Definition
White blood cells (WBC) are a heterogeneous group of nucleated cells that can
be found in circulation for at least a period of their life. Their normal
concentration in blood varies between 5000 and 11,000 per microliter. They
play a most important role in phagocytosis and immunity and therefore in
defense against infection.

WBC Test
The white blood cell (WBC) count indicates the number of white blood cells in
a sample of blood. This count provides a clue to the presence of illness. White
blood cells are made in the bone marrow and protect the body against
infection and aid in the immune response. If an infection develops, white blood
cells attack and destroy the bacteria or other micro-organisms causing the
infection.

Technique
Leukocytes can be evaluated through several techniques of varying complexity
both quantitative and qualitative properties can be assessed in the laboratory.

The simplest test is the WBC count and differential. White cells can be counted
manually in specially designed chambers (Neubauer) or with automated
counters. The latter are widely used, offering the advantage of higher accuracy
and speed over manual techniques
Principle
 Whole blood collected in EDTA is diluted according to the

type of cell count obtained.

 The diluted blood suspension is then placed in a chamber and

the cell counted

 The count is multiplied by dilution factor and reported as

number of cells per microlitter (µL) of whole blood

Material
1. Hemocytometer with Neubauer grid.
2. Cover glass
3. Diluents
4. Microscope.

Methodology
 we Put the cover slip or glass slip on the top of grid area in the

Chamber

 Then dilute the sample:

1: 20 for WBC count

 Load the sample into the laoding area


in the chamber
 Count the cells in the 4 large squares
for WBC by Microscope
 calculate the number of cells (N × 50 )
Our result
We counted the number (N) of cells in the 4 large squares located at the four
corners of the chamber using the microscope and found:

4\55

By combine the 4 number together and then using this equation (N x 50) we
found the total number of WBC is 9750 cell in mm3 in our sample
