Lab Test Instructions

1. Go to Genebank. Search and download COX1 gene sequences of the species

listed below:
i. Rousettus amplexicaudatus
ii. Macroglossus minimus
iii. Macroglossus sobrinus
iv. Eonycteris spelaea
v. Megaderma spasma (the outgroup).

2. Prepare a DNA alignment using the sequences downloaded.

3. Save the DNA Alignment using your student ID as the file name.

4. Perform a phylogenetic analysis of the aligned DNA using the following parameters:
Neighbor Joining tree with Kimura 2 parameter model and 1000 bootstrap
replicates.

5. Save the results as PNG photo, paste in MsWords and describe the type of tree
generated (Monophyletic, paraphyletic or polyphyletic). Name your MsWord file
using your student ID as the file name.

6. Submit both files as results for lab test.
