“What? Will you just calm down and start talking in plain English! What are you trying to say?”
“It was a flesh wound…plenty of blood, but not enough to kill a person. Without arterial damage it would take a
long time to bleed out from this wound. But there is more to it…this guy died shortly after he was stabbed, but it
wasn’t from the knife, that much is for sure!”
The detective cursed under his breath…it was going to be a long day. “Okay, so the guy didn’t die from the knife
wound, but he is dead, so from what?!!”
Two weeks have passed and you have been brought in to help investigate this case. You have heard the
interrogation tapes and been briefed by both the lead detective and the coroner. The coroner has ruled the cause
of death of Dr. John Bleu a homicide, but by poisoning, rather than because of the knife-wound inflicted by Mrs.
Bleu, presumably now by accident. Small quantities of an unknown substance, hereafter referred to as “Compound
X”, were discovered in the decaf-coffee Dr. Bleu was drinking when he died. Dr. Bleu had brought the coffee home
with him from work as he did everyday according to co-workers and his wife. His wife is still under suspicion, but
it has not escaped the notice of the investigators that Dr. Bleu leads a large research group studying potentially
lethal enzyme inhibitors for the drug company Hinesbiopharma.
The company is working with the U.S. government in a counter-terrorism effort to study the effects of chemical
weapons so that potential treatments can be created in advance. Three scientists who Dr. Bleu supervised are
running independent projects studying different inhibitors toward the same enzyme, referred to in company
documents as XYZase. XYZase is critical to human metabolism and so development of chemical inhibitors of this
enzyme by terrorist groups is viewed as a real and present threat by counter-terrorism experts. The three
scientists are Dr. Greene, Dr. Gray, and Dr. Wight. According to the company policies, only Dr. Bleu and the single
scientist assigned to each project have access to the highly valuable but extremely lethal compounds each one is
studying.
To complete this case, you will be given enzyme kinetic data, and you will need to use Microsoft EXCEL (or another
graphing program) to graph the data in order to determine the kinetic parameters and type of inhibition. A tutorial
to guide you through using MS EXCEL is also provided.
Results: The compound found in the coffee (Compound X) has yet to be identified, but using the
small amount extracted, you have discovered that it is an inhibitor of XYZase. Given this
revelation, it seems clear that this compound is the most likely the murder weapon. Shown below
is the data you have gathered. Note: [S] is given in nM; other values represent observed V0 values
given in units of µmol product formed per minute.

Given that Dr. Bleu appears to have been killed by an inhibitor to XYZase, there is now sufficient
cause to search the labs of Drs. Greene, Gray, and Wight so that you can recover and test samples
of the compounds they are each developing. This discovery has opened up a new investigation
option! You decide to…
Results: By gathering samples from the labs of Drs. Greene, Gray and Wight you hope to discover
which of these people may have had access to what is now clearly the murder weapon, Compound
X. Shown below is the data you have gathered. Note: [S] is given in nM; other values represent
observed V0 values given in units of µmol product formed per minute.

solution (for example, the amount of the murder weapon you are adding to the assay), you
cannot directly compare the magnitude of inhibition between the murder weapon and the

You notice, however, that the samples you collected from the labs are now labeled 1, 2 and 3. You
ask your lab assistants which samples correspond to which labs, but they mixed up the samples
(somebody didn’t label the tubes when running the assays!). This is a disaster! Now, in addition
to determining which sample (if any) matches the murder weapon, you are going to have to testify
in court as a biochemist to convince a jury that the guilty sample (1, 2, or 3) belongs to the guilty
person (Drs. Greene, Wight, or Gray) based on what you know about enzyme kinetics! Are you up
to the challenge? Who killed Dr. Bleu?
The good news is that you may now go back to Hinesbiopharma and interview the three scientists
about the molecular mechanism of the inhibitor each is working with.
Dr. Gray
Results: Dr. Gray is studying the effects of a transition-state analog (Tymoczko 3E pgs. 104-105 &
138). This molecule mimics the geometry of the theoretical transition state in the XYZase reaction
mechanism. Dr. Gray has solved crystal structures that have confirmed that the inhibitor binds
non-covalently in the active site as expected.
Dr. Greene
Results: Dr. Greene is studying a compound that binds non-covalently to an allosteric site
(Tymoczko 3E pgs. 118-122) on the protein approximately 20Å away from the active site. She has
solved crystal structures, which reveal that this allosteric site is blocked in the absence of
substrate. When the enzyme binds the substrate, a shift in a helix connecting the active site and
the allosteric site, clears the site and allows the inhibitor to bind. Binding studies have confirmed
predictions made by the crystal structures: in the absence of substrate, the inhibitor fails to bind
to the enzyme.

Dr. Wight
Results: Dr. Wight works on an inhibitor of XYZase that acts by binding to a lysine residue in the
enzyme active site, to form a Schiff-base, which is a covalent linkage. This lysine is critical to
function, normally acting as an acid in the enzyme mechanism. Stability studies have confirmed
that under physiological conditions, the inhibitor- Schiff-base linkage formed in the enzyme active
site is extremely stable, with a half-life of more than 24 hours.

concentration of 100 nM.

Also Note: When V0 is given in units of molarity per unit time, then kcat = Vmax/[Et]
where [Et] is the concentration of the enzyme in the assay; however, when Vmax is given
in moles per unit time, then kcat = Vmax/Et, where Et is the number of moles of Et in the
assay.

… Egyedűl ülök műtermemben, s folyton az óra mutatóját lesem.
Alig tudom az ecsetet tartani, – hogyne, hisz az Ő megkezdett képe
áll előttem, hisz Őt várom! Inkább czeruzát veszek a kezembe, írni
próbálok, – de hát hiába, nem vagyok író, s mikor szívemet akarnám
kiönteni, csak ezt az egy szót tudom papirra vetni: boldogság,
boldogság!… Oh, te áldott szó, hogy ujjong minden betűd, ezerszer
leírnálak, – világgá szeretném kiáltani azt a mindenható örömöt, a
mit te nekem jelentesz! Emberek, nézzétek bennem a csodát:
megtértem! hiszek, szeretek! De csitt… Tudjátok-e, mikor szeret a
megtérő világfi igazán? Magamon látom; mikor még annak a legjobb
barátnak sem árulja el szíve titkát, a kinek eddig mindent elmondott,
– mikor végre megtanul hallgatni. Sose voltam fecsegő, de
bevalljam-e? Hízelgett hiúságomnak, ha sikereimről suttogtak. Most
csak azt látom, hogy az első dolog, a mit a nagy szerelem megöl, a
hiúság. Alázatos lettem, nem fitogtatom, hanem féltve, titkolódzva
őrzöm kincsemet. A második, a mit a nagy szerelem megöl: a
barátság. Elbámulok magamon, – mint a bűnt kerülöm azt a benső
hívemet, kinek gyónással tartoznám. A pajtások csodálkozva néznek
rám az utczán: «mi lelte Iharossy Zoltánt? Úgy megváltozott…»
Őt várom. Előttem megkezdett arczképe, s két kezem önkéntelen
imára kulcsolódik, a hogy rajongva rápillantok. Kék szemei olyan
megadóan, olyan búsan néznek rám szőke haja arany keretéből…
És én még mindig nem tudok magamhoz térni ámulatomból, még
mindig nem tudom elhinni boldogságomat: hát csakugyan te vagy
az, te magamnak jobbik fele, kihez öröktől fogva tartozom, s kire
csak így elkésve találtam rá? Hát így nézesz ki, hát így hívnak?…
Csodálkozva ismétlem nevedet: Ilona… Ki hitte volna, hogy ez lesz
az a név, mely eldönti éltemet? Pár héttel ezelőtt még nem is
ismertük egymást, fogalmunk se volt egymás létéről, – lehetséges-
e? Oh, azok az elvesztett évek!… De azért még se voltunk mi
idegenek, – nem kellett nekünk megismerkednünk: egymásra
ismertünk mi azonnal! Voltak közös emlékeink, – vágyaink
ugyanazokra a dolgokra emlékeztek vissza a multban… Én
harmincz éves vagyok, te huszonhat, csalódtunk, szenvedtünk
mindketten eleget, – mint hajótöröttek hánykolódtunk, szinte
menekültünk egymáshoz, emlékszel-e? Multamra gondolok, sok
léha kalandomra, oh, ha visszalophatnám azt a sok eltékozolt
csókot! Megtisztulni, – hogy méltó legyek nagy szerelmemhez…
Kopogtatnak, s ő belép. Édes vihar az az ölelés…
– Bűnös asszony vagyok, – suttogja remegve, – mi lesz velünk?
Mámorosan csókolom le az aggodalmat ajkairól:
– Ne törődj semmivel! Nincsen se jövő, se jelen, se mult, –
örökkévalóság a szerelem! Egyedül vagyunk a mindenségben…
Február.
Csak ennyit: szeretek, szeretek… Egyre azt kérdem magamtól:
szeretett-e már valaki én előttem? Úgy érzem, én fedeztem fel a
boldogságot… Hogyan alakulhat át így valaki egy hónap alatt? Én, a
kétkedő ember, ki eddig agyonelemeztem minden érzésemet, most
ujjongva tombolok: dolgozik a démon olykor bennem, de analizise
nem fog többé a boldogságomon…
És az az irigy világ körülöttünk! Milyen kajánul leskelődik titkos
szerelmünkre! Rettegünk tőle, – mámorunkban még
megfeledkezünk, s eláruljuk magunkat. Gyülölök már mindenkit, – ha
az utczán megszólítanak, előre félek a kérdésektől és menekülök.
Türelmetlenül szakítok barátaimmal, ismerőseimmel. Csakhogy nem
olyan könnyű ám egyszerre elvágni azt az ezer szálat, mely
bennünket a társasághoz fűz! Főleg az asszonyi szálakat… Művész
embernek mindig van egynéhány olyan viszonya, a melyet nem
lehet csak úgy máról holnapra felbontani. Az elhagyott szeretők
boszúállók, kifürkésznek mindent, – vigyázni kell! Pirulok, ha arra
gondolok, hogy már Ilonát szeretve, még ölelnem kellett egyik-másik
régi kedvesemet… De nem tehettem másként, épp mert szakítani
akartam velük. Olyan undor fogott el…
– Hanem milyen furcsák a nők! Határozottan megérzik ők az
emberen, ha szerelmes, – s a férfi lekötöttségének a sejtése csak
annál jobban kihivja a kaczérságukat. És szívesen beállnak
szerelemtolvajnak, ha arról van szó, hogy meglopják egy kollégájuk
boldogságát… «De maga milyen érdekesen sápadt!» – hallom
jobbról-balról, – «kétszer akkorák a szemei és milyen fényesek!…»
Soha annyi kalandra alkalmam nem volt, mint a mennyi most
lehetne. És mentől visszatartóbb az ember, annál merészebbek ők.
Mosolyognom kell… De azért a multkor még egyszer utolsót lobbant
bennem a hiúság. Levelet kaptam Mariettától. Hogyan, az a ragyogó
bálvány, Marietta ír nekem, ki eddig, minden erőlködésem daczára,
mindig csak kinevetett? Tán az egyedüli nő, a kiről egészen biztos
voltam, hogy soha meg nem adja magát nekem… Alig hittem a
szememnek: írt zavarodottan, érzelgően és sorai közt ott
szégyenkezett a vallomás. Három hónap óta nem voltam nála,
keressem fel azonnal. Olyas aggodalom vett erőt rajtam, mint a
gimnazistán érettségi előtt. A szinházban kerestem fel a nagy
művésznőt, öltözőjében. A tükör előtt ült s a hogy meglátott, egy
sikolylyal ejtette ki az ecsetet a kezéből. Láttam, elpirult a festék
alatt, – sietve tette be az ajtót. Fecsegtünk, én ügyetlenül és
kitérőleg feleltem. Olyan elragadó volt pongyolájában… legjobban
szerettem volna felugrani és megszökni. Ő is zavarban volt. Végre
egy hosszabb szünet után hirtelen rám nézett és így szólt:
– Ej, milyen félénk lett maga! Úgy látszik, nekem kell beszélnem,
– hát tudja mit, Iharossy, ne vesztegessük az időt. Meggondoltam
magamat…
Idegesen nevetett:
– Jöjjön el hozzám holnap délután… Zoltán, itt vagyok!…
Felkelt, én is felszöktem székemről, de hátraléptem előle.
Megdöbbent.
– Zoltán, mi lelte magát?
– Semmi…
Fürkészőleg nézett rám s aztán egyszerre kitört:
– Micsoda, – maga visszautasít engem?!
Mosolyogni próbáltam:
– De kérem… ha parancsolja…
– Maga szerelmes! És tudom is, hogy kibe… igen, igen!
Kimondta a nevét…
– Nem akarod velem megcsalni, – takarodjál!… Oh átkozott élet,
nem fogom tudni a szerepemet! – –
… Ahogy kiértem az utczára, sajátságos büszkeséggel
egyenesedtem ki s pillantottam fel a havas éjszakába. Ilonára
gondoltam… kiálltam hát a nagy vizsgát, ellenálltam,
visszautasítottam egy gyönyörű asszonyt. Úgy éreztem: most lettem
igazán férfivá! Hű vagyok, – micsoda erő! Van-e nagyobb erő a
hűségnél?…
Márczius.
Szeretem őt és hű vagyok hozzá… És életemben most érzem át
először, hogy micsoda végtelen boldogság az, hűnek lenni ahhoz, a
kit igazán szeretünk! Gyermekes örömmel hajtom a végletekbe ezt
az állhatatosságot: már azért is szemrehányást tennék magamnak,
ha az utczán egy csinos nő után megfordulnék… Neki, vagy az ő
gondolatának szentelem minden perczemet. Minek is veszem a
kezembe ezt a naplót, miért olvasom át olyan szorongva első lapjait?
Aggódva kérdem magamtól: változott-e azóta valamit a mi
boldogságunk? Oh, úgy féltem a mi szép szerelmünket! Hát nem, –
nem változott semmi, boldogok vagyunk, csak én… alig merem
bevallani magamnak: én szenvedek. Tisztázni szeretném
gondolataimat! Félek, a dolgok rendes folyamata ez – nekem
szükségképen szenvednem kell, éppen mert hű vagyok, és mert a
mióta hű lettem, egészen más szemekkel látom a világot. Óriási egy
fordulat! Az eddigi szerelmes költeményből egy egész életre szóló
szerelmes valóság lett. Megtettem azt a nagy lépést, a mi a szeretőt
a férjtől elválasztja, – életem súlypontját az imádott nőbe helyeztem
át. Ábrándjaimban mily boldogan neveztem őt feleségemnek… de
hisz ez hazugság! Ő már másnak a felesége! Milyen galádul bukom
a fenségesből a nevetségesbe: én férfi létemre hű vagyok hozzá és
ő?… Vannak dolgok, a melyekre gondolni se merek. Titok van
közöttünk, – egyikünk se meri említeni. Sajnálom őt és gyűlölöm
egyszerre… Az első időben mámorom káprázatától mit se láttam, ha
láttam is, költői magaslatomról nem törődtem vele. Most, hogy a
hűségem oly lázadóan követelődzik bennem, most látok sokat és
minden nap többet. Nézem a férjét, azt a feketeszakállas, hallgatag
embert, nézem aranyhajú kis leánykáját… Beszélgetek azzal a
komoly úrral s valami ádáz figyelemmel lesem minden mozdulatát:
ezekkel a karokkal ölelte át, ezekkel az ajkakkal csókolta meg
először imádottamat esküvő után. Ő eltudna mondani nekem
mindent… Ha a felesége nevét kimondja, összerázkódom, ha nevén
szólítja ökölbe szorúl a kezem. Szeretném a fülébe harsogni: nekem
van ahhoz jogom!… Olykor feltámad bennem a becsületérzet: mit
tolakodom én ide ebbe a békés családi életbe? Nyomorult gazember
vagyok. Van még más nő is a világon, szabadulni fogok. Oh, hiszen
ha megcsalhatnám, de nem tudom, – már késő. Szeretem,
szeretem… És aljasul megadom magam megint. Micsoda szatira!
Azt mondják, a szerető neveti ki a férjet, – hazugság! Ha tudná, neki
kellene nevetnie rajtam, hisz én többet szenvedek, mint a mennyit ő
szenvedne. Neki van felesége, családja, gyermeke, övé volt az én
imádottam legszebb ifjúsága… végeredményében én maradok
hoppon! Hiszen ha közönséges lelkű ember volnék és
megalkudnám! De nem tudok, és csak szenvedek…
A férje? Csak egyre vigyázok: valahogy meg ne barátkozzam
vele…
Április.
… Ilona látja szenvedésemet és csak megadóan hajtja le a fejét.
– Hát én, – suttogja – azt hiszed, én nem szenvedek még
százszor annyit?…
Szemeink már értik egymást, de ajkaink még nem tudnak, nem
mernek nyiltan szólni. Csak annál észvesztőbb csókokba fojtjuk
keserüségünket. Ő összekulcsolja kezeit és könyörög:
– Oh, hisz csak boldoggá tehetnélek téged! Mondd, mit tegyek?
Elszorul a torkom, de aztán így felelek:
– Legalább a jövőd legyen az enyém, – az tiszta… Igérd meg,
hogy az enyém lesz. Úgy érzem, akkor megnyugszom…
S a hogy karjaim közt van, a műterem félhomályában, suttogva
boldog ábrándokba merülünk. Menekülünk ebből a gyűlöletes
világból, el az álmok, tündérmesék országába. Hiszünk a
lélekvándorlásban, hiszszük, hogy még egyszer e világra jövünk, –
megesküszünk, hogy akkor majd hamarabb egymásra találunk, és
boldogok leszünk… De aztán félve légvárakat építünk e mostani
életünk jövőjére is. Még boldogok lehetünk, ugy-e? Könnyes
csókokkal biztatjuk egymást. De mikor, – hogyan?… Nem tudunk,
nem merünk rá felelni. Ilona szorongva lesi arczomat:
– Tied a jövőm, mienk az, – ugy-e, megnyugszol?…
Május.
Azt hittem, megnyugszom, de hát nyilván beteg vagyok: a
féltékenység kórja tovább harapódzik bennem. A jövő? Micsoda
hóbort, micsoda agyrém! A jelen éget engem, – némelykor már azt
hiszem, hogy megőrülök. A rögeszme nem hagy el: egyre férje
karjaiban látom őt. Ordítanék kínomban. A minap férje két heti
távollét után hazatért, – éppen édesen beszélgettünk, mikor
belépett. Szokatlan szenvedélyességgel ölelte meg előttem a
védekező asszonyt. Milyen könyörögve nézett rám Ilona, mikor
gunyos mosolylyal bucsúztam! Azt hittem, az eszem vesztem el
