Laboratory Activity:
Extraction of Deoxyribonucleic Acid (DNA)
Introduction
All organisms contain DNA--the information storage molecule. In fact, all organisms
share at least 500 of the same genes. Changes in DNA lead to biological evolution. In
this activity, you will extract DNA and see what it looks like. Scientists use variations of
this method to isolate DNA from all kinds of organisms.
Materials
Laboratory Gown or Gloves Isopropyl Alcohol
Coat Hair Net or Cap Blender
Printed Laboratory Mung Beans Barbeque Stick
Worksheet Disposable Plastic Cup Meat Tenderizer
Writing Materials: Spoon Strainer or Cheesecloth
Ballpen and Pencil Clear Dishwashing
Cellphone Liquid
Methodology/Procedures and Illustrations
Step 1:
-Soak Mung beans in H2O overnight and then remove hulls.
-Put the Mung beans without hulls into the blender and blend for 15 seconds.
Step 2:
-Pour the soup through a strainer or cheesecloth into the beaker.
-Add two tablespoons of dishwashing liquid (about 30ml) and swirl to mix.
-Let the mixture sit for five to 10 minutes.
-Carefully pour the mixture into the test tube.
Step 4:
Tilt your test tube and slowly pour alcohol into the test tube down the side so that
it forms a layer on top of the Mung bean mixture.
