with heavily overlapping emission spectra, discrimina-
tion of autofluorescent signals in the stained specimens, and detailed analysis of diverse autofluorescence of
different cells—from mammalian to chlorophyll-containing cells like cyanobacteria. In this paper, we review
the history, compare modern conventional and spectral flow cytometers, and discuss several applications of
spectral flow cytometry.

1 Introduction

Wade and colleagues made one of the first attempts to extract full
emission spectra during flow cytometry analysis in 1979 [5]. They
used a grating spectrograph and projected the spectrum of the
fluorescent signal onto TV type Vidicon detector (objects: Anacyc-
tis nidulans—chlorophyll and phycobilin, 600–750 nm; 3T3 fibro-
blasts from Balb/c mice, propidium iodide (PI) and fluorescamine
staining) [5]. The recorded signal from individual cells has a very
low signal-to-noise (S/N) ratio, and reasonable spectra were
obtained only by averaging recordings from hundreds of cells
(fibroblasts).
The next configuration of the spectral flow cytometer was
based on a photomultiplier tube (PMT) as a detector and grating
monochromator. This cytometer had a 10 nm bandwidth spectral
resolution, and signal detection was performed when cells were
running through the cuvette at a rate of hundreds of events per
second [6]. The system eliminated problems of the noisy back-
ground using PMT with adjustable gain and offset as a detector
(analyzed objects—fixed rat thymocytes, stained with Hoechst
33258) [6]. However, the spectrum measured was only between
400 and 600 nm, not including far-red and infrared
(IR) wavelengths.
Buican [7] described in 1990 a “real-time FT spectrometer”
that was an interferometer-based spectral detector using PMT with
minimal time needed for the recording of the spectrum (only
3.2 μs). However, this instrument was never used as a commercially
available cytometer. Subsequently, several more spectral systems
were created in an attempt to obtain spectra from short measure-
ments using conventional cytometers in 1990–2000. Thus, Gauci
and co-authors described configuration with the prism and
512-element intensified photodiode array based on the FACS IV
laser flow cytometer. They analyzed spectra obtained from Dictyos-
telium discoideum spores stained with Cy3, fluorescein isothiocya-
nate, R-phycoerythrin (R-PE), and calibration beads [8]. This
system was relatively slow (operating at 62.5 Hz) and not sensitive

(633 nm) lasers [16]. This system achieved a speed of 3000 random
events per second; however, the sensitivity was lower than that of
conventional filter-based detectors.
A similar system based on a modified BD FACSCalibur cyt-
ometer equipped with argon-ion laser and 100 W mercury lamp
was built by Goddard and co-authors [17] using a grating spectro-
graph and Hamamatsu CCD array with 80% quantum yield. The
spectra analyzed by this instrument were in the range of
500–800 nm. This instrument allowed recording spectra with
great linearity, making spectral subtraction to remove background
signals from labeled specimens such as Rayleigh scattering, Raman
light scatter, and even cell autofluorescence feasible. Also, the
sensitivity of the instrument was significantly lower (10–30 times)
than that of the conventional cytometer [17].
Alternative spectral cytometry systems used a charge-coupled
device (CCD) camera as a detector to measure spectra from single
cells and beads [17–19]. In 2012 Nolan’s group [20] developed
spectral FCM instruments and data analysis algorithms suitable for
everyday use. Their two systems were based on FACSCanto
equipped with 405 and 488 nm lasers and using EM-CCD (elec-
tron-multiplying CCD) detector (11.3 nm resolution in the
500–800 nm range) and Coulter Elite cytometer using 785 nm
laser for IR emission (at 3.23 nm resolution in 790–930 nm range).
Their spectral flow cytometers used a holographic grating and
EM-CCD detector for high-speed spectra detection. Customized
software was developed for the spectral unmixing and production
of spectra-derived parameters for individual cells.
Instrument calibration and data analysis were very complicated
at these early stages of spectral FCM development (circa 2012)
[21]. Instrument design was not standardized, requiring thorough
spectral calibration for each instrument. Also, different instruments
used different data formats, making cross-platform spectral analysis
tricky. In the first spectral cytometers, spectral unmixing was per-
formed through the least square unmixing algorithm or indirectly
through principal component analysis [22]. Overall comparing the
spectral data obtained by different instruments was practically
impossible. So far, at that time, the advantages of spectral FCM
over conventional multichannel flow cytometry were impossible to
use in many applications. The next step was done when commer-
cially available spectral cytometers with standardized parameters
appeared.
The system patented by Purdue University was licensed by
Sony Inc., which is producing the first-generation commercial
spectral cytometry system (sometimes named hyperspectral
cytometer)—the Sony SP6800 Spectral Analyzer was announced
at the end of 2012 and came to the market in 2014. Also, in 2014
Cytek Biosciences (USA) developed and soon released its Aurora

the quality of these filters plays a crucial role in the spreading when
unmixing similar spectra [3]. The commercially available filters
might slightly deviate from the characteristics provided by the
supplier and, thus, sometimes, do not adequately exclude the fluo-
rescent emission of other fluorophores and/or autofluorescent
molecules that overlap with the desired signal. In commercial spec-
tral cytometers from SONY, instead of the optical filters, specialized
prism-based optics are used to measure and separate emissions from
different fluorophores [38].
Another advantage of spectral unmixing in spectral cytometry is
better extracting of autofluorescence signal that could be treated as
an additional fluorophore [36], while compensation cannot be
applied to autofluorescence until its spectrum is recorded.
11-color labeling showed that CD69 low naive T cell subset was
replaced in lymph node faster than CD69 high memory T-cell
subsets [36]. Schmutz and co-authors (2016) [48], using a
two-laser Sony SP6800 instrument (405 and 488 nm), demon-
strated by detailed fluorescence-minus-one control (FMO) that
while the staining index (SI) for individual dyes in spectral FCM
was the same as in conventional FCM, spectral FCM gives much
better discrimination of dyes with similar fluorescent properties.
Spectral FCM allowed discrimination of dyes with the same peak
fluorescence intensity when the overall spectra were different and
dyes with similar spectra but shifted for 10–20 nm peaks using
Kaluza software (YFP versus GFP; both proteins versus
FITC) [48].
Besides, spectral FCM allowed discrimination of lymphocytes
among the cells isolated from the tissues with high autofluores-
cence. Complete elimination of autofluorescent signal makes it
possible to discriminate dye-positive and dye-negative cells using
dyes with emission spectra close to the autofluorescent spectra for
further analysis [48].
The Sony SP6800 Spectral Cell Analyzer instrument utilized
a 32 multianode PMT (Hamamatsu), and spectrum separation is
achieved through a complex prism-based monochromator.
SONY Inc. demonstrated a prototype instrument and reported
on hyperspectral technology during the ISAC Congress in Seat-
tle in 2012 and announced the launch of the new hyperspectral
flow cytometer product—an SP6800 Spectral Cell Analyzer—
in 2012.
In some applications, the multiplexing by spectral tags may not
require spectral unmixing. In this setting, it may be beneficial to
classify the spectra directly instead of classification based on
unmixed intensities. Many techniques may be utilized here, includ-
ing unsupervised data reduction (using, for example, principal
component analysis, independent component analysis, or factor
analysis) or supervised techniques (such as neural networks or
support vector machines).
Advantages of spectral cytometry such as a large number of
studied parameters in one panel with better resolution due to the
removal of the autofluorescence signal and a rate of several thou-
sand events per second (Sony SP6800 10,000–20,000 events/
second, Cytek Aurora 35,000 events/s), have led to an increase in
the practical use of spectral cytometers in immunophenotyping.
One of the first multicolor panels (nine colors) was created by
Futamura and co-authors [38] at the presentation of the Spectral
Analyzer SP 6800 to study the movement of KikGr protein after
photoconversion in the inguinal lymph node cells. The remaining
immune cells, after photoconversion, changed their emission from
green to red (KikGrGreen-KikGrRed) while migrated cells stayed

Chapter 2
Abstract
Fluorescence methods are widely used for the study of marine and freshwater phytoplankton communities.
However, the identification of different microalgae populations by the analysis of autofluorescence signals
remains a challenge. Addressing the issue, we developed a novel approach using the flexibility of spectral
flow cytometry analysis (SFC) and generating a matrix of virtual filters (VF) which allowed thorough
examination of autofluorescence spectra. Using this matrix, different spectral emission regions of algae
species were analyzed, and five major algal taxa were discriminated. These results were further applied for
tracing particular microalgae taxa in the complex mixtures of laboratory and environmental algal popula-
tions. An integrated analysis of single algal events combined with unique spectral emission fingerprints and
light scattering parameters of microalgae can be used to differentiate major microalgal taxa. We propose a
protocol for the quantitative assessment of heterogenous phytoplankton communities at the single-cell
level and monitoring of phytoplankton bloom detection using a virtual filtering approach on a spectral flow
cytometer (SFC-VF).

the most variable and elongated regions. A matrix of several virtual filters corresponding to the variable

Based on the most variable spectral regions, combination of virtual filters corresponding to spectrum regions
in channels 15–20 (488 nm excitation) and channel 32 (488 nm excitation), channels 31–32 (488 nm
excitation) and channels V1-CH9 (405 nm excitation), and in channel 32 (488 nm excitation) and channels
4–15 (405 nm excitation) were selected to achieve the best discrimination of the two cell populations. Spectra
of gated populations were then plotted to confirm the identity of discriminated populations

2.2 List of Microalgae cell cultures from major microalgae taxa, including
Microalgae Cell Cyclotella sp. CCMP334, Chlorella sp. CCMP251, Dinobryon
Cultures divergens CCMP3055, Cryptomonas pyrenoidifera CCMP1177,
Aphanizomenon sp. CCMP2764, Acutodesmus obliquus SAG
276-1, and Porphyridium sordidum SAG 114.79 were obtained
from the National Center for Marine Algae and Microbiota
(NCMA; Bigelow Laboratory for Ocean Sciences, USA) and Göt-
tingen University’s collection of algal cultures (Germany).
2.3 Reagents 1. Microalgae cell culture media: (1) DY-V medium; (2) L1
medium; (3) L1 derivative, L1–11 psu medium.

3 Methods

1:10 volume ratio making up to 250 μL sample. The next steps
are accommodated for the use of the ID7000 spectral system
(Sony Biotechnlogy Inc., USA).
4. Turn on the spectral flow cytometer and run autocalibration
using calibration beads. Use Ultra Rainbow calibration beads
(Spherotech, USA or Sony Biotechnology Inc., USA) for auto-
matic calibration.

3. Set the sample flow rate to 1 under the “Flow Control” tab to
keep the intermediate flow velocity.
4. Set the stopping condition to 50,000.
5. Create FSC_A vs. SSC_A dot plot and ribbon plot for all lasers.
(see Note 3).
6. Place the round-bottom tube with the Chlorella sp., Acutodes-
mus obliquus, and two mixed cultures at different ratios in
24 Tube rack.
7. Place the rack in the multi-well plate holder and click “Load”.
8. Highlight sample positions as a “Target” and move all samples
to sample group 1.
9. Choose Set current position in the first sample tube by right-
clicking, and then click “Preview.”
10. Once the sample is being processed, observe if any parameters
from the “Detector & Threshold,” e.g., fluorescence PMT
voltage and/or FSC/SSC gain, need to be tuned.
11. After tuning, click “Auto Acquire” to record the samples (see
Notes 4–7, Fig. 5).
12. Record mixed samples with ID 7000 spectral flow cytometer
(Fig. 6).

allerdings nicht wahrzunehmen, wohl aber eine ganz frische
Wasserlinie, die zeigte, daß der Wasserspiegel um einen Meter
gesunken war, was seinen Grund im Fallen des Tarim während des
Winters hatte. Die bedeutenden Dünen, die den See auf allen Seiten
einrahmen, ja bisweilen kleine Inseln bilden, beweisen, daß sie sich
an Ort und Stelle befunden haben, als diese Überschwemmung das
Land unter Wasser setzte. Die Anordnung der Dünen würde sonst
eine ganz andere sein; sie hätten im Süden und im Südwesten des
Sees gefehlt, wo sie nun statt dessen am höchsten waren.
2. April. Tschernoff gab sich nicht eher zufrieden, als bis er eine
90 Zentimeter tiefe Furt mit tragfähigem Sandboden gefunden hatte.
Den Kamelen gefiel das Bad. Auf der anderen Seite drangen wir
wieder in hohe Dünen ein, und der See entschwand uns bald aus
den Augen. Der Sand wurde immer gewaltiger; 8–11 Meter hohe
Dünenkämme wurden mit dem Nivellierspiegel gemessen.
Die Muselmänner waren mißmutig und glaubten, der See, den
wir verlassen hatten, sei der Kara-koschun gewesen. Ich selbst
begann unwillkürlich darüber nachzugrübeln. War es so, dann waren
wir verloren! Die Kamele keuchten in dem weichen Sande und
sahen so melancholisch aus, als zerbrächen sie sich den Kopf
darüber, warum wir den See sogleich verlassen hatten. Die Hitze
war unerträglich, und die Sonne schien uns gerade ins Gesicht.
So gefährlich war es jedoch nicht. Das Terrain veränderte sein
Aussehen in erfreulicher Weise. Der Sand nahm ab, ein Gürtel von
toten Pappeln wurde durchquert, und dann kam einer von lebenden
Tamarisken. Vor uns erhob sich ein 5 Meter hoher Hügel. Ich sagte
Faisullah, er solle mich hinaufbegleiten, um den Kara-koschun in
Augenschein zu nehmen. Und richtig! Von dieser Anhöhe sah man
im Südwesten, Süden, Südosten und Osten große und kleine
Flächen reinen, blauen Wassers, die durch gelbe Kamischfelder
voneinander getrennt waren.
Auf einer Landzunge in dem nächsten großen See wurden die
Zelte unmittelbar am Strande aufgeschlagen. Mit Genuß betrachtete
ich von meinem Zelte aus durch das Fernrohr die Scharen von
Gänsen, Enten und Schwänen, die auf dem See schwammen und
tauchten. Ein Wüstenreisender kann sich keine schönere Aussicht
träumen. Leider waren die Schwimmvögel zu weit vom Ufer entfernt.
Aus ihrer Art zu tauchen konnte man schließen, daß der See nicht
sehr tief war. Sein Wasser war ganz süß.
Das Zelttuch flatterte im Seewinde. Welch ein Unterschied gegen
den Wüstenwind und seinen Staub! Das Wasser plätscherte so
herrlich am Ufer, und halb träumend lag ich da, über diese jetzt so
glücklich beendete Fahrt nachdenkend. Das Vorhandensein des
Seebeckens des alten Lop-nor war festgestellt, ein ehemals
bewohnter Ort war entdeckt, ein neuer See in der Wüste angetroffen
worden. Weder Menschen noch Tiere waren dabei verloren
gegangen.
Was das Profil quer durch die ganze Lopwüste betraf, so fand
ich, daß es nicht genügte. Man kann die Ergebnisse des
Kochthermometers und des Aneroids in einem Lande mit so
unbedeutenden Höhenunterschieden nicht verwerten. Mein schon
gefaßter Entschluß, diese Gegend noch einmal zu untersuchen,
wurde nur noch fester, denn nur ein Präzisionsnivellement des
Beckens konnte über die äußerst unbedeutenden
Höhenunterschiede zwischen dem Lop-nor und dem Kara-koschun
Auskunft geben. Ich hatte nach meiner früheren Reise gerade
deshalb betont, daß der Lop-nor ein wandernder See sein müsse,
weil dieses ganze Gebiet nahezu in ein und derselben Fläche liege.
Zweiundzwanzigstes Kapitel.
Fünfundzwanzig Tage im Kahn.
112. Die Pontonfähren auf dem Wege nach Abdall. (S. 275.)
Auf der Rückfahrt schaute ich mit großer Spannung zu, wie die
Ruderer einen großen schönen Schwan mit den Händen griffen. Wir
sahen ihn auf dem offenen Wasser am Schilfrande schwimmen; er
tauchte aber unter, als wir uns näherten. Die Leute senkten die
Ruder mit aller Kraft ins Wasser und ruderten nach der Stelle hin, wo
der Schwan sich wahrscheinlich wieder zeigen würde, was an den
Ringeln auf der Wasserfläche zu sehen war. Als der Schwan wieder
emporkam, waren wir ihm so nahe, daß er sich in seiner Verwirrung
in das Schilf hineinbegab; damit war er verloren, denn dort konnte er
nicht die Flügel zur Flucht ausbreiten. Der Kahn flog ihm wie ein Pfeil
nach; der alte Jaman Kullu sprang ins Wasser, das ihm bis an die
Hüften reichte, fiel über das arme Tier her und schleppte dann seine
Beute in das Boot. Der Schwan war so verängstigt, daß er mit
schlaffem Halse und hängendem Kopfe wie tot dalag; er wurde
sofort getötet.
Zahllose Wildenten und Gänse bevölkerten den See. Vielleicht
hatten sich einige von ihnen schon zu Prschewalskijs Zeit hier als
Junge aufgehalten. Die Generation der menschlichen Eingeborenen,
die damals in ihrer vollen Kraft stand, ist jetzt zu Greisen und
Greisinnen geworden, und ein neues Geschlecht ist seitdem
herangewachsen. Nicht einmal die Seen sind dieselben geblieben,
sie sind geschrumpft und zugewachsen und haben die Lage
gewechselt; die Sanddünen sind vorgerückt, alles hat sich in der
kurzen Zeit von 20 Jahren verändert, und das Bild, das man von
einem flüchtigen Besuche mitnimmt, ist, genau genommen, nur eine
Momentphotographie.
Wir verbrachten die Nacht an einem sehr notwendigen Feuer auf
einem kleinen Eilande im „See der vergessenen Fischhaut“ und
kehrten am nächsten Tage bei starkem Winde und hohem Seegange
nach Kum-tschappgan zurück. Faisullah, der nie in einem Boote
gesessen hatte, wurde bleich und sehnte sich, wieder an Land zu
kommen.
Am 10. April wurde an dem Punkte, wo der Fluß sich in Sümpfe
auflöst, die Wassermenge des Tarim gemessen; sie betrug 29,7
Kubikmeter in der Sekunde. Im Jahre 1896 hatte ich an derselben
Stelle und um dieselbe Zeit 50,5 Kubikmeter gefunden. Der
bedeutende Unterschied beruht teils auf dem neugebildeten Schirge-
tschappgan-Arme, teils auf einer Menge von natürlichen Kanälen,
die sich seit meinem vorigen Besuche oberhalb des Kum-
tschappgan vom Hauptflusse abgetrennt haben.
Die Sanddüne, die dem Kum-tschappgan seinen Namen
gegeben hat, ist 10,24 Meter hoch. Die größte Tiefe des Sees
beträgt 5,15 Meter. Man kann diesen Vertikalunterschied von 15,39
Meter als den größten im ganzen unteren Lop-nor-Becken
betrachten.
Am 11. April unternahmen wir eine Kahnfahrt nach den südlichen
Teilen des Kara-koschun, die den nördlichen sehr unähnlich sind. So
betrug die größte gemessene Tiefe nur 1,90 Meter, aber während
des ganzen späteren Teiles der Fahrt hatten wir nur 0,3 und noch
weniger. Der Grund besteht aus feinem gelbem Schlamm, der auf
schwarzem Moore oder blauem Tone ruht; sobald das Ruder ihn
streift, steigt es wie Tinte im Wasser auf. Als das Wasser so seicht
wurde, daß die Kähne nicht mehr darauf schwammen, mußten die
Leute aussteigen und sie an Stricken weiterziehen; als aber auch
dies bald unmöglich war, mußten wir umkehren. Man sinkt 10
Zentimeter tief in den Schlamm ein, findet dann aber einen festen,
aus einer dünnen Salzschicht bestehenden Untergrund. Der Sate-
köll ist eine ausgedehnte, seichte Wasserfläche mit wenig Schilf und
liegt ganz in der Nähe des Wüstenweges, der am Südufer von Abdall
nach Sa-tscheo führt. Weder Seevögel noch Fische gibt es in diesen
sterilen Seebecken, die zu baldigem Verschwinden verurteilt sind;
keine Algen bedecken ihren Boden. Während des Sommers
trocknen sie vollständig aus, und ihr Bett verwandelt sich dann in
eine harte, rissige Lehmschicht. Der ganze Kara-koschun wandert
langsam nach Norden zurück.
Den Tag darauf ruderten wir nach Abdall und lagerten auf dem
westlichen Ufer, wohin auch die Kamele geführt wurden. Der Tarim
führte hier nicht weniger als 93,3 Kubikmeter in der Sekunde, verlor
also auf der kurzen Strecke bis Kum-tschappgan 63,5 Kubikmeter,
die sich in vielen Tschappganen oder Kanälen von dem Hauptflusse
abtrennen. Das Tarimdelta wandert flußaufwärts, wie auch die Seen,
die von den vielen Armen gebildet werden. Der Fluß hatte hier jetzt
infolge der Eisschmelze seinen höchsten Wasserstand und war
ungefähr ebenso hoch angeschwollen wie beim Hochwasser im
Herbst vor dem Zufrieren. Im Sommer steht die Wasserfläche volle 2
Meter niedriger.
Eine kurze Tagereise nördlich von der Gegend von Abdall wird
die Wüste von einem neugebildeten Arme durchschnitten, der von
Schirge-tschappgan am unteren Tarim ausgeht. Dieser Flußarm war
bisher nur von Jägern aus Abdall besucht worden; ich wollte eine
Karte von ihm aufnehmen. Da das Gebiet aber nur mit Kähnen
untersucht werden konnte, mußten solche auf irgendeine Weise
über den Wüstenstreifen, welcher den neuen Arm vom Tarim
trennte, mitgenommen werden; es handelte sich nun darum, wie
dies zu bewerkstelligen sei. Einen schweren Kahn auf einem Kamele
zu balancieren, ist unmöglich, und zwei hintereinander gehende
Kamele mit zwei Kähnen in der Länge zu beladen, gelang
ebensowenig; durch den ungleichen Takt ihres Ganges war es ein
ständiges Hin- und Herreißen, und die Tiere scheuten auch vor
diesen ungewöhnlichen Lasten.
Die einzige Möglichkeit war, sie wie Schlitten auf der Erde
weiterzuziehen, wobei vor jeden Kahn ein Kamel gespannt wurde
(Abb. 91). Die Tiere mußten vorsichtig geführt werden, damit sie
nicht scheuten und durchgingen. So gelangten die Fahrzeuge mit ein
wenig abgescheuertem Boden, sonst aber in gutem Zustand an Ort
und Stelle.
Unser Zug nahm sich recht komisch aus. Die Kamele hatten den
größten Teil ihrer Wolle verloren und waren, ein paar hier und dort
noch sitzende Haarbüschel abgerechnet, nackt. Sie zogen die
langen, knirschenden Kähne geschickt durch den Sand, in welchem
dadurch eine abgerundete Furche entstand. Alle Mann gingen zu
Fuß; ein drittes Kamel trug das Gepäck, alle notwendigen
Instrumente und Proviant für sieben Tage.
Faisullah sollte die Kamele wieder zurückführen und dafür
sorgen, daß sie nach Mian gebracht würden, worauf er und Ördek
uns wieder bei Schirge-tschappgan zu treffen hatten.
Ich, Tschernoff, Tokta Ahun, Jaman Kullu und noch zwei Ruderer
lagerten bei J a n g i - j e r (neue Stelle), wo wir die erste seeartige
Anschwellung des Schirge-tschappgan-Armes erreichten, die
größtenteils mit Schilf zugewachsen war, im Norden aber von
ziemlich hohem Sand begrenzt wurde.
Die Kähne wurden sofort ins Wasser gebracht, und die Männer
machten eine kleine Rekognoszierung, um die Strömung zu suchen,
der wir aufwärts folgen mußten, um uns in diesen Irrgängen von
Kamisch und Wasser nicht ganz zu verlieren.
Der Tag war so weit vorgeschritten, daß wir unser Nachtlager da,
wo wir uns befanden, aufschlugen. Die Nacht unter freiem Himmel
war jedoch wenig angenehm. Ein heftiger Nordoststurm erhob sich
und brachte starken Regen. Als Tschernoff mich gegen 4 Uhr mit
einem Filzteppiche bedeckte, war ich schon ziemlich durchweicht,
hatte einen kleinen See an den Füßen und ein paar kleine Bäche auf
dem Kopfkissen, schlief aber wieder ein, ohne mich um den Regen
zu bekümmern.
Unsere erste Beschäftigung am Morgen war, unsere Kleider an
einem schönen Feuer zu trocknen, worauf das Geschwader
ausgerüstet wurde. Ich hatte die Instrumente in meinem Kahne,
Tschernoff den Proviant in dem seinen; dieser war aber so schwer
belastet, daß die Reling nur 4 Zentimeter über dem Wasser lag, und
an offneren Stellen des Flusses schlugen die Wellen dann und wann
in den Kahn. Jolldasch, der ohne Erlaubnis bei uns geblieben war,
durfte auch mitkommen, fiel uns aber nur lästig; wenn es ihm im
Kahne zu langweilig wurde, sprang er ins Wasser und schwamm in
das Schilf hinein, und wir hatten viele Mühe, ihn wieder zu
erwischen.
Die Fahrt war ziemlich mühsam. Der Buran fuhr fort, im Schilfe
zu heulen und zu pfeifen, und wir mußten an dem dichten
Schilfbestande entlang rudern, um Schutz zu finden. Auf offenem
Wasser drohten die Kähne sich selbst bei ziemlich unbedeutendem
Wellenschlage mit Wasser zu füllen und unterzugehen. In dem
Schilfdickicht war es halbdunkel. Hier und dort plätscherte ein Fisch;
Schwäne und Gänse eilten fort, und bei ein paar Gelegenheiten
wurden sie ihrer Eier beraubt.
Solange die Strömung deutlich war, ging alles gut; dann aber
wurden wir durch ein vollständiges Labyrinth von dichtem Kamisch,
aus dem Wasser herausguckenden Tamariskenkegeln,
Anschwemmungen, Landzungen und Landengen gehemmt. Über
drei Stunden lang suchten wir hier kreuz und quer nach einem
Durchgange, gingen denselben Weg, den wir gekommen waren,
wieder zurück, verloren uns in schlängelnden Buchten, wo wir
wieder umkehren mußten, forcierten den Schilfbestand, indem wir
die Kähne mit den Rudern durch seine knackenden Wände trieben,
und schleppten sogar die Fahrzeuge über schilfbewachsene
Landengen, die benachbarte Wasserflächen voneinander trennten.
Jede Düne, die dabei passiert wurde, mußte einer von uns
besteigen; doch der Blick reichte bei dem Nebel nicht weit, und die
ganze Umgebung war ein einziges dichtes Dschungel.
Als dieses Suchen umsonst war, steckten wir das Kamisch in
Brand. Es gab in dieser mit Dämmerung gesättigten Atmosphäre ein
großartiges Schauspiel, als sich die Flammen in die regenfeuchten
Schilfhecken hineinwarfen und diese knallten, knisterten und
dampften und rabenschwarze Wolken emporsteigen ließen, die vom
Sturme zerzaust wurden und wie ein Trauerflor über diese
irreführenden Sümpfe mit ihren überwachsenen Irrgängen führten.
Rußflocken erfüllen die Luft, und man wird ebenso schmutzig wie
naß, während man in dem seichten Wasser umherpatscht und die
Kähne in die vom Feuer gebahnte Gasse schleppt, die jetzt auch
einen Blick nach vorn gestattet und uns sehen läßt, wo wir den
nächsten Weg zum offenen Wasser haben (s. bunte Tafel).
Endlich waren wir wieder auf dem rechten Wege, wo das Wasser
tüchtig strömte. Die Strömung war so saugend und so schnell, daß
die Ruderer all ihre Kraft aufbieten mußten, um den Kahn gegen sie
vorwärtszubringen. Gegen Abend sahen wir uns nach einem
geeigneten Lagerplatze an dem hier überall feuchten Ufer um; da es
uns aber nicht gelang, einen solchen zu finden, blieben wir bei
einigen Tamarisken und legten, um wenigstens trocken zu liegen,
Kamisch auf die Erde. In schneidendem Wind machte ich meine
Aufzeichnungen beim Scheine des Lagerfeuers, und der Sturm
peitschte den Flußarm derart, daß von den Wogenkämmen weißer
Gischt sprühte.
Der Morgen war wenig verlockend zur Fortsetzung der Fahrt. Im
Norden war der Himmel schwarzgrau von Flugsand, und bei
Wellenschlag läßt sich die Strömung schwer unterscheiden. Erst um
11 Uhr konnten wir aufbrechen und zogen nun nach Nordwesten
über langgestreckte Seen, die uns wieder in ein deutliches Flußbett
führten. Manchmal verengte sich dieses bis nur auf 10 Meter Breite
und hatte dann eine Stromgeschwindigkeit von 0,9 Meter. Dann folgt
eine verwickelte Strecke, von den Jägern Tokkus-Tarim (neun
Flüsse) genannt, weil der Strom hier in mehrere Arme geteilt ist.
In brennendem Schilfe.
Der größte See auf dem ganzen Wege zog sich glücklicherweise
von Norden nach Süden hin. Indem wir seinem östlichen, von hohen
Dünen eingefaßten Ufer folgten, blieben wir vor dem Sturme
geschützt. Gerade über den See hinüberzufahren, wäre unmöglich
gewesen; er war ganz weiß von den schäumenden Wellen, und es
war mehr Glück als Geschicklichkeit, daß wir uns auf der anderen
Seite wieder in den Flußarm hineinfanden. Der Wasserweg lief
alsdann nach Südwesten, und der Wind neutralisierte den
hemmenden Einfluß der Strömung. Bei dem Lager Nr. 30 führte
dieser Arm 10,6 Kubikmeter Wasser, die dem unteren Tarim
entzogen worden waren.
Auch die folgende Tagereise war verwickelt, und der Wind
dauerte fort. Die Minimaltemperatur war auf −0,3 Grad
heruntergegangen. Wir ruderten längs des Ufers weiter, gerieten
aber oft in Sackgassen. Ein junger Hirt, auf den Tokta Ahun
gestoßen war, diente uns als Lotse, bis wir eine Sattma erreichten,
von welcher aus ein alter Fischer uns zu Boot begleitete. Ohne seine
Hilfe wäre es uns nicht möglich gewesen, die Mündung des
Flußarmes zu finden, denn sie war total vom Schilfe verdeckt. Ein
wenig weiter aufwärts lotste er uns durch einen kaum meterbreiten
Kanal, wo die Kähne an einer Stelle auf das Land und durch das
Schilfdickicht gezogen werden mußten, um an einem etwa 55
Zentimeter hohen Wasserfalle vorbeizukommen. Ein zweiter
Katarakt hatte eine Höhe von 60 Zentimeter.
Man erhält durch diese Wasserfälle den Eindruck, daß der
Schirge-tschappgan-Arm stärkeres Gefälle hat als der Hauptfluß,
sich also auf einem etwas höheren Niveau befinden muß. Hierdurch
erklärt sich auch die Tendenz des ganzen hydrographischen
Systems, nach Norden zu wandern und sich nach diesen flachen
Depressionen hinüberzuwerfen. Ein Niveauunterschied von einem
Meter spielt in einem Lande, das beinahe ganz horizontal ist, eine
sehr große Rolle.
Bei J e k k e n - ö i fanden wir ein Dörfchen (Abb. 92) von 4 Sattmen
und 20 Einwohnern, ausschließlich aus Greisen, Frauen und kleinen
Kindern bestehend, denn die kräftigere männliche Bevölkerung hatte
sich nach Tscharchlik begeben, um Ackerbau zu treiben. Die
Bevölkerung lebt hier von Fischfang, Wildenten, die sie
massenweise fangen, und Enteneiern. Sie besitzen auch 150 Schafe
und eine Anzahl Kühe. Vor vier Jahren waren sie von Tscheggelik-ui
hierher gezogen und sie erzählten, daß der neue Flußarm erst vor
sieben Jahren angefangen habe, sich von ihrem See aus einen Arm
nach Osten zu bahnen.
Von einem ganzen Geschwader von Kähnen begleitet, steuerten
wir am 18. nach Südwesten über eine Seenreihe, die Tiefen bis zu
4,6 Meter zeigten. Diese Seen sind dadurch eigentümlich, daß das
Wasser sich von ihnen nach zwei Seiten teilt; die Bifurkation findet
nach Osten und Westen statt. Die Hauptmasse geht ostwärts und
bildet den Arm, dem wir gefolgt waren, ein Teil aber fließt bei
Schirge-tschappgan in den Tarim, und der Spiegel des Sees liegt 60
Zentimeter über dem Niveau des Tarim.
Allmählich kommen wir in einen Kok-ala (kleinen Flußarm) hinein,
der sich nach dem Tarim hinunterschlängelt, wo sein kristallhelles
Wasser sofort in den trüben Fluten des Flusses verschwindet.
Eine Strecke weiter abwärts machen wir an den Hütten von
S c h i r g e - t s c h a p p g a n Halt und haben eine prachtvolle Aussicht
über den gewaltigen Fluß, der hier gerade und regelmäßig ist und
von ehrwürdigen dichtbelaubten Pappeln eingefaßt wird. Hier
werden 5,6 Kubikmeter Wasser aus den Seen von Jekken-öi wieder
an den Tarim abgegeben, der selbst an diesem Punkte 108,4
Kubikmeter in der Sekunde führte, die größte Wassermenge, die ich
bis dahin in dem Flusse gefunden hatte (Abb. 93).
In der Nacht auf den 20. April ging die Temperatur wieder auf −4
Grad herunter, was für diese Jahreszeit recht ungewöhnlich ist.
Nachdem wir uns mit Faisullah, Ördek und Maschka wiedervereinigt
hatten, war unser Plan, auf den östlichen Seen, die ich das vorige
Mal entdeckt hatte, nach Tikkenlik zurückzurudern. Es war recht
ärgerlich, zwei Tage lang denselben Weg wie damals gehen zu
müssen, nach Kum-tschekke, aber die hydrographischen
Verhältnisse hatten sich so verändert, daß ich auf mehrere neue
Erfahrungen hoffen konnte.
So ließen wir denn jetzt den Nias-köll zur Linken liegen und
ruderten über den Tschong-köll, der seinen Namen (großer See) mit
Recht führt und auf dem man sich mit den wenig seetüchtigen
Kähnen nicht zu weit hinauswagen darf.
Sodann gehen wir einen mächtigen Flußarm hinauf, der lauter
Sand durchschneidet und Lailik-darja heißt. Indem ich von Zeit zu
Zeit die Wassermenge in diesem östlichen Arme auf dem Wege
aufwärts maß, würde ich allmählich seinen Charakter erforschen und
ausfindig machen können, wieviel Wasser unterwegs in den Seen
verloren geht, sich durch Verdunstung verflüchtigt, in den Boden
einsickert usw.
Der Sadak-köll hatte sein Aussehen in den vier Jahren
vollständig verändert. Er war mit Schilf zugewachsen und voller
Sand und Anschwemmungen, in denen ein Flußarm mit starker
Strömung entstanden war. Die Hütten, bei denen ich damals vom
Sturme aufgehalten wurde, standen noch. Ihre Bewohner waren
größtenteils noch dieselben; sie erkannten mich wieder und
empfingen uns mit der größten Freundlichkeit. Sie nennen ihr mit 26
Menschen bewohntes Dorf M e r d e k t i k , nach einem neugebildeten
Arme des Merdek-köll. Schritt für Schritt sehen wir, wie das ganze
hydrographische System nach Norden und Osten wandert, um
dereinst wieder in das Seebecken des alten Lop-nor
zurückzukehren.
Von dem Dorfe begleitete uns ein Fischer in seinem Kahne und
zeigte uns den gegen die ziemlich reißende Strömung angehenden
Weg nach Norden. Tschernoff hatte uns eine 6 Meter lange Stange
besorgt, die er in Meter und Dezimeter eingeteilt hatte, so daß er die
Tiefen direkt ablesen konnte, wenn ich Sondieren für wünschenswert
hielt. Längs der Ufer stand reicher Tograkwald in seinem ersten,
zarten Lenzgrün und wirkte da, wo er, wie hier, die gelbe, öde
Sandwüste als Hintergrund hatte, besonders anziehend.
In K u l a k t s c h a machten wir eine kurze Frühstücksrast; dort
wohnten noch immer fünf Familien, lauter alte Bekannte von 1896.
Seit meinem vorigen Besuch hatten die Hirten der Gegend eine
ziemlich feste Brücke (Abb. 94) von Balken, Ästen und Kamisch über
den Fluß geschlagen, um ihr Vieh im Sommer von einem Ufer nach
dem anderen hinübertreiben zu können. Sie ist so niedrig, daß man
mit belasteten Kähnen nur gerade unter ihr durchfahren kann; aber
sie ist pittoresk, wie sie ihre Pfähle und Balken in dem Ilek spiegelt,
dessen Wasser schwarz wie Tinte, aber auch klar wie Kristall ist.
Dann ruderten wir mit einer Geschwindigkeit von 1,8 Meter in der
Sekunde zwischen üppigen Wäldern und undurchdringlichen
Schilfdickichten weiter und langten bei Sonnenuntergang unter
Mückentanz in K u m - t s c h e k k e an, wo wir gleichfalls von Freunden
aus dem Jahre 1896 empfangen wurden.
Mit ihnen machte ich am folgenden Tage eine Fahrt nach dem
Merdek-köll, um von diesem eine Karte aufzunehmen. Dieser Fluß
empfängt sieben Kubikmeter Wasser vom Ilek und hat Tiefen bis zu
7,4 Meter, also viel bedeutendere als der Kara-koschun.
Von Kum-tschekke gingen wir weiter den Fluß hinauf, der noch
immer außergewöhnlich tief ist und von prächtigen Wäldern
eingefaßt wird. Es ist eine sehr schöne Gegend, die mit ihrem
Kanale und ihrem blanken, dunkeln Wasserspiegel einem Parke
gleicht, und es ist eine Freude, die unaufhörlich wechselnden
Uferszenerien zu betrachten (Abb. 95, 96).
Von dem See und Dorfe To s g a k - t s c h a n t s c h d i an nahmen wir
neue Ruderer, und ich machte einen Ausflug nach dem auf der
vorigen Reise entdeckten Arka-köll.
Auf der folgenden Tagereise wurde mitten im Flusse eine der
größten Tiefen gelotet, die ich im ganzen Tarimsysteme je gefunden
habe, 12,55 Meter. Von dem Punkte, wo wir am Ufer lagerten,
unternahm ich wieder eine Bootexkursion nach dem nahegelegenen
Ta j e k - k ö l l . Ich hatte nur einen Mann bei mir, und der Kahn mußte
einen halben Kilometer über Land von dem Flusse nach dem See
getragen werden. Der Tajek-köll, an dessen östlichem Ufer wir 1896
mit Kamelen durch mühsames Terrain wanderten, ist ziemlich offen
und hat in der Mitte Tiefen von 5,7 Meter, 6,9 Meter und 9,5 Meter,
die also beinahe doppelt so groß sind wie die tiefsten Stellen des
Kara-koschun.
Als ich wieder ins Lager kam, war das Zelt so voller Mücken, daß
sie ausgeräuchert werden mußten, worauf die Leinwand auf allen
Seiten zugezogen und Jolldasch angebunden wurde, damit er nicht,
wie er zu tun pflegte, aus und ein liefe und diese verwünschten
Insekten, die uns jetzt abends zu plagen begannen, mitbrächte. Es
ist eine Art großer hellgrauer Mücken, die ihr Opfer mit unglaublicher
Energie und unglaublichem Eigensinn umschwärmen; sie hinderten
mich am Schreiben sowie an jeder anderen Beschäftigung. Man ist
ihrem Blutdurste, gegen den der eines Tigers nichts ist, völlig
preisgegeben. Sie zeigen eine Todesverachtung, die nicht Mut,
sondern Dummdreistigkeit ist, greifen von allen Seiten an und gehen
gern in den Tod, wenn sie nur erst eine tüchtige Mahlzeit Blut haben
einsaugen können.
Am 27. brachen wir bei Tagesgrauen aus diesem unwirtlichen
Mückenneste auf. Es dauerte nicht lange, so fegte ein warmer,
dunstiger Südwestwind über das Wasser in diesem unheimlich
verwickelten Labyrinthe von Seen, Sümpfen und Flüssen hin. Wir
folgten unserem alten Ilek aufwärts; er gleicht kaum einem Flusse,
sondern eher einer offenen Gasse in einem Sumpfsee. Unsere
Richtung ist anfangs nördlich, aber beim S u j i - s a r i k - k ö l l (See des
gelben Wassers) biegen wir nach Westen ab, um in ungeheuer
verwickelten Dickichten und Dschungeln von Kamisch, durch die ein
schlecht instandgehaltener Tschappgan führt, zu verschwinden (Abb.
97). Drinnen ist es dunkel und schwül; das Kamisch ist von den
Stürmen über den engen Wasserweg gelegt worden und ist mit
Staub und Flugsand bedeckt. Stellenweise bildet das Ganze eine
Brücke, auf der man bequem weite Strecken über das im
allgemeinen 2 Meter tiefe Wasser gehen kann. Es war jedoch nicht
immer ganz leicht, unter diesen mächtigen natürlichen Gewölben,
wo man so staubig wird wie auf einer Landstraße, vorzudringen.
Zwischen Milliarden von Schilfstengeln rinnt das Wasser nach dem
Ilek hinab. Diesen Abend lagerten wir im Dorfe S c h e i t l a r , das wir
auch im Winter besucht hatten (Abb. 98); ich erhielt also einen
Anknüpfungspunkt an die Karte der Expedition nach Tschertschen.
