STUDY OF EFFECT OF DRUGS ON FROG'S RECTUS
MUSCLE PREPARATION
At the end of the practical class the student shall be able to:
1. Record the dose-response curve of acetyl choline on isolated frog's
rectus muscle and demonstrate the effect of eserine and d-tubocurarine on
it.
2. Interpret the observations and explain the basis for the same.
3. Understand the rationale for the use of skeletal muscle relaxants in
surgery and the pharmacological basis for reversal with neostigmine.
Dose-response curve:
It’s the curve obtained by plotting :Log doses on x-axis and its
consequential responses plotting on y-axis.the curve become sigmoid and
a linear relationship between log of does and the response are seen in the
intermediate (30-70%)response zone.
Animal: Frog
Apparatus: Isolated organ bath, student’s physiograph / recording drum
with smoked paper, writing lever, aerator, syringes and needles
Preparation :
Isolated frog rectus muscle is mounted in an organ bath filled with
aerated frog's Ringer solution.
The muscle is allowed to relax for 45 mints under 5 gm tension and
washed intermittently.
