INTRODUCTION
Antibiotics are the organic secretion produced by
micro – organisms, which in low concentrations
are antagonistic to the growth of other micro-
organisms (mostly pathogens). Antibiotics have
proved very useful in combating several bacterial
diseases in man and animal. Antibiotics are
commonly obtained from Actinomycetes and
some eubacteria. Some of the important
antibiotics are Streptomycin, Aureomycin,
Terramycin, Chloromycetin, Erythromycin,
Neomycin etc.

Soil is a natural medium that harbours several


types of micro-organism. These micro-organisms
can be grown on culture media. The effect of
different types of antibiotics can be studied on the
growth of micro-organism growing in culture
medium. This is an important subject, therefore,
the study of effect of antibiotics on micro-
organism has been taken for the present project.
AIM
To study the effect of antibiotics on micro-
organisms.

MATERIALS REQUIRED
Potato, agar, dextrose, distilled water, four
different types of antibiotics (such as penicillin,
streptomycin, aureomycin, terramycin), syringe,
oven sterilized petridish, flasks, beakers, pipettes,
garden soil, glass, marker pen etc.

EXPERIMENTAL PROCEDURE
A. Preparation of culture Medium
Potato Dextrose Agar (PDA) Medium: 200 g of
peeled potato chips taken. They are then
boiled with 500 ml of water in a beaker for 15
minutes. The squeezed potato pulp thus
obtained through a muslin cloth and kept it in
a flask. 20 g of agar is taken in a beaker and
warmed with 500 ml of water. Both the
solution of potato and agar mixed and 20 g
dextrose is added to it. Thus one litre of PDA
medium is prepared.
Autoclave the medium at 15 pounds pressure
for 15 minutes.

B. Effects of antibiotics on soil micro-


organisms
i) 2 g of soil is taken and dissolved in 10 ml of
water in a beaker. Let the soil particle settle
down.
ii) 5 oven sterilized petridishes is taken and 1 ml
of soil suspension is poured in each of the
plates. Now 1ml of the four antibiotics is
poured separately in four petridishes with the
help of syringe, and marked with marker pen.
The fifth petridish is left without antibiotic to
serve as control.

iii) PDA is poured in each of the petridishes and


the suspension is mixed by rotating the
petridishes. The petridishes are left
undisturbed at a warm place for a week.

OBSERVATION
Colonies of micro-organisms appeared on the
culture medium, the colonies in each petridishes
is counted and presented my observation in the
following table:

The effect of different antibiotics on the micro-


organisms were assessed by counting the number
and size of the colonies growing in the
petridishes.
PRECAUTION
1.Did not expose the culture of petridish to the
atmosphere.
2.Sterilise the petridishes properly in the oven
before use.
3.Use proper kind of stains for different type
micro-organisms.

CONCLUSION
Pencillin and Terramycin were the most effective
antibiotic against micro-organism in soil.
